    A Photometrically and Morphologically Variable Infrared Nebula in L483

    We present narrow and broad K-band observations of the Class 0/I source IRAS 18148-0440 that span 17 years. The infrared nebula associated with this protostar in the L483 dark cloud is both morphologically and photometrically variable on a time scale of only a few months. This nebula appears to be an infrared analogue to other well-known optically visible variable nebulae associated with young stars, such as Hubble's Variable Nebula. Along with Cepheus A, this is one of the first large variable nebulae to be found that is only visible in the infrared. The variability of this nebula is most likely due to changing illumination of the cloud rather than any motion of the structure in the nebula. Both morphological and photometric changes are observed on a time scale only a few times longer than the light crossing time of the nebula, suggesting very rapid intrinsic changes in the illumination of the nebula. Our narrow-band observations also found that H_2 knots are found nearly twice as far to the east of the source as to its west, and that H_2 emission extends farther east of the source than the previously known CO outflow.Comment: 19 pages, 6 figures, 1 tabl

    PIER: protein interface recognition for structural proteomics

    Abstract Recent advances in structural proteomics call for development of fast and reliable automatic methods for prediction of functional surfaces of proteins with known three-dimensional structure, including binding sites for known and unknown protein partners as well as oligomerization interfaces. Despite significant progress the problem is still far from being solved. Most existing methods rely, at least partially, on evolutionary information from multiple sequence alignments (MSA) projected on protein surface. The common drawback of such methods is their limited applicability to the proteins with a sparse set of sequential homologs, as well as inability to detect interfaces in evolutionary variable regions. In this study, we developed an improved method for predicting interfaces from a single protein structure, that is based on local statistical properties of the protein surface derived at the level of atomic groups. It was also demonstrated that the evolutionary conservation signal only marginally influenced the overall prediction performance on a diverse benchmark; moreover, for certain classes of proteins, using this signal actually resulted in a deteriorated prediction. The proposed Protein IntErface Recognition method (PIER) yielded improved performance as compared to several alignment-free or alignment-dependent predictions. PIER achieved the overall precision of 60% at the recall threshold of 50% at the residue level on a benchmark of 490 homodimeric, 62 heterodimeric and 196 transient interfaces. For 696 of 748 proteins (93%) the binding patch residues were successfully detected with precision exceeding 25% at 50% recall; for 524 proteins (70%) the corresponding precision was above 50%. The calculation only took seconds for an average 300-residue protein. The accuracy, efficiency, and dependence on structure alone make PIER a suitable tool for automated high-throughput annotation of protein structures emerging from structural proteomics projects

    Персональная телемедицинская система «Оберег» для дистанционного мониторинга пациентов

    The objective: to assess the potential use of the personalized telemedical system (PTS) of Obereg to ensure monitoring and constant medical control over the state of vital systems of the patient's body in the hospital, during transportation, and in out-of-hospital conditions.Subjects and methods. The Obereg system was tested in leading Russian clinics through simultaneous measurement of vital activity parameters with other standard patient monitoring systems. Comparative evaluation criteria were the following: functionality, measurement accuracy in comparison with stationary systems, reliability of operation, the impact on the operation of PTS of other equipment in the intensive care unit and possible interference, user friendliness for personnel and patients, and verification of communication capabilities based on field experiments. Additional parameters of the system were also evaluated. The need for it by medical units of the Ministry of Health of Russia was estimated.Results. The operability of the system has been confirmed in clinical conditions for patients of various profiles, including the most severe cases; it was found that the functionality of the system and accuracy of measurement met relevant requirements. It was estimated that the total demand for such systems in Russia might amount to 4,250,000 units.Conclusion. The device can be used for individual and group monitoring in intensive care units, in in-patient settings after transfer from intensive care, during rehabilitation for remote monitoring at home, and during patients' transportation to monitor their condition in case of emergency.Цель: оценить возможность использования персональной телемедицинской системы (ПТС) «Оберег» для обеспечения мониторинга и постоянного врачебного контроля состояния жизненно важных систем организма пациентов в стационаре, в процессе транспортировки и во внегоспитальных условиях.Материал и методы. Проведены испытания системы «Оберег» в ведущих российских клиниках путем одновременного измерения параметров жизнедеятельности другими штатными мониторами пациента. Критерии сравнительной оценки: функциональные возможности; точность измерения в сравнении со стационарными системами; надежность работы; влияние на работу ПТС другого оборудования отделения реанимации и интенсивной терапии и возможные помехи с его стороны; удобство работы персонала и использования пациентами; проверка возможностей связи на основании натурных экспериментов. Оценивались также дополнительные возможности системы. Проведен расчет потребности в ней медицинских организаций Минздрава России.Результаты. Подтверждена работоспособность устройства в клинических условиях для пациентов различного профиля, включая самых тяжелых; установлено, что функциональные возможности системы и точность измерения параметров соответствуют необходимым требованиям. Подсчитано, что общая потребность в подобных системах в России может составить до 4 250 000 шт.Заключение. Устройство может применяться для индивидуального и группового мониторинга в отделениях реанимации и интенсивной терапии, в обычных отделениях стационаров после перевода из реанимации, при реабилитационных мероприятиях для дистанционного мониторинга на дому, при перемещениях пациентов, для контроля их состояния в чрезвычайных условиях

