    Characterization of cadmium proteinuria in man and rat.

    In workers chronically exposed to cadmium and without signs of renal insufficiency, plasma proteins with molecular weight ranging from 11,800 to 450,000 are excreted in greater amount in urine. Increased urinary excretion of low and high molecular weight proteins can occur independently. Because of its greater stability in urine and provided a sensitive immunological technique is used, the determination of retinol-binding protein is a more practical and reliable test of proximal tubular function than beta 2-microglobulin. The evaluation of renal function of workers removed from cadmium exposure indicates that cadmium-induced renal lesions, albeit of slow progression, are not reversible when exposures ceases. In workers chronically exposed to cadmium or removed from cadmium exposure, metallothionein in urine is directly correlated with cadmium in urine but not with cadmium in blood or years of cadmium exposure. The association between cadmium in urine and metallothionein in urine is independent of the status of renal function and the intensity of current exposure to cadmium. Whereas the repeated IP injection of high doses of cadmium to rat gives rise to a mixed or tubular type proteinuria, the prolonged oral administration of cadmium results mainly in the development of a glomerular type proteinuria. The former is usually reversible after cessation of treatment whereas the latter is not. Circulating antiglomerular basement membrane antibodies have been found in man and in rat chronically exposed to cadmium. The pathogenic significance of this finding deserves further investigation

    Uncovering functions of long-chain polyphosphate in higher eukaryotes: storage and synthesis of polyphosphate in matrix vesicles during mineralization

    Poster: abstract no. 0052Conference Theme: The Extracellular Matrix NicheInorganic polyphosphate (polyP) is a fundamental molecule that occurs throughout life. PolyP has been found in mammalian tissues with particularly high levels of polyP in bone and cartilage, however its metabolism and functions remain poorly understood. Here, we initially demonstrate the highest concentrations of polyP yet observed in a higher eukaryote in matrix vesicles (MVs) secreted from mineralized osteoblasts. We also observed that MVs isolated from both osteoblasts and chondrocytes have a remarkable ability to synthesize very long-chain polyP under physiological condition. PolyP could be hydrolysed to Pi by one of the critical enzymes of mineralization, tissue non-specific alkaline phosphatase (TNAP). PolyP was consumed during mineralization induced by MVs. Extravesicular PolyP acted as an efficient inhibitor to suppress MV-induced HA formation. Biochemical properties of polyP synthesis were consistent with membrane-bound enzyme activity within the MV.     The fecal microbiota of piglets during weaning transition and its association with piglet growth across various farm environments

    This study describes the fecal microbiota from piglets reared in different living environments during the weaning transition, and presents the characteristics of microbiota associated with good growth of piglets after weaning. Fecal samples were collected pre- (d26) and post-weaning (d35) from 288 male piglets in 16 conventional indoor commercial farms located in the West of France. The changes one week after weaning on the most abundant microbial families was roughly the same in all farms: alpha diversity increased, the relative abundance of Bacteroidaceae (-61%), Christensenellaceae (-35%), Enterobacteriaceae (-42%), and Clostridiaceae (-32%) decreased, while the relative abundance of Prevotellaceae (+143%) and Lachnospiraceae (+21%) increased. Among all the collected samples, four enterotypes that were ubiquitous in all farms were identified. They could be discriminated by their respective relative abundances of Prevotella, Faecalibacterium, Roseburia, and Lachnospira, and likely corresponded to a gradual maturational shift from pre- to post-weaning microbiota. The rearing environment influenced the frequency of enterotypes, as well as the relative abundance of 6 families at d26 (including Christensenellaceae and Lactobacillaceae), and of 21 families at d35. In all farms, piglets showing the highest relative growth rate during the first three weeks after weaning, which were characterized as more robust, had a higher relative abundance of Bacteroidetes, a lower relative abundance of Proteobacteria, and showed a greater increase in Prevotella, Coprococcus, and Lachnospira in the post-weaning period. This study revealed the presence of ubiquitous enterotypes among the farms of this study, reflecting maturational stages of microbiota from a young suckling to an older cereal-eating profile.     Renal and Neurologic Effects of Cadmium, Lead, Mercury, and Arsenic in Children: Evidence of Early Effects and Multiple Interactions at Environmental Exposure Levels

