    Effects of temperature on wheat-pathogen interactions

    Climate change is affecting UK agriculture, and research is needed to prepare crops for the future. Wheat is the UK’s most important crop, and needs to be protected from losses caused by disease. While direct effect of the environment on pathogen spread is often reported, effect of the environment on host defence is not. Many wheat resistance genes are temperature sensitive and these were used as a starting point to investigate defence temperature sensitivity in wheat starting with yellow rust resistance gene Yr36, previously shown to be temperature-sensitive. The effect of temperature on resistance was shown to be independent of Yr36 in breeding line UC1041, and was more likely to be due to a previously-uncharacterised background temperature sensitivity. These results suggest that temperature changes, rather than thresholds, might influence some disease resistance mechanisms. Understanding this phenomenon could enable the breeding of more stable defence in crops. In order to gain further insight into how temperature changes influence resistance, plants were grown under different thermoperiods and challenged with different types of pathogens; Results showed that resistance to multiple pathogens in one cultivar Claire was enhanced under variable temperatures, compared to constant temperatures. Taken together, the research presented revealed that defence temperature sensitivity in plants is much more complex than previously thought, considering that both temperature changes and different thermoperiods can influence aspects of wheat defence. To ascertain which research approaches will be most valuable in preparing for climate change, the effect of the environment on take-all was also investigated. Vulnerable periods for wheat from the threat of take-all development were identified by analysing historical datasets, and controlled environment experiments. Results showed a relationship between initial post-sowing temperatures and spring take-all levels in 2nd 3rd or 4th winter wheats, depending on the location. The work on yellow rust resistance and take-all both identify vulnerable periods for wheat caused by the environment, be it weakening of host defence responses, or increased threat from disease pressure. Further characterisation and understanding of vulnerable periods will be essential to control disease outbreaks under an increasingly unstable climate

    Students speak : obtaining youth input regarding tobacco policies

    This study evaluated youth attitudes regarding the effectiveness of a variety of smoking-prevention strategies. The attitudes of young people have largely been overlooked in the field of tobacco-control. Here, students’ attitudes towards the following approaches were examined: financial disincentives to smoke; smoking prevention education based on the information deficit, affective education, and social influences models; and a variety of punishments and rewards derived from theories of compliance. Five hundred and seventy-seven middle and high school students in Knox County, Tennessee, were surveyed regarding their attitudes. Students’ attitudes towards the perceived effectiveness of these strategies varied based on students’ demographic characteristics. The three most significant characteristics were students’ current smoking status, their race, and their income level. Current smokers indicated that the majority of approaches that are incorporated in contemporary anti-smoking efforts are very unlikely to discourage them from smoking. In particular, they indicated little support for the effectiveness of social influences-based education. Rather, smokers indicated that a variety of punishments and, in particular, a variety of rewards were the approaches most likely to deter them from smoking. The responses of black and lower income students were also notable regarding their evaluations of the effectiveness of social influences-based education. Significant numbers of students in both of these categories indicated that social influences-based education is not an effective deterrent to their smoking

    A change in temperature modulates defence to yellow (stripe) rust in wheat line UC1041 independently of resistance gene Yr36

    Background Rust diseases are of major importance in wheat production worldwide. With the constant evolution of new rust strains and their adaptation to higher temperatures, consistent and durable disease resistance is a key challenge. Environmental conditions affect resistance gene performance, but the basis for this is poorly understood. Results Here we show that a change in day temperature affects wheat resistance to Puccinia striiformis f. sp tritici (Pst), the causal agent of yellow (or stripe) rust. Using adult plants of near-isogenic lines UC1041 +/- Yr36, there was no significant difference between Pst percentage uredia coverage in plants grown at day temperatures of 18°C or 25°C in adult UC1041 + Yr36 plants. However, when plants were transferred to the lower day temperature at the time of Pst inoculation, infection increased up to two fold. Interestingly, this response was independent of Yr36, which has previously been reported as a temperature-responsive resistance gene as Pst development in adult UC1041 -Yr36 plants was similarly affected by the plants experiencing a temperature reduction. In addition, UC1041 -Yr36 plants grown at the lower temperature then transferred to the higher temperature were effectively resistant and a temperature change in either direction was shown to affect Pst development up to 8 days prior to inoculation. Results for seedlings were similar, but more variable compared to adult plants. Enhanced resistance to Pst was observed in seedlings of UC1041 and the cultivar Shamrock when transferred to the higher temperature. Resistance was not affected in seedlings of cultivar Solstice by a temperature change in either direction. Conclusions Yr36 is effective at 18°C, refining the lower range of temperature at which resistance against Pst is conferred compared to previous studies. Results reveal previously uncharacterised defence temperature sensitivity in the UC1041 background which is caused by a change in temperature and independently of Yr36. This novel phenotype is present in some cultivars but absent in others, suggesting that Pst defence may be more stable in some cultivars than others when plants are exposed to varying temperatures