    Large introns in relation to alternative splicing and gene evolution: a case study of Drosophila bruno-3

    Background: Alternative splicing (AS) of maturing mRNA can generate structurally and functionally distinct transcripts from the same gene. Recent bioinformatic analyses of available genome databases inferred a positive correlation between intron length and AS. To study the interplay between intron length and AS empirically and in more detail, we analyzed the diversity of alternatively spliced transcripts (ASTs) in the Drosophila RNA-binding Bruno-3 (Bru-3) gene. This gene was known to encode thirteen exons separated by introns of diverse sizes, ranging from 71 to 41,973 nucleotides in D. melanogaster. Although Bru-3's structure is expected to be conducive to AS, only two ASTs of this gene were previously described. Results: Cloning of RT-PCR products of the entire ORF from four species representing three diverged Drosophila lineages provided an evolutionary perspective, high sensitivity, and long-range contiguity of splice choices currently unattainable by high-throughput methods. Consequently, we identified three new exons, a new exon fragment and thirty-three previously unknown ASTs of Bru-3. All exon-skipping events in the gene were mapped to the exons surrounded by introns of at least 800 nucleotides, whereas exons split by introns of less than 250 nucleotides were always spliced contiguously in mRNA. Cases of exon loss and creation during Bru-3 evolution in Drosophila were also localized within large introns. Notably, we identified a true de novo exon gain: exon 8 was created along the lineage of the obscura group from intronic sequence between cryptic splice sites conserved among all Drosophila species surveyed. Exon 8 was included in mature mRNA by the species representing all the major branches of the obscura group. To our knowledge, the origin of exon 8 is the first documented case of exonization of intronic sequence outside vertebrates. Conclusion: We found that large introns can promote AS via exon-skipping and exon turnover during evolution likely due to frequent errors in their removal from maturing mRNA. Large introns could be a reservoir of genetic diversity, because they have a greater number of mutable sites than short introns. Taken together, gene structure can constrain and/or promote gene evolution

Явление, лежащее в основе принципа действия этих МДС, было позднее признано научным открытием (диплом № 79 от 17.03.1970) [1]. Необычность «явления синфазного и направленного разветвления энергии» тогда была объяснена наличием у МДС особого «плеча», которое отражает физическую возможность эффективного (практически полного) излучения из МДС в определенных режимах его работы (при практически полном отсутствии излучения в других режимах).     The interaction of the T-lymphocytes and epithelial cells of the thymus, line HTSC has been investigated. It has been shown, that the HTSC cells cause the aptoptosis of the predecessors of the T-lymphocytes, non-mature cells, activated T-cells, T-lymphoblasts patients with lymphoblast leucoses, which was being registered by the presence of the internucleosome degradation DNA by the electrophoresis method. The important role in the apoptosis inducted play the surface molecules - the pairs SD8/MNS-1 and LFA-1-ISAM-1, supernatants from the activated HTSC induce the aptoptosis of the lymphoid cells. The developed luminescent method of determination of the ATP is being used in the Institute of Immunology and on the chair of chemical enzymology of the Moscow State University.
