Replicability of data collected for empirical estimation of relative pollen productivity
The effects of repeated survey and fieldwork timing on data derived from a recently proposed standard field methodology for empirical estimation of relative pollen productivity (RPP) have been tested. Seasonal variations in vegetation and associated pollen assemblages were studied in three contrasting cultural habitat types; semi-natural ancient woodlands, lowland heaths, and unimproved, traditionally managed hay meadows. Results show that in woodlands and heathlands the standard method generates vegetation data with a reasonable degree of similarity throughout the field season, though in some instances additional recording of woodland canopy cover should be undertaken, and differences were greater for woodland understorey taxa than for arboreal taxa. Large differences in vegetation cover were observed over the field season in the grassland community, and matching the phenological timing of surveys within and between studies is clearly important if RPP estimates from these sites are to be comparable. Pollen assemblages from closely co-located moss polsters collected on different visits are shown to be variable in all communities, to a greater degree than can be explained by the sampling error associated with pollen counting, and further study of moss polsters as pollen traps is recommended

Cell patterning on photolithographically defined parylene-C:SiO2 substrates
Cell patterning platforms support broad research goals, such as construction of predefined in vitro neuronal networks and the exploration of certain central aspects of cellular physiology. To easily combine cell patterning with Multi-Electrode Arrays (MEAs) and silicon-based ‘lab on a chip’ technologies, a microfabrication-compatible protocol is required. We describe a method that utilizes deposition of the polymer parylene-C on SiO(2 )wafers. Photolithography enables accurate and reliable patterning of parylene-C at micron-level resolution. Subsequent activation by immersion in fetal bovine serum (or another specific activation solution) results in a substrate in which cultured cells adhere to, or are repulsed by, parylene or SiO(2) regions respectively. This technique has allowed patterning of a broad range of cell types (including primary murine hippocampal cells, HEK 293 cell line, human neuron-like teratocarcinoma cell line, primary murine cerebellar granule cells, and primary human glioma-derived stem-like cells). Interestingly, however, the platform is not universal; reflecting the importance of cell-specific adhesion molecules. This cell patterning process is cost effective, reliable, and importantly can be incorporated into standard microfabrication (chip manufacturing) protocols, paving the way for integration of microelectronic technology
Influence of Isoflurane on Immediate-Early Gene Expression
Background: Anterograde amnesia is a hallmark effect of volatile anesthetics. Isoflurane is known to affect both the translation and transcription of plasticity-associated genes required for normal memory formation in many brain regions. What is not known is whether isoflurane anesthesia prevents the initiation of transcription or whether it halts transcription already in progress. We tested the hypothesis that general anesthesia with isoflurane prevents learning-induced initiation of transcription of several memory-associated immediate-early genes (IEGs) correlated with amnesia; we also assessed whether it stops transcription initiated prior to anesthetic administration. Methods: Using a Tone Fear Conditioning paradigm, rats were trained to associate a tone with foot-shock. Animals received either no anesthesia, anesthesia immediately after training, or anesthesia before, during, and after training. Animals were either sacrificed after training or tested 24 h later for long-term memory. Using Cellular Compartment Analysis of Temporal Activity by Fluorescence in situ Hybridization (catFISH), we examined the percentage of neurons expressing the IEGs Arc/Arg3.1 and Zif268/Egr1/Ngfi-A/Krox-24 in the dorsal hippocampus, primary somatosensory cortex, and primary auditory cortex. Results: On a cellular level, isoflurane administered at high doses (general anesthesia) prevented initiation of transcription, but did not stop transcription of Arc and Zif268 mRNA initiated prior to anesthesia. On a behavioral level, the same level of isoflurane anesthesia produced anterograde amnesia for fear conditioning when administered before and during training, but did not produce retrograde amnesia when administered immediately after training. Conclusion: General anesthesia with isoflurane prevents initiation of learning-related transcription but does not stop ongoing transcription of two plasticity-related IEGs, Arc and Zif268, a pattern of disruption that parallels the effects of isoflurane on memory formation. Combined with published research on the effects of volatile anesthetics on memory in behaving animals, our data suggests that different levels of anesthesia affect memory via different mechanisms: general anesthesia prevents elevation of mRNA levels of Arc and Zif268 which are necessary for normal memory formation, while anesthesia at lower doses affects the strength of memory by affecting levels of plasticity-related proteins
Approaches to quantitative reconstruction of woody vegetation in managed woodlands from pollen records