    Interventions targeted at women to encourage the uptake of cervical screening

    BACKGROUND World-wide, cervical cancer is the second most common cancer in women. Increasing the uptake of screening, alongside increasing informed choice is of great importance in controlling this disease through prevention and early detection. OBJECTIVES To assess the effectiveness of interventions aimed at women, to increase the uptake, including informed uptake, of cervical cancer screening. SEARCH STRATEGY We searched the Cochrane Gynaecological Cancer Group Trials Register, Cochrane Central Register of Controlled Trials (CENTRAL), Issue 1, 2009. MEDLINE, EMBASE and LILACS databases up to March 2009. We also searched registers of clinical trials, abstracts of scientific meetings, reference lists of included studies and contacted experts in the field. SELECTION CRITERIA Randomised controlled trials (RCTs) of interventions to increase uptake/informed uptake of cervical cancer screening. DATA COLLECTION AND ANALYSIS Two review authors independently abstracted data and assessed risk of bias. Where possible the data were synthesised in a meta-analysis. MAIN RESULTS Thirty-eight trials met our inclusion criteria. These trials assessed the effectiveness of invitational and educational interventions, counselling, risk factor assessment and procedural interventions. Heterogeneity between trials limited statistical pooling of data. Overall, however, invitations appear to be effective methods of increasing uptake. In addition, there is limited evidence to support the use of educational materials. Secondary outcomes including cost data were incompletely documented so evidence was limited. Most trials were at moderate risk of bias. Informed uptake of cervical screening was not reported in any trials. AUTHORS' CONCLUSIONS There is evidence to support the use of invitation letters to increase the uptake of cervical screening. There is limited evidence to support educational interventions but it is unclear what format is most effective. The majority of the studies are from developed countries and so the relevance to developing countries is unclear.Output Type: Revie

    Childhood overeating is associated with adverse cardiometabolic and inflammatory profiles in adolescence

    Childhood eating behaviour contributes to the rise of obesity and related noncommunicable disease worldwide. However, we lack a deep understanding of biochemical alterations that can arise from aberrant eating behaviour. In this study, we prospectively associate longitudinal trajectories of childhood overeating, undereating, and fussy eating with metabolic markers at age 16 years to explore adolescent metabolic alterations related to specific eating patterns in the first 10 years of life. Data are from the Avon Longitudinal Study of Parents and Children (n = 3104). We measure 158 metabolic markers with a high-throughput (1H) NMR metabolomics platform. Increasing childhood overeating is prospectively associated with an adverse cardiometabolic profile (i.e., hyperlipidemia, hypercholesterolemia, hyperlipoproteinemia) in adolescence; whereas undereating and fussy eating are associated with lower concentrations of the amino acids glutamine and valine, suggesting a potential lack of micronutrients.     A large bioassay identifies Stb resistance genes that provide broad resistance against Septoria tritici blotch disease in the UK

    Introduction: Septoria tritici blotch (STB) is one of the most damaging fungal diseases of wheat in Europe, largely due to the paucity of effective resistance genes against it in breeding materials. Currently dominant protection methods against this disease, e.g. fungicides and the disease resistance genes already deployed, are losing their effectiveness. Therefore, it is vital that other available disease resistance sources are identified, understood and deployed in a manner that maximises their effectiveness and durability. Methods: In this study, we assessed wheat genotypes containing nineteen known major STB resistance genes (Stb1 through to Stb19) or combinations thereof against a broad panel of 93 UK Zymoseptoria tritici isolates. Seedlings were inoculated using a cotton swab and monitored for four weeks. Four infection-related phenotypic traits were visually assessed. These were the days post infection to the development of first symptoms and pycnidia, percentage coverage of the infected leaf area with chlorosis/necrosis and percentage coverage of the infected leaf area with pycnidia. Results: The different Stb genes were found to vary greatly in the levels of protection they provided, with pycnidia coverage at four weeks differing significantly from susceptible controls for every tested genotype. Stb10, Stb11, Stb12, Stb16q, Stb17, and Stb19 were identified as contributing broad spectrum disease resistance, and synthetic hexaploid wheat lines were identified as particularly promising sources of broadly effective STB resistances. Discussion: No single Z. tritici isolate was found to be virulent against all tested resistance genes. Wheat genotypes carrying multiple Stb genes were found to provide higher levels of resistance than expected given their historical levels of use. Furthermore, it was noted that disease resistance controlled by different Stb genes was associated with different levels of chlorosis, with high levels of early chlorosis in some genotypes correlated with high resistance to fungal pycnidia development, potentially suggesting the presence of multiple resistance mechanisms. The knowledge obtained here will aid UK breeders in prioritising Stb genes for future breeding programmes, in which optimal combinations of resistance genes could be pyramided. In addition, this study identified the most interesting Stb genes for cloning and detailed functional analysis

