    An Integrin-Linked Machinery of Cytoskeletal Regulation that Enables Experimental Tumor Initiation and Metastatic Colonization

    Recently extravasated metastatic cancer cells use the Rif/mDia2 actin-nucleating/polymerizing machinery in order to extend integrin β1-containing, filopodium-like protrusions (FLPs), which enable them to interact productively with the surrounding extracellular matrix; this process governs the initial proliferation of these cancer cells. Here, we identify the signaling pathway governing FLP lifetime, which involves integrin-linked kinase (ILK) and β-parvin, two integrin:actin-bridging proteins that block cofilin-mediated actin-filament severing. Notably, the combined actions of Rif/mDia2 and ILK/β-parvin/cofilin pathways on FLPs are required not only for metastatic outgrowth but also for primary tumor formation following experimental implantation.     Electron density and collision frequency of microwave‐resonant‐cavity‐produced discharges

    A review of perturbation diagnostics applied to microwave resonant cavity discharges is presented. The classical microwave perturbation technique examines the shift in the resonant frequency and cavity quality factor of the resonant cavity caused by low‐electron density discharges. However, the modifications presented allow the analysis to be applied to discharges with electron densities beyond the limit predicted by perturbation theory. An ‘‘exact’’ perturbation analysis is presented which models the discharge as a separate dielectric, thereby removing the restrictions on electron density imposed by the classical technique. The ‘‘exact’’ method also uses measurements of the shifts in the resonant conditions of the cavity. Third, an electromagnetic analysis is presented which uses a characteristic equation, based upon Maxwell’s laws, and predicts the discharge conductivity based upon measurements of a complex axial wave number. By allowing the axial wave number of the electromagnetic fields to be complex, the fields are experimentally and theoretically shown to be spatially attenuated. The diagnostics are applied to continuous‐wave microwave (2.45 GHz) discharges produced in an Asmussen resonant cavity. Double Langmuir probes, placed directly in the discharge at the point where the radial electric field is zero, act as a comparison with the analytic diagnostics. Microwave powers ranging from 30 to 100 W produce helium and nitrogen discharges with pressures ranging from 0.5 to 6 Torr.     Maritime Economics in a Post-Expansion Panama Canal Era

    (First paragraph) The 2016 opening of an expanded Panama Canal will allow for Post-Panamax containerships up to 12 500 twenty-foot equivalent unit (TEU) in size to transit the Panama Canal. In response, some US East Coast container ports are having their channels and berths dredged deeper—to allow Post-Panamax containerships from Asia (transiting the expanded canal) to call at their ports. What are the implications for the US West Coast ports? Will there be a cargo shift from West Coast to East Coast ports? These topics as well as the impacts of other changes in global shipping lanes (e.g., the Suez Canal and the Arctic shipping lanes) on global trade and ports in the Post-Expansion Panama Canal Era were discussed in various sessions of the International Association of Maritime Economists (IAME) 2014 Conference. This special issue is dedicated to the study of the above impacts. The goal of this special issue is to encourage research in this important area by highlighting the influence of the Panama Canal expansion to the global maritime sectors and examining the potentially dramatic changes in the Post-Expansion Era. Hence, five IAME conference papers and an additional paper by Ducruet are chosen for this reason

    The Outgrowth of Micrometastases Is Enabled by the Formation of Filopodium-like Protrusions

    Disseminated cancer cells that have extravasated into the tissue parenchyma must interact productively with its extracellular matrix components to survive, proliferate, and form macroscopic metastases. The biochemical and cell biologic mechanisms enabling this interaction remain poorly understood. We find that the formation of elongated integrin β1-containing adhesion plaques by cancer cells that have extravasated into the lung parenchyma enables the proliferation of these cells via activation of focal adhesion kinase. These plaques originate in and appear only after the formation of filopodium-like protrusions (FLP) that harbor integrin β1 along their shafts. The cytoskeleton-regulating proteins Rif and mDia2 contribute critically to the formation of these protrusions and thereby enable the proliferation of extravasated cancer cells. Hence, the formation of FLPs represents a critical rate-limiting step for the subsequent development of macroscopic metastases. SIGNIFICANCE: Although the mechanisms of metastatic dissemination have begun to be uncovered, those involved in the establishment of extravasated cancer cells in foreign tissue microenvironments remained largely obscure. We have studied the behavior of recently extravasated cancer cells in the lungs and identified a series of cell biologic processes involving the formation of filopodium-like protrusions and the subsequent development of elongated, mature adhesion plaques, which contribute critically to the rapid proliferation of the micrometastatic cells and thus are prerequisites to the eventual lung colonization by these cells.National Institutes of Health (U.S.) (Grant P01-CA080111

