    Hydrocarbon molar water solubility predicts NMDA vs. GABAA receptor modulation.

    BackgroundMany anesthetics modulate 3-transmembrane (such as NMDA) and 4-transmembrane (such as GABAA) receptors. Clinical and experimental anesthetics exhibiting receptor family specificity often have low water solubility. We hypothesized that the molar water solubility of a hydrocarbon could be used to predict receptor modulation in vitro.MethodsGABAA (α1β2γ2s) or NMDA (NR1/NR2A) receptors were expressed in oocytes and studied using standard two-electrode voltage clamp techniques. Hydrocarbons from 14 different organic functional groups were studied at saturated concentrations, and compounds within each group differed only by the carbon number at the ω-position or within a saturated ring. An effect on GABAA or NMDA receptors was defined as a 10% or greater reversible current change from baseline that was statistically different from zero.ResultsHydrocarbon moieties potentiated GABAA and inhibited NMDA receptor currents with at least some members from each functional group modulating both receptor types. A water solubility cut-off for NMDA receptors occurred at 1.1 mM with a 95% CI = 0.45 to 2.8 mM. NMDA receptor cut-off effects were not well correlated with hydrocarbon chain length or molecular volume. No cut-off was observed for GABAA receptors within the solubility range of hydrocarbons studied.ConclusionsHydrocarbon modulation of NMDA receptor function exhibits a molar water solubility cut-off. Differences between unrelated receptor cut-off values suggest that the number, affinity, or efficacy of protein-hydrocarbon interactions at these sites likely differ

    Flight test and analyses of the B-1 structural mode control system at supersonic flight conditions

    A practical structural mode control system (SMCS) that could be turned on at takeoff and be left on for the entire flight was demonstrated. The SMCS appears to be more effective in damping the key fuselage bending modes at supersonic speeds than at the design point of Mach 0.85 (for fixed gains). The SMCS has an adverse effect on high frequency symmetric modes; however, this adverse effect did not make the system unstable and does not appear to affect ride quality performance. The vertical ride quality analyses indicate that the basic configuration without active systems is satisfactory for long term exposure. If clear air turbulence were to be encountered, indications are that the SMCS would be very effective in reducing the adverse accelerations. On the other hand, lateral ride quality analyses indicate that the aircraft with the SMCS on does not quite meet the long term exposure criteria, but would be satisfactory for shot term exposure at altitude. Again, the lateral SMCS was shown to be very effective in reducing peak lateral accelerations

    Trading Behavior Between Conspecifics in Chimpanzees, Pan Troglodytes

    Bartering of commodities between individuals is a hallmark of human behavior that is not commonly seen in other species. This is difficult to explain because barter is mutually beneficial, and appears to be within the cognitive capabilities of many species. It may be that other species do not recognize the gains of trade, or that they do not experience conditions (e.g., low risk) in which barter is most beneficial. To answer these questions, we instituted a systematic study of chimpanzees’ ability to barter with each other when doing so materially benefits them. Using tokens derived from symbols they have used since infancy, pairs of adult chimpanzees could trade between themselves to obtain tokens needed to get foods. Subjects flexibly used the tokens to obtain foods from an experimenter; however, they did not spontaneously trade with their partner. After extensive training, subjects engaged in accurate trade behavior as long as an experimenter enforced the structure of the interaction; however, trade between partners disappeared when this enforcement was removed. We discuss possible reasons for these findings as well as implications for the evolution of barter across the primate lineage
