    Regeneration Ecology of Chrysopogon aucheri and Cymbopogon jwarancusa in Grasslands of Upland Balochistan , Pakistan

    Field experiments were conducted to investigate the seed attributes, movements and fates of dispersal units, and seedling establishment of Chrysopogon aucheri and Cymbopogon jwarancusa in a representative grassland ecosystem in upland Balochistan, Pakistan. Cymbopogon jwarancusa had more filled and viable caryopses than Chrysopogon aucheri. Seeds (spikelets) of both species had similar morphological features. Chrysopogon aucheri had one dispersal unit, a triplet spikelet. Cymbopogon jwarancusa had four types of dispersal units: a paired spikelet, a partial raceme, an entire raceme, and a partial inflorescence comprised of two racemes. Paired spikelets and partial racemes of Cymbopogon jwarancusa had greater mean dispersal distances (94 and 101 cm) from the edge of the basal crown of marked plants to the ground surface than triplet spikelets of Chrysopogon aucheri (79 cm). Spikelets of Cymbopogon jwarancusa and Chrysopogon aucheri moved mean distances of 26 and 32 cm, respectively, on the ground surface before becoming trapped in a microhabitat. The mean angle of dispersal for both species was toward the northeast, according to the prevailing wind direction. An ant (Tica verona) was the only detected seed (spikelet) predator for Chrysopogon aucheri. Both species had a weakly persistent soil seed bank, with higher amounts of seeds found under plant canopies compared to open interspaces. The recruitment of Chrysopogon aucheri and Cymbopogon jwarancusa seedlings from the natural seed bank was monitored in seven different microhabitats under natural and above-normal precipitation regimes . Above-normal precipitation increased seedling recruitment for both species in all microhabitats. Cymbopogon jwarancusa had higher seedling densities than Chrysopogon auchfiri. Seedling survival and tiller development for both species were greatest in the gravel microhabitat in the natural precipitation treatment. Monsoon rains in late July enhanced emergence of both species from recently dispersed seeds but emerged seedlings did not survive to the end of the growing season. The field studies indicate that Cymbopogon jwarancusa has a greater regeneration potential than Chrysopogon aucheri in this grassland ecosystem in upland Balochistan. It may be difficult to increase the composition of Chrysopogon aucheri, the more desirable species in these grasslands, when using management techniques that rely on natural regeneration

    Comparison of Early and Late Conversion of Sirolimus in Experimental Model of Chronic Cyclosporine Nephropathy

    Sirolimus (SRL) is a promising drug for replacing calcineurin inhibitors. We performed this study to determine the optimal time of conversion from cyclosporine (CsA) to SRL in an experimental model of chronic CsA nephropathy. Three separate studies were performed. In the first study, SRL was given to rats with or without CsA for 4 weeks. In the second study, rats were treated initially with CsA for 1 week, and then switched to SRL (early conversion). In the third study, CsA was given for 4 weeks and then replaced by SRL for 4 weeks treatment of CsA (late conversion). The influence of SRL on CsA-induced renal injury was evaluated by assessing renal function, histopathology (interstitial inflammation and fibrosis), and apoptotic cell death. Combined CsA and SRL treatment significantly impaired renal function, increased apoptosis, and interstitial fibrosis and inflammation compared with CsA or SRL treatment alone. Early conversion to SRL did not change renal function, histopathology, or apoptosis compared with early CsA withdrawal. By contrast, late conversion to SRL significantly aggravated these parameters compared with late CsA withdrawal. In conclusion, early conversion from CsA to SRL is effective in preventing CsA-induced renal injury in a setting of CsA-induced renal injury

    Changes in histopathology and tumor necrosis factor-α levels in the hearts of rats following asphyxial cardiac arrest

    Objective Post cardiac arrest (CA) syndrome is associated with a low survival rate in patients who initially have return of spontaneous circulation (ROSC) after CA. The aim of this study was to examine the histopathology and inflammatory response in the heart during the post CA syndrome. Methods We induced asphyxial CA in male Sprague-Dawley rats and determined the survival rate of these rats during the post resuscitation phase. Results Survival of the rats decreased after CA: 66.7% at 6 hours, 36.7% at 1 day, and 6.7% at 2 days after ROSC following CA. The rats were sacrificed at 6 hours, 12 hours, 1 day, and 2 days after ROSC, and their heart tissues were examined. Histopathological scores increased at 12 hours post CA and afterwards, histopathological changes were not significant. In addition, levels of tumor necrosis factor-α immunoreactivity gradually increased after CA. Conclusion The survival rate of rats 2 days post CA was very low, even though histopathological and inflammatory changes in the heart were not pronounced in the early stage following CA

