    Mistakes in identity: sexual orientation and credibility in the asylum process

    This thesis examines the prejudice that exists on the part of decision-makers responsible for determining refugee status and adjudicating asylum claims in jurisdictions that accept claims based on sexual orientation. An analysis of case law from both common law and civil law jurisdictions uncovers the negative impact of judicial stereotypes about sexuality on refugees and asylum-seekers. It follows the increasing importance placed on proving the genuineness of the claimants\u27 professed sexual identity that has coincided with an increased emphasis on credibility, a trend that has heightened the impact of decision-makers\u27 biases regarding sexuality. In addition to analyzing case law, the opinions of lawyers and other experts are included to add nuance and further illuminate decisions. The unique challenges that distinguish lesbian, gay, bisexual and transgender (lGBT) claimants from other refugees as well as the behavior of deciSion-makers are subsequently interpreted through the lens of sociologist Erving Goffman\u27s theories on stigma and self-presentation. The thesis then suggests that Western stereotypes about sexuality (and non normative sexuality in particular) that revolve around appearance, demeanor, past relationships, sexual activity, cultural values, and other experiences and elements of identity are particularly problematic for lGBT refugees, most of whom come from a non-Western context. The thesis further asserts that to be understood properly, the refugee narrative must be examined with regards to the intersection of multiple identities-gender, ethnic, religious, and others. The conclusion ultimately drawn is that lGBT refugees who are multiply marginalized as a result of these identities must be seen as having been excluded from participation in the political and religious discourses that regulate and restrict their lives who attempt to rectify this injustice through their transgressions of social norms. In light of thiS, recommendations are made to consider lGBT claims based on political opinion and religious grounds rather than relying on membership in a particular social group which does not recognize the political and religious dimensions of sexual identity. Recognizing the considerable difficulty of such a shift, further suggestions are made with regard to combating stereotypes within the current particular social group approach

    (UA) 1764 р. вийшла публікація Г. Міллера про наслідки перших археологічних розкопок в Україні. Виявлений тоді Литий скарб потрапив до Кунсткамери, а потім – до Ермітажу. Наступні автори вивчали, характеризували цю колекцію, виходячи з отриманих нових даних скіфологією. З ’являлася у світ інформація й про інші надходження до Ермітажу з території, яка сьогодні входить до Кіровоградщини. Ці публікації можуть стати певними свідченнями для обґрунтування постановки питання повернення історичних художніх цінностей з-за кордону до музейного фонду України.(EN) In 1764 the publication of G. Muller went out about the consequences of the first archaeological excavations in Ukraine. Poured treasure is educed then got to Cabinet of curiosities, and then to the Hermitage. Next authors studied, characterized this collection, coming from the obtained new data, as scythology. Information was published about other receivabless to the Hermitage from territory that is today included to the Kirovohrad area. These publications can become certain certificates for the ground of statement of a question of return of historical artistic values in the museum fund of Ukraine

    Interplay of Mre11 Nuclease with Dna2 plus Sgs1 in Rad51-Dependent Recombinational Repair

    The Mre11/Rad50/Xrs2 complex initiates IR repair by binding to the end of a double-strand break, resulting in 5′ to 3′ exonuclease degradation creating a single-stranded 3′ overhang competent for strand invasion into the unbroken chromosome. The nuclease(s) involved are not well understood. Mre11 encodes a nuclease, but it has 3′ to 5′, rather than 5′ to 3′ activity. Furthermore, mutations that inactivate only the nuclease activity of Mre11 but not its other repair functions, mre11-D56N and mre11-H125N, are resistant to IR. This suggests that another nuclease can catalyze 5′ to 3′ degradation. One candidate nuclease that has not been tested to date because it is encoded by an essential gene is the Dna2 helicase/nuclease. We recently reported the ability to suppress the lethality of a dna2Δ with a pif1Δ. The dna2Δ pif1Δ mutant is IR-resistant. We have determined that dna2Δ pif1Δ mre11-D56N and dna2Δ pif1Δ mre11-H125N strains are equally as sensitive to IR as mre11Δ strains, suggesting that in the absence of Dna2, Mre11 nuclease carries out repair. The dna2Δ pif1Δ mre11-D56N triple mutant is complemented by plasmids expressing Mre11, Dna2 or dna2K1080E, a mutant with defective helicase and functional nuclease, demonstrating that the nuclease of Dna2 compensates for the absence of Mre11 nuclease in IR repair, presumably in 5′ to 3′ degradation at DSB ends. We further show that sgs1Δ mre11-H125N, but not sgs1Δ, is very sensitive to IR, implicating the Sgs1 helicase in the Dna2-mediated pathway

