ein Projekt stellt sich vor
BNE (Bildung fĂŒr nachhaltige Entwicklung) ins Lehramtsstudium der Physik und
Informatik an der Freien UniversitÀt Berlin zu integrieren, ist das Ziel des
neuen Projektes BNE-Lehramtsausbildung im SchĂŒlerlabor. In diesem Projekt wird
das bereits etablierte und vorgestellte Konzept der Praxisseminare (Krofta et
al. 2013) weiterentwickelt. Im neu eingerichteten Praxisseminar Smart Grid
sollen Lehramtsstudierende BNE-Lehrerkompetenzen (nach Rauch et al. 2008)
erwerben, indem sie kompetenzorientierten Unterricht planen, im SchĂŒlerlabor
durchfĂŒhren und reflektieren. Als Grundlage der Unterrichtsentwicklung dient
das Konzept der Gestaltungskompetenz (de Haan, 2008). Das Nachhaltigkeitsthema
des Seminars, Smart Grid/ Stromnetz der Zukunft, ist durch zahlreiche
Wechselwirkungen und KomplexitÀt gekennzeichnet. Dies erfordert eine
interdisziplinÀre Herangehensweise und das Know-How einer mehrköpfigen
Arbeitsgruppe, die in unserem Praxissemester aus Studierenden der Informatik-
und der Physikdidaktik besteht. Das Projekt- und Seminarkonzept sowie die
Erfahrungen aus dem ersten Seminardurchlauf werden vorgestellt
Comprehensive Analysis of Cellular Galectin-3 Reveals No Consistent Oncogenic Function in Pancreatic Cancer Cells
Galectin-3 (Gal-3), a 31 kDa member of the family of beta-galactoside-binding proteins, has been implicated in the progression of different human cancers. However, the proposed roles differ widely, ranging from tumor-promoting cellular functions and negative impact on patient prognosis to tumor-suppressive properties and positive prognostic impact. We and others have previously identified Gal-3 as overexpressed in pancreatic cancer as compared to chronic pancreatitis and normal pancreatic tissue. The purpose of this study was thus the comprehensive analysis of putative cellular functions of Gal-3 by transient as well as stable silencing or overexpression of Gal-3 in a panel of 6 well-established pancreatic cancer cell lines. Our results confirm that galectin-3 is upregulated at the mRNA level in pancreatic cancer and strongly expressed in the majority of pancreatic cancer cell lines. In individual cell lines, transient knockdown of Gal-3 expression resulted in moderate inhibitory effects on proliferation, migration or anchorage-independent growth of the cells, but these effects were not consistent across the spectrum of analyzed cell lines. Moreover, functional effects of the modulation of Gal-3 expression were not observed in stable knockdown or overexpression approaches in vitro and did not alter the growth characteristics of nude mouse xenograft tumors in vivo. Our data thus do not support a direct functional role of Gal-3 in the malignant transformation of pancreatic epithelial cells, although paracrine or systemic effects of Gal-3 expression are not excluded
Funktionelle Charakterisierung der Gene NPC2 und ADRBK1 im Pankreaskarzinom
Das duktale Adenokarzinom des Pankreas stellt heute immer noch eines der aggressivsten und am schlechtesten behandelbaren Malignome dar. Deshalb ist es umso wichtiger, Genetik und Pathomechanismen dieser Erkrankung im Detail aufzuklĂ€ren und Marker zur FrĂŒh- bzw. Differenzialdiagnose sowie potentielle Ziele fĂŒr targeted therapies zu finden.
Mit Hilfe von enzymatisch generierten, Transkriptom-basierten shRNA-Bibliotheken
konnten in Hochdurchsatzanalysen potentielle Onko- bzw. Tumorsuppressorgene in PDAC identifiziert werden. NPC2 wurde als eines dieser potentiellen Onkogene fĂŒr weitere funktionelle Assays im Bezug auf ZellvitalitĂ€t, -proliferation und -motilitĂ€t ausgewĂ€hlt. Expressionsanalysen auf mRNA-Ebene zeigten eine deutliche Ăberexpression des Gens sowohl in pankreatischem Tumorgewebe als auch in verschiedenen Pankreaskarzinomzelllinien. Von vier in dieser Arbeit untersuchten Zelllinien zeigte sich nach siRNA-vermitteltem Knockdown jedoch nur in BxPC3-Zellen ein akuter funktioneller Effekt im Sinne einer hoch signifikanten Abnahme der ZellvitalitĂ€t. Dieser wachstumsfördernde Effekt von NPC2 lĂ€sst sich nicht auf das Pankreaskarzinom im Allgemeinen ĂŒbertragen. Die Zellproliferation und âmotilitĂ€t waren nach transienter Repression von NPC2 im Kurzzeit-Versuch nicht beeintrĂ€chtigt.