    Metformin Represses Self-Renewal of the Human Breast Carcinoma Stem Cells via Inhibition of Estrogen Receptor-Mediated OCT4 Expression

    Metformin, a Type II diabetic treatment drug, which inhibits transcription of gluconeogenesis genes, has recently been shown to lower the risk of some diabetes-related tumors, including breast cancer. Recently, “cancer stem cells” have been demonstrated to sustain the growth of tumors and are resistant to therapy. To test the hypothesis that metformin might be reducing the risk to breast cancers, the human breast carcinoma cell line, MCF-7, grown in 3-dimensional mammospheres which represent human breast cancer stem cell population, were treated with various known and suspected breast cancer chemicals with and without non-cytotoxic concentrations of metformin. Using OCT4 expression as a marker for the cancer stem cells, the number and size were measured in these cells. Results demonstrated that TCDD (100 nM) and bisphenol A (10 µM) increased the number and size of the mammospheres, as did estrogen (10 nM E2). By monitoring a cancer stem cell marker, OCT4, the stimulation by these chemicals was correlated with the increased expression of OCT4. On the other hand, metformin at 1 and 10 mM concentration dramatically reduced the size and number of mammospheres. Results also demonstrated the metformin reduced the expression of OCT4 in E2 & TCDD mammospheres but not in the bisphenol A mammospheres, suggesting different mechanisms of action of the bisphenol A on human breast carcinoma cells. In addition, these results support the use of 3-dimensional human breast cancer stem cells as a means to screen for potential human breast tumor promoters and breast chemopreventive and chemotherapeutic agents

    The Relationship between Neuroticism, Hopelessness, and Depression in Older Korean Immigrants.

    PURPOSE:This study aimed to investigate the relationship between neuroticism, hopelessness, and depression among older Korean immigrants. To extend this line of research, this study aimed to examine the effects of neuroticism and hopelessness in predicting depression among older Korean immigrants. METHODS:Data for this study came from a survey of 220 first generation Korean immigrants aged 65 years or older in Los Angeles County in 2012. Data were collected by face-to-face interviews with trained social workers using a structured questionnaire translated into Korean. All interviews were conducted in Korean. The neuroticism sub-scale of the Eysenck Personality Questionnaire was used to assess neuroticism (EPQN). Hopelessness was measured by the Beck Hopelessness Scale (BHS). Depression was measured by the 20-item Center of Epidemiological Studies Depression (CES-D) scale. RESULTS:The study found that age (β = .26, p< .01), gender (β = -.13, p< .01), income (β = -.13, p< .01), neuroticism (β = .51, p< .01), and hopelessness (β = .15, p< .01) were significant predictors of depression. CONCLUSION:The study provides preventive strategies that would help in the development of depression-reduction services or programs for the population, especially for those living with neuroticism and hopelessness

    Does Methylphenidate Affect Cystometric Parameters in Spontaneously Hypertensive Rats?

    Purpose: Methylphenidate (MPH) is one of the most commonly prescribed psychostimulants for attention deficit hyperactivity disorder (ADHD). However, there is limited research on its effects on lower urinary tract function. This study investigated changes in cystometric parameters after intragastric administration of MPH in conscious spontaneously hypertensive rats (SHRs), an animal model of ADHD. Methods: Fourteen- to 16-week-old male SHRs (n=10), weighing between 280 and 315 g, were used. Three micturition cycles were recorded before administering MPH. One hour after each intragastric MPH injection, three cycles of cystometrogram were obtained in the awake condition. Various cystometric parameters were evaluated, including basal pressure (BP), maximal pressure (MP), threshold pressure (TP), bladder capacity (BC), micturition volume (MV), micturition interval (MI), and residual volume (RV). The data were analyzed using paired Student t-tests. Results: Five SHRs were each administered a dose of 3-mg/kg MPH, and the other five received a dose of 6-mg/kg MPH. BP and MP increased significantly in the rats that received the 3-mg/kg MPH injection, but not in those that received the 6-mg/kg injection. BC, MV, and MI significantly increased in the rats that received the 6-mg/kg MPH injection, but not in those that received the 3-mg/kg injection. There were no significant changes in TP after either injection. Conclusions: Significant increases in BC, MV, and MI after the 6-mg/kg MPH injection suggest that the peripheral and the central nervous systems may play important roles in bladder function in those receiving MPH for ADHD