There has been increasing interest in developing quantitative methods for reconstructing the dynamics of cultural landscapes over the last 15 years. This paper adds to this literature by using various approaches to reconstruct the vegetation of two woodlands subject to rotational coppicing (the periodic cutting of broadleaved trees and shrubs for wood products). Pollen deposition at ground level was determined at both sites using ‘Tauber’ traps placed near to the centre of 14 compartments of differing age in the coppice rotation. For the main woody taxa, Relative Pollen Productivity (RPP) estimates were derived using linear regression for pollen influx data and Extended R-value analysis for percentage data. The vegetation around three ponds was reconstructed by applying four methods (inverting the two RPP estimate approaches, the modern analogue technique and correction for pollen productivity using the linear regression estimated RPP values) to pollen data obtained from the uppermost sample of sediment from the ponds. To determine whether these methods gave better estimates of the vegetation composition than the original pollen proportions, the results were compared with the surveyed vegetation around each pond using the Bray–Curtis Index. Linear regression of pollen influx produced RPP values which are comparable with previous European studies, whilst for some taxa the Extended R-value analysis produced estimates which are orders of magnitude different both from values derived from the linear regression and previous work. No single approach performed equally well at reconstructing the vegetation around the ponds, and at two of the three locations the uncorrected pollen proportions were most similar to the surveyed vegetation.We conclude that applying quantitative reconstruction methods to individual small sites is, currently, not likely to be useful in complex cultural landscapes. In the context of coppiced woodland, deficiencies in our understanding of pollen taphonomy and the impact of the practice on pollen production first need to be rectified, and we identify strategies to address this situation
Family-Owned Newspapers: Filling Niches in Local U.S. Communities

Journal of Media Business Studies. 2(11)Since small town newspapers are facing increased competition and technological changes that are threatening their survival, the purpose of this paper was to analyze the strategy-making activities of these organizations from an ecological perspective. Findings indicate that family-owned newspapers were finding stability in retaining their core print businesses while migrating content to the web. Too many variations from the norm appeared to weaken financial footholds and were often eventually abandoned. Such behaviors indicate a rationale for upholding a “tried-and-true” approach in newspapers. While profits may fluctuate, the uncertainty that occurred with variation seemed to play a role in increasing the threat of failure for these small, family-owned newspapers
Pollen signals of ground flora in managed woodlands
This paper explores the vegetation signals contained in the non-arboreal pollen and spore (NAPS) components of pollen assemblages from Tauber traps placed in woodlands subject to rotational cutting (coppicing) in lowland England. Sets of three Tauber traps were placed in compartments of different ages at multiple locations within each woodland for 1 year, and pollen assemblages recorded along with a vegetation survey using a modified pin-frame method in an area of 10 m radius around each trap array. Cluster analysis suggests that, as expected, the ground layer vegetation broadly reflects the different environmental conditions in the three woods, with the main subdivisions within woods apparently driven by changes in ground cover between the early and late stages of the coppice cycle. Non-arboreal pollen and spores (NAPS) assemblages group according to woodland of origin, with subdivisions which relate to pollen abundance but lack a simple relationship with years since cutting. Indices of Association between NAPS records and plant presence in the area around each array were calculated using presence–absence data for multiple distances of vegetation survey. All values tend towards an asymptote, which is interpreted as implying an effective source area for the single taxon presence–absence indicator values of ground flora taxa in coppiced woodlands on the order of a 10 m radius. Only four taxa, Poaceae, Ranunculus acris-type, Cyperaceae, and Scilla-type, have an Index of Association (A) greater than 0.5, implying that the presence of pollen can be interpreted in terms of the local presence of the relevant plant taxon with some confidence. Estimates of Pollen Productivity relative to Poaceae are presented for five taxa: Apiaceae, Asteraceae (Cardueae), Cyperaceae, Mercurialis perennis, and Scilla-type. Years since cutting does affect the ground vegetation and NAPS assemblage trapped in these woods, but that the effect is more clearly seen at an assemblage level in the vegetation than in the pollen assemblages. The interpretative significance of NAPS taxa does not seem to be in providing information about the local conditions around the sampling point, but in reflecting the ground flora of the wider woodland
Uniqueness properties of the Kerr metric
We obtain a geometrical condition on vacuum, stationary, asymptotically flat
spacetimes which is necessary and sufficient for the spacetime to be locally
isometric to Kerr. Namely, we prove a theorem stating that an asymptotically
flat, stationary, vacuum spacetime such that the so-called Killing form is an
eigenvector of the self-dual Weyl tensor must be locally isometric to Kerr.
Asymptotic flatness is a fundamental hypothesis of the theorem, as we
demonstrate by writing down the family of metrics obtained when this
requirement is dropped. This result indicates why the Kerr metric plays such an
important role in general relativity. It may also be of interest in order to
extend the uniqueness theorems of black holes to the non-connected and to the