    Prognostic models for predicting posttraumatic seizures during acute hospitalization, and at 1 and 2 years following traumatic brain injury

    Objective Posttraumatic seizures (PTS) are well-recognized acute and chronic complications of traumatic brain injury (TBI). Risk factors have been identified, but considerable variability in who develops PTS remains. Existing PTS prognostic models are not widely adopted for clinical use and do not reflect current trends in injury, diagnosis, or care. We aimed to develop and internally validate preliminary prognostic regression models to predict PTS during acute care hospitalization, and at year 1 and year 2 postinjury. Methods Prognostic models predicting PTS during acute care hospitalization and year 1 and year 2 post-injury were developed using a recent (2011–2014) cohort from the TBI Model Systems National Database. Potential PTS predictors were selected based on previous literature and biologic plausibility. Bivariable logistic regression identified variables with a p-value < 0.20 that were used to fit initial prognostic models. Multivariable logistic regression modeling with backward-stepwise elimination was used to determine reduced prognostic models and to internally validate using 1,000 bootstrap samples. Fit statistics were calculated, correcting for overfitting (optimism). Results The prognostic models identified sex, craniotomy, contusion load, and pre-injury limitation in learning/remembering/concentrating as significant PTS predictors during acute hospitalization. Significant predictors of PTS at year 1 were subdural hematoma (SDH), contusion load, craniotomy, craniectomy, seizure during acute hospitalization, duration of posttraumatic amnesia, preinjury mental health treatment/psychiatric hospitalization, and preinjury incarceration. Year 2 significant predictors were similar to those of year 1: SDH, intraparenchymal fragment, craniotomy, craniectomy, seizure during acute hospitalization, and preinjury incarceration. Corrected concordance (C) statistics were 0.599, 0.747, and 0.716 for acute hospitalization, year 1, and year 2 models, respectively. Significance The prognostic model for PTS during acute hospitalization did not discriminate well. Year 1 and year 2 models showed fair to good predictive validity for PTS. Cranial surgery, although medically necessary, requires ongoing research regarding potential benefits of increased monitoring for signs of epileptogenesis, PTS prophylaxis, and/or rehabilitation/social support. Future studies should externally validate models and determine clinical utility

    Telomerase Maintains Telomere Structure in Normal Human Cells

    AbstractIn normal human cells, telomeres shorten with successive rounds of cell division, and immortalization correlates with stabilization of telomere length. These observations suggest that human cancer cells achieve immortalization in large part through the illegitimate activation of telomerase expression. Here, we demonstrate that the rate-limiting telomerase catalytic subunit hTERT is expressed in cycling primary presenescent human fibroblasts, previously believed to lack hTERT expression and telomerase activity. Disruption of telomerase activity in normal human cells slows cell proliferation, restricts cell lifespan, and alters the maintenance of the 3′ single-stranded telomeric overhang without changing the rate of overall telomere shortening. Together, these observations support the view that telomerase and telomere structure are dynamically regulated in normal human cells and that telomere length alone is unlikely to trigger entry into replicative senescence

    Contacts and behaviours of university students during the COVID-19 pandemic at the start of the 2020/2021 academic year

    University students have unique living, learning and social arrangements which may have implications for infectious disease transmission. To address this data gap, we created CONQUEST (COroNavirus QUESTionnaire), a longitudinal online survey of contacts, behaviour, and COVID-19 symptoms for University of Bristol (UoB) staff/students. Here, we analyse results from 740 students providing 1261 unique records from the start of the 2020/2021 academic year (14/09/2020–01/11/2020), where COVID-19 outbreaks led to the self-isolation of all students in some halls of residences. Although most students reported lower daily contacts than in pre-COVID-19 studies, there was heterogeneity, with some reporting many (median = 2, mean = 6.1, standard deviation = 15.0; 8% had ≥ 20 contacts). Around 40% of students’ contacts were with individuals external to the university, indicating potential for transmission to non-students/staff. Only 61% of those reporting cardinal symptoms in the past week self-isolated, although 99% with a positive COVID-19 test during the 2 weeks before survey completion had self-isolated within the last week. Some students who self-isolated had many contacts (mean = 4.3, standard deviation = 10.6). Our results provide context to the COVID-19 outbreaks seen in universities and are available for modelling future outbreaks and informing policy

    Leptospirosis during Dengue Outbreak, Bangladesh

    We collected acute-phase serum samples from febrile patients at 2 major hospitals in Dhaka, Bangladesh, during an outbreak of dengue fever in 2001. A total of 18% of dengue-negative patients tested positive for leptospirosis. The case-fatality rate among leptospirosis patients (5%) was higher than among dengue fever patients (1.2%)