    Lead, cadmium, mercury, and arsenic are common environmental pollutants in industrialized countries, but their combined impact on children’s health is little known. We studied their effects on two main targets, the renal and dopaminergic systems, in > 800 children during a cross-sectional European survey. Control and exposed children were recruited from those living around historical nonferrous smelters in France, the Czech Republic, and Poland. Children provided blood and urine samples for the determination of the metals and sensitive renal or neurologic biomarkers. Serum concentrations of creatinine, cystatin C, and ÎČ(2)-microglobulin were negatively correlated with blood lead levels (PbB), suggesting an early renal hyperfiltration that averaged 7% in the upper quartile of PbB levels (> 55 ÎŒg/L; mean, 78.4 ÎŒg/L). The urinary excretion of retinol-binding protein, Clara cell protein, and N-acetyl-ÎČ-d-glucosaminidase was associated mainly with cadmium levels in blood or urine and with urinary mercury. All four metals influenced the dopaminergic markers serum prolactin and urinary homovanillic acid, with complex interactions brought to light. Heavy metals polluting the environment can cause subtle effects on children’s renal and dopaminergic systems without clear evidence of a threshold, which reinforces the need to control and regulate potential sources of contamination by heavy metals

    A simple formalism describing the light response of CsI(Tl) to heavy ions, which quantifies the luminescence and the quenching in terms of the competition between radiative transitions following the carrier trapping at the Tl activator sites and the electron-hole recombination, is proposed. The effect of the delta rays on the scintillation efficiency is for the first time quantitatively included in a fully consistent way.     Multifragmentation process for different mass asymmetry in the entrance channel around the Fermi energy

    The influence of the entrance channel asymmetry upon the fragmentation process is addressed by studying heavy-ion induced reactions around the Fermi energy. The data have been recorded with the INDRA 4pi array. An event selection method called the Principal Component Analysis is presented and discussed. It is applied for the selection of central events and furthermore to multifragmentation of single source events. The selected subsets of data are compared to the Statistical Multifragmentation Model (SMM) to check the equilibrium hypothesis and get the source characteristics. Experimental comparisons show the evidence of a decoupling between thermal and compresional (radial flow) degrees of freedom in such nuclear systems.Comment: 28 pages, 15 figures, article sumitted to Nuclear Physics

    Tumour-derived alkaline phosphatase regulates tumour growth, epithelial plasticity and disease-free survival in metastatic prostate cancer

    BACKGROUND: Recent evidence suggests that bone-related parameters are the main prognostic factors for overall survival in advanced prostate cancer (PCa), with elevated circulating levels of alkaline phosphatase (ALP) thought to reflect the dysregulated bone formation accompanying distant metastases. We have identified that PCa cells express ALPL, the gene that encodes for tissue nonspecific ALP, and hypothesised that tumour-derived ALPL may contribute to disease progression. METHODS: Functional effects of ALPL inhibition were investigated in metastatic PCa cell lines. ALPL gene expression was analysed from published PCa data sets, and correlated with disease-free survival and metastasis. RESULTS: ALPL expression was increased in PCa cells from metastatic sites. A reduction in tumour-derived ALPL expression or ALP activity increased cell death, mesenchymal-to-epithelial transition and reduced migration. Alkaline phosphatase activity was decreased by the EMT repressor Snail. In men with PCa, tumour-derived ALPL correlated with EMT markers, and high ALPL expression was associated with a significant reduction in disease-free survival. CONCLUSIONS: Our studies reveal the function of tumour-derived ALPL in regulating cell death and epithelial plasticity, and demonstrate a strong association between ALPL expression in PCa cells and metastasis or disease-free survival, thus identifying tumour-derived ALPL as a major contributor to the pathogenesis of PCa progression.British Journal of Cancer advance online publication, 22 December 2016; doi:10.1038/bjc.2016.402 www.bjcancer.com

    Multifragmentation in Xe(50A MeV)+Sn Confrontation of theory and data

    We compare in detail central collisions Xe(50A MeV) + Sn, recently measured by the INDRA collaboration, with the Quantum Molecular Dynamics (QMD) model in order to identify the reaction mechanism which leads to multifragmentation. We find that QMD describes the data quite well, in the projectile/target region as well as in the midrapidity zone where also statistical models can be and have been employed. The agreement between QMD and data allows to use this dynamical model to investigate the reaction in detail. We arrive at the following observations: a) the in medium nucleon nucleon cross section is not significantly different from the free cross section, b) even the most central collisions have a binary character, c) most of the fragments are produced in the central collisions and d) the simulations as well as the data show a strong attractive in-plane flow resembling deep inelastic collisions e) at midrapidity the results from QMD and those from statistical model calculations agree for almost all observables with the exception of d2σdZdE{d^2 \sigma \over dZdE}. This renders it difficult to extract the reaction mechanism from midrapidity fragments only. According to the simulations the reaction shows a very early formation of fragments, even in central collisions, which pass through the reaction zone without being destroyed. The final transverse momentum of the fragments is very close to the initial one and due to the Fermi motion. A heating up of the systems is not observed and hence a thermal origin of the spectra cannot be confirmed.Comment: figures 1 and 2 changed (no more ps -errors