Auch das Gen ADRBK1 wurde in einem Hochdurchsatz-Screening, welches auf Pankreaskarzinom-spezifischen cDNA-Microarrays und dem Prinzip der reversen Transfektion basierte, identifiziert. ADRBK1 zeigte eine zeitabhĂ€ngige Translokation in den Nukleus und es stellte sich die Frage, ob die Kinase eine direkte Rolle bei der Genregulation spielt. Vorversuche der Arbeitsgruppe zeigten zudem, dass ADRBK1 im humanen PDAC nicht nur signifikant ĂŒberexprimiert wird, sondern auch pro- proliferative Effekte vermittelt. Rekombinante Ăberexpression der Kinase beschleunigte das Zellwachstum, wĂ€hrend ihr Knockdown dieses signifikant inhibierte. Als Ursache fĂŒr die beobachtete Wachstumshemmung kamen potentiell die Induktion eines Zellzyklusarrests oder Apoptose in Frage. Die daraufhin von mir im Western-Blot untersuchten Proteinlevel von CDKN1A/p21, CDKN1B/p27, Cyclin A, Cyclin D1, pRB und c-myc waren nach ADRBK1-Repression jedoch unverĂ€ndert. Eine Spaltung der Apoptosemarker PARP und Caspase 3 war nur in der Zelllinie PaTu-8988t nachweisbar. Somit kann weder die Induktion von Apoptose noch ein Zellzyklusarrest als maĂgebliche Ursache der Wachstumsabnahme nach ADRBK-Knockdown gelten. Um die Bedeutung des Gens fĂŒr PDAC weiter aufzuklĂ€ren, sollte die nukleĂ€re Translokation und deren potentielle Auswirkungen auf die Genfunktion weiter untersucht werden. Zudem sollten zellulĂ€re Zielproteine von ADRBK1, die Relevanz der Kinase beim Zusammenspiel von Krebszellen mit infiltrierenden Immunzellen und ihre Rolle bezĂŒglich der ZellmotilitĂ€t identifiziert werden
Double Clipping: Less-Biased Variance Reduction in Off-Policy Evaluation
"Clipping" (a.k.a. importance weight truncation) is a widely used
variance-reduction technique for counterfactual off-policy estimators. Like
other variance-reduction techniques, clipping reduces variance at the cost of
increased bias. However, unlike other techniques, the bias introduced by
clipping is always a downward bias (assuming non-negative rewards), yielding a
lower bound on the true expected reward. In this work we propose a simple
extension, called , which aims to compensate this
downward bias and thus reduce the overall bias, while maintaining the variance

Depletion of Macrophages Improves Therapeutic Response to Gemcitabine in Murine Pancreas Cancer.
