    Conception optimale des moteurs à réluctance variable à commutation électronique pour la traction des véhicules électriques légers

    Le domaine de la traction électrique a suscité un très grand intérêt dans les dernières années. La conception optimale de l'ensemble moteur électrique de traction onduleur doit prendre en compte une variété de critères et contraintes. Étant donnée la liaison entre la géométrie du moteur et la stratégie de commande de l'onduleur, l'optimisation de l'ensemble de traction doit prendre en considération, en même temps, les deux composants.L'objectif de la thèse est la conception d'un outil d'optimisation appliqué à un système de traction électrique légère qu'emploie un moteur à réluctance variable alimenté (MRVCE) par un onduleur triphasé en pont complet. Le MRVCE est modélisé en utilisant la technique par réseau de perméances. En même temps, la technique de commande électronique peut être facilement intégrée dans le modèle pour effectuer l'analyse dynamique du fonctionnement du moteur. L'outil d'optimisation réalisé utilise l'algorithme par essaim de particules, modifié pour résoudre des problèmes multi-objectif. Les objectifs sont liés à la qualité des caractéristiques de fonctionnement du moteur, en temps que les variables d'optimisation concernent la géométrie du moteur aussi que la technique de commande. Les performances de l'algorithme sont comparées avec ceux de l'algorithme génétique (NSGA-II) et d'une implémentation classique de l'algorithme par essaim de particules multi-objectif.Finalement, un prototype de moteur à réluctance variable est construit et le fonctionnement du MRVCE alimenté depuis l'onduleur triphasé en pont complet est implémenté et les outils de modélisation et d'optimisation sont validésThe interest for the electric traction applications has been growing in the last few years. The optimal design of the electric motor and of the inverter that powers it needs to consider a long list of restrictions and criteria. Because of the fact that the geometry of the motor and the switching strategy are closely linked, the optimization of the traction solution needs to consider both, at the same time.The objective of this thesis is the development of an optimization tool applied for the optimization of an electric traction solution that uses the switched reluctance motor (SRM) fed from a three phase full bridge inverter. The SRM is modeled using Permeance Network Analysis (PNA). The switching technique can be easily integrated in the model, which gives the possibility to run a dynamic analysis. The optimization tool created uses the Particle Swarm Optimization (PSO) algorithm, modified for multi-objective problems. The algorithms performances are compared with those of the Genetic Algorithm, using the NSGA-II multi-objective technique and with a classic version of multiple objective particle swarm optimizer (MOPSO).Finally, a SRM prototype is constructed and the drive solution using a full-bridge three phase inverter is implemented.     A Plasmodium membrane receptor platform integrates cues for egress and invasion in blood forms and activation of transmission stages

    Critical events in the life cycle of malaria-causing parasites depend on cyclic guanosine monophosphate homeostasis by guanylyl cyclases (GCs) and phosphodiesterases, including merozoite egress or invasion of erythrocytes and gametocyte activation. These processes rely on a single GCalpha, but in the absence of known signaling receptors, how this pathway integrates distinct triggers is unknown. We show that temperature-dependent epistatic interactions between phosphodiesterases counterbalance GCalpha basal activity preventing gametocyte activation before mosquito blood feed. GCalpha interacts with two multipass membrane cofactors in schizonts and gametocytes: UGO (unique GC organizer) and SLF (signaling linking factor). While SLF regulates GCalpha basal activity, UGO is essential for GCalpha up-regulation in response to natural signals inducing merozoite egress and gametocyte activation. This work identifies a GC membrane receptor platform that senses signals triggering processes specific to an intracellular parasitic lifestyle, including host cell egress and invasion to ensure intraerythrocytic amplification and transmission to mosquitoes

    : Regulated exocytosis by secretory organelles is important for malaria parasite invasion and egress. Many parasite effector proteins, including perforins, adhesins, and proteases, are extensively proteolytically processed both pre- and postexocytosis. Here we report the multistage antiplasmodial activity of the aspartic protease inhibitor hydroxyl-ethyl-amine-based scaffold compound 49c. This scaffold inhibits the preexocytosis processing of several secreted rhoptry and microneme proteins by targeting the corresponding maturases plasmepsins IX (PMIX) and X (PMX), respectively. Conditional excision of PMIX revealed its crucial role in invasion, and recombinantly active PMIX and PMX cleave egress and invasion factors in a 49c-sensitive manner.<br/

    Pseudomonas protegens are multi-talented plant-colonizing bacteria that suppress plant pathogens and stimulate plant defenses. In addition, they are capable of invading and killing agriculturally important plant pest insects that makes them promising candidates for biocontrol applications. Here we assessed the role of type VI secretion system (T6SS) components of type strain CHA0 during interaction with larvae of the cabbage pest Pieris brassicae. We show that the T6SS core apparatus and two VgrG modules, encompassing the respective T6SS spikes (VgrG1a and VgrG1b) and associated effectors (RhsA and Ghh1), contribute significantly to insect pathogenicity of P. protegens in oral infection assays but not when bacteria are injected directly into the hemolymph. Monitoring of the colonization levels of P. protegens in the gut, hemolymph, and excrements of the insect larvae revealed that the invader relies on T6SS and VgrG1a module function to promote hemocoel invasion. A 16S metagenomic analysis demonstrated that T6SS-supported invasion by P. protegens induces significant changes in the insect gut microbiome affecting notably Enterobacteriaceae, a dominant group of the commensal gut bacteria. Our study supports the concept that pathogens deploy T6SS-based strategies to disrupt the commensal microbiota in order to promote host colonization and pathogenesis

    IMGT/V-QUEST: the highly customized and integrated system for IG and TR standardized V-J and V-D-J sequence analysis

    IMGT/V-QUEST is the highly customized and integrated system for the standardized analysis of the immunoglobulin (IG) and T cell receptor (TR) rearranged nucleotide sequences. IMGT/V-QUEST identifies the variable (V), diversity (D) and joining (J) genes and alleles by alignment with the germline IG and TR gene and allele sequences of the IMGT reference directory. New functionalities were added through a complete rewrite in Java. IMGT/V-QUEST analyses batches of sequences (up to 50) in a single run. IMGT/V-QUEST describes the V-REGION mutations and identifies the hot spot positions in the closest germline V gene. IMGT/V-QUEST can detect insertions and deletions in the submitted sequences by reference to the IMGT unique numbering. IMGT/V-QUEST integrates IMGT/JunctionAnalysis for a detailed analysis of the V-J and V-D-J junctions, and IMGT/Automat for a full V-J- and V-D-J-REGION annotation. IMGT/V-QUEST displays, in ‘Detailed view’, the results and alignments for each submitted sequence individually and, in ‘Synthesis view’, the alignments of the sequences that, in a given run, express the same V gene and allele. The ‘Advanced parameters’ allow to modify default parameters used by IMGT/V-QUEST and IMGT/JunctionAnalysis according to the users’ interest.