    Unexpected diversity and complexity of the Guerrero Negro hypersaline microbial mat

The definitive version was published in Applied and Environmental Microbiology 72 (2006): 3685-3695, doi:10.1128/AEM.72.5.3685-3695.2006.We applied nucleic acids-based molecular methods, combined with estimates of biomass (ATP), pigments and microelectrode measurements of chemical gradients, to map microbial diversity vertically on the mm-scale in a hypersaline microbial mat from Guerrero Negro, Baja California Sur, Mexico. To identify the constituents of the mat, small-subunit ribosomal RNA genes were amplified by PCR from community genomic DNA extracted from layers, cloned and sequenced. Bacteria dominated the mat and displayed unexpected and unprecedented diversity. The majority (1336) of 1586 bacterial 16S rRNA sequences generated were unique, representing 752 species (≥97% rRNA sequence identity) in 42 of the main bacterial phyla, including 15 novel candidate phyla. The diversity of the mat samples differentiated according to the chemical milieu defined by concentrations of O2 and H2S. Chloroflexi formed the majority of the biomass by percentage of bulk rRNA and of clones in rRNA gene libraries. This result contradicts the general belief that Cyanobacteria dominate these communities. Although Cyanobacteria constituted a large fraction of the biomass in the upper few mm (>80% of total rRNA and photosynthetic pigments), Chloroflexi sequences were conspicuous throughout the mat. Filamentous Chloroflexi were identified by fluorescent in-situ hybridization within the polysaccharide sheaths of the prominent cyanobacterium Microcoleus chthonoplastes in addition to free-living in the mat. The biological complexity of the mat far exceeds that observed in other polysaccharide-rich microbial ecosystems, such as human and mouse distal guts, and suggests that positive feedbacks exist between chemical complexity and biological diversity.R.     Interleukin 4 and interleukin 13 downregulate the lipopolysaccharide-mediated inflammatory response by human gestation-associated tissues†

    Local and systemic inflammation is a key feature of preterm and term labour and delivery. Pro-inflammatory mediators are produced by gestation-associated tissues in response to a broad range of pathogen-associated molecular patterns (PAMPs) or damage-associated molecular patterns (DAMPs). Interleukin (IL)-4, IL-10 and IL-13 are well recognised anti-inflammatory cytokines and might have potential as anti-inflammatory therapies to prevent preterm birth. The objective of this study was to determine if IL-4 and IL-13 exert anti-inflammatory effects on lipopolysaccharide (LPS)-stimulated production of pro-inflammatory cytokines produced by human term gestation-associated tissues (placenta, choriodecidua and amnion). Both IL-4 and IL-13 reduced LPS-stimulated IL-1β and macrophage inflammatory protein (MIP)-1α with this effect diminishing with delay to exposure to IL-4 or IL-13; there was no effect on LPS-stimulated prostaglandin production. IL-4 receptor α (IL-4Rα) was expressed throughout the placenta, choriodecidua and amnion, and the inhibitory effects of IL-4 and IL-13 were IL-4Rα-dependent. A combination of IL-4 and IL-13 did not enhance the anti-inflammatory potential of either cytokine; however, a combination of IL-4 and IL-10 had a greater anti-inflammatory effect than either cytokine alone. These findings demonstrate that human term gestation-associated tissues are responsive to the anti-inflammatory cytokines IL-4 and IL-13, which could down-regulate LPS induced cytokine production in these tissues. Anti-inflammatory cytokines might offer an adjunct to existing therapeutics to prevent adverse obstetric outcome