BACKGROUND: The tumor microenvironment (TME) is composed of fibro-inflammatory cells and extracellular matrix (ECM) components. However, the exact contribution of the various TME compartments towards therapeutic response is unknown. Here, we aim to dissect the specific contribution of tumor-associated macrophages (TAMs) towards drug delivery and response in pancreatic ductal adenocarcinoma (PDAC). METHODS: The effect of gemcitabine was assessed in human and murine macrophages, human pancreatic stellate cells (hPSCs), and tumor cells (L3.6pl, BxPC3 and KPC) in vitro. The drug metabolism of gemcitabine was analyzed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Preclinical studies were conducted using KrasG12D;p48-Cre and KrasG12D;p53172H;Pdx-Cre mice to investigate gemcitabine delivery at different stages of tumor progression and upon pharmacological TAM depletion. RESULTS: Gemcitabine accumulation was significantly increased in murine PDAC tissue compared to pancreatic intraepithelial neoplasia (PanIN) lesions and healthy control pancreas tissue. In vitro, macrophages accumulated and rapidly metabolized gemcitabine resulting in a significant drug scavenging effect for gemcitabine. Finally, pharmacological TAM depletion enhanced therapeutic response to gemcitabine in tumor-bearing KPC mice. CONCLUSION: Macrophages rapidly metabolize gemcitabine in vitro, and pharmacological depletion improves the therapeutic response to gemcitabine in vivo. Our study supports the notion that TAMs might be a promising therapeutic target in PDAC
Significantly improved precision of cell migration analysis in time-lapse video microscopy through use of a fully automated tracking system
<p>Abstract</p> <p>Background</p> <p>Cell motility is a critical parameter in many physiological as well as pathophysiological processes. In time-lapse video microscopy, manual cell tracking remains the most common method of analyzing migratory behavior of cell populations. In addition to being labor-intensive, this method is susceptible to user-dependent errors regarding the selection of "representative" subsets of cells and manual determination of precise cell positions.</p> <p>Results</p> <p>We have quantitatively analyzed these error sources, demonstrating that manual cell tracking of pancreatic cancer cells lead to mis-calculation of migration rates of up to 410%. In order to provide for objective measurements of cell migration rates, we have employed multi-target tracking technologies commonly used in radar applications to develop fully automated cell identification and tracking system suitable for high throughput screening of video sequences of unstained living cells.</p> <p>Conclusion</p> <p>We demonstrate that our automatic multi target tracking system identifies cell objects, follows individual cells and computes migration rates with high precision, clearly outperforming manual procedures.</p
Lösungen und LeitfĂ€den fĂŒr das institutionelle Forschungsdatenmanagement
Hochschulen und deren Zentraleinrichtungen beschĂ€ftigen sich zunehmend mit dem Thema Forschungsdatenmanagement (FDM), um ihre Forschenden adĂ€quat zu unterstĂŒtzen. Nicht zuletzt aufgrund neuer Verlags- und Förderanforderungen wĂŒnschen sich Forschende Beratung und Orientierung, wie sie mit ihren Forschungsdaten umgehen sollen. Damit Hochschulen schnell und nachhaltig Lösungen zum institutionellen FDM etablieren können, haben fĂŒnf Berliner und Brandenburger UniversitĂ€ten im gemeinsamen Verbundvorhaben FDMentor mit Förderung des Bundesministeriums fĂŒr Bildung und Forschung (BMBF) entsprechende LeitfĂ€den und Werkzeuge erarbeitet. Die innerhalb von zwei Jahren (2017â2019) entstandenen Ergebnisse in den Bereichen Strategieentwicklung, Forschungsdaten-Policy, rechtliche Aspekte und Kompetenzausbau finden ĂŒber das Verbundprojekt hinaus ihre Anwendung.Peer Reviewe
Matrix-comparative genomic hybridization from multicenter formalin-fixed paraffin-embedded colorectal cancer tissue blocks
<p>Abstract</p> <p>Background</p> <p>The identification of genomic signatures of colorectal cancer for risk stratification requires the study of large series of cancer patients with an extensive clinical follow-up. Multicentric clinical studies represent an ideal source of well documented archived material for this type of analyses.</p> <p>Methods</p> <p>To verify if this material is technically suitable to perform matrix-CGH, we performed a pilot study using macrodissected 29 formalin-fixed, paraffin-embedded tissue samples collected within the framework of the EORTC-GI/PETACC-2 trial for colorectal cancer. The scientific aim was to identify prognostic genomic signatures differentiating locally restricted (UICC stages II-III) from systemically advanced (UICC stage IV) colorectal tumours.</p> <p>Results</p> <p>The majority of archived tissue samples collected in the different centers was suitable to perform matrix-CGH. 5/7 advanced tumours displayed 13q-gain and 18q-loss. In locally restricted tumours, only 6/12 tumours showed a gain on 13q and 7/12 tumours showed a loss on 18q. Interphase-FISH and high-resolution array-mapping of the gain on 13q confirmed the validity of the array-data and narrowed the chromosomal interval containing potential oncogenes.</p> <p>Conclusion</p> <p>Archival, paraffin-embedded tissue samples collected in multicentric clinical trials are suitable for matrix-CGH analyses and allow the identification of prognostic signatures and aberrations harbouring potential new oncogenes.</p