    Expression of myriapod pair rule gene orthologs

    Background Segmentation is a hallmark of the arthropods; most knowledge about the molecular basis of arthropod segmentation comes from work on the fly Drosophila melanogaster. In this species a hierarchic cascade of segmentation genes subdivides the blastoderm stepwise into single segment wide regions. However, segmentation in the fly is a derived feature since all segments form virtually simultaneously. Conversely, in the vast majority of arthropods the posterior segments form one at a time from a posterior pre-segmental zone. The pair rule genes (PRGs) comprise an important level of the Drosophila segmentation gene cascade and are indeed the first genes that are expressed in typical transverse stripes in the early embryo. Information on expression and function of PRGs outside the insects, however, is scarce. Results Here we present the expression of the pair rule gene orthologs in the pill millipede Glomeris marginata (Myriapoda: Diplopoda). We find evidence that these genes are involved in segmentation and that components of the hierarchic interaction of the gene network as found in insects may be conserved. We further provide evidence that segments are formed in a single-segment periodicity rather than in pairs of two like in another myriapod, the centipede Strigamia maritima. Finally we show that decoupling of dorsal and ventral segmentation in Glomeris appears already at the level of the PRGs. Conclusions Although the pair rule gene network is partially conserved among insects and myriapods, some aspects of PRG interaction are, as suggested by expression pattern analysis, convergent, even within the Myriapoda. Conserved expression patterns of PRGs in insects and myriapods, however, may represent ancestral features involved in segmenting the arthropod ancestor

    Müller cell activation, proliferation and migration following laser injury.

    PurposeMüller cells are well known for their critical role in normal retinal structure and function, but their reaction to retinal injury and subsequent role in retinal remodeling is less well characterized. In this study we used a mouse model of retinal laser photocoagulation to examine injury-induced Müller glial reaction, and determine how this reaction was related to injury-induced retinal regeneration and cellular repopulation.MethodsExperiments were performed on 3-4-week-old C57BL/6 mice. Retinal laser photocoagulation was used to induce small, circumscribed injuries; these were principally confined to the outer nuclear layer, and surrounded by apparently healthy retinal tissue. Western blotting and immunohistochemical analyses were used to determine the level and location of protein expression. Live cell imaging of green fluorescent protein (GFP)-infected Müller cells (AAV-GFAP-GFP) were used to identify the rate and location of retinal Müller cell nuclear migration.ResultsUpon injury, Müller cells directly at the burn site become reactive, as evidenced by increased expression of the intermediate filament proteins glial fibrillary acidic protein (GFAP) and nestin. These reactive cells re-enter the cell cycle as shown by expression of the markers Cyclin D1 and D3, and their nuclei begin to migrate toward the injury site at a rate of approximately 12 microm/hr. However, unlike other reports, evidence for Müller cell transdifferentiation was not identified in this model.ConclusionsRetinal laser photocoagulation is capable of stimulating a significant glial reaction, marked by activation of cell cycle progression and retinal reorganization, but is not capable of stimulating cellular transdifferentiation or neurogenesis

    Reactive oxygen species induce virus-independent MAVS-oligomerization in systemic lupus erythematosus

    The increased expression of genes induced by type I interferon (IFN) is characteristic of viral infections and systemic lupus erythematosus (SLE). We showed that mitochondrial antiviral signaling (MAVS) protein, which normally forms a complex with retinoic acid gene I (RIG-I)–like helicases during viral infection, was activated by oxidative stress independently of RIG-I helicases. We found that chemically generated oxidative stress stimulated the formation of MAVS oligomers, which led to mitochondrial hyperpolarization and decreased adenosine triphosphate production and spare respiratory capacity, responses that were not observed in similarly treated cells lacking MAVS. Peripheral blood lymphocytes of SLE patients also showed spontaneous MAVS oligomerization that correlated with the increased secretion of type I IFN and mitochondrial oxidative stress. Furthermore, inhibition of mitochondrial reactive oxygen species (ROS) by the mitochondria-targeted antioxidant MitoQ prevented MAVS oligomerization and type I IFN production. ROS-dependent MAVS oligomerization and type I IFN production were reduced in cells expressing the MAVS-C79F variant, which occurs in 30% of sub-Saharan Africans and is linked with reduced type I IFN secretion and milder disease in SLE patients. Patients expressing the MAVS-C79F variant also had reduced amounts of oligomerized MAVS in their plasma compared to healthy controls. Together, our findings suggest that oxidative stress–induced MAVS oligomerization in SLE patients may contribute to the type I IFN signature that is characteristic of this syndrome
