    Petrography and mineralogy of the white marble and black stone of Göktepe (Mugla, Turkey) used in antiquity: New data for provenance determination

    The discovery near Göktepe (Mugla province, Western Turkey) of an ancient quarrying site of white marbles and black stones has recently been reported by some authors. Assigning the provenance of stone from ancient artifacts to Göktepe is currently possible mainly thanks to chemical, EPR and MGS data. Petrographic description, which many researchers use to characterize ancient marbles, is still incomplete. Several thin sections of both types of stone were thus examined in this study, and also used for cathodoluminescence analysis. As the rock is >99% calcite, trace minerals could only be detected in some samples by XRD analysis of insoluble residues after acetic acid attack. Data on strontium and manganese contents and carbon and oxygen isotopes were also recorded, for better understanding of some petrographic features. A new method of grain size characterization was tentatively introduced to improve the description of grain size variability in the white marble. Microstructure and grain size measurements on thin sections of this marble identify two petrographic varieties: the first is extremely fine with signs of dynamic recrystallization, and the second exhibits texture and MGS similar to those of Carrara marble. Statuary samples of white marble from Villa Adriana (Tivoli, Rome), preliminarily assigned in a previous study partly to Carrara and partly to Göktepe quarry, are reconsidered here. A certain degree of variability was found in the structures and textures in the thin sections of the Göktepe black stone. It may have undergone transformations at an advanced stage of diagenesis. One important source of this variability seems to be a fluid alteration event, revealed by both isotopic and chemical data and trace mineral assemblages

    Grotta Romanelli (Southern Italy, Apulia). Legacies and issues in excavating a key site for the Pleistocene of the Mediterranean

    Grotta Romanelli, located on the Adriatic coast of southern Apulia (Italy), is considered a key site for the Mediterranean Pleistocene for its archaeological and palaeontological contents. The site, discovered in 1874, was re-evaluated only in 1900, when P. E. Stasi realised that it contained the first evidence of the Palaeolithic in Italy. Starting in 1914, G. A. Blanc led a pioneering excavation campaign, for the first-time using scientific methods applied to systematic palaeontological and stratigraphical studies. Blanc proposed a stratigraphic framework for the cave. Different dating methods (C-14 and U/Th) were used to temporally constrain the deposits. The extensive studies of the cave and its contents were mostly published in journals with limited distribution and access, until the end of the 1970s, when the site became forgotten. In 2015, with the permission of the authorities, a new excavation campaign began, led by a team from Sapienza University of Rome in collaboration with IGAG CNR and other research institutions. The research team had to deal with the consequences of more than 40 years of inactivity in the field and the combined effect of erosion and legal, as well as illegal, excavations. In this paper, we provide a database of all the information published during the first 70 years of excavations and highlight the outstanding problems and contradictions between the chronological and geomorphological evidence, the features of the faunal assemblages and the limestone artefacts

    Il dispositivo teatrale alla prova del Covid-19. Mediatizzazione, liveness e pubblici

    The COVID-19 emergency had a profound impact on the artistic and cultural sectors, and on performing arts in particular. The lockdown required the suspension of all live performances and rehearsals, including the cancellation of seasons and festivals. Because physical proximity is an essential component of the live shows, this sector will be on pause longer than the others. Since the early stages of the lockdown, the Italian theatre field developed several online initiatives to counterbalance the suspension of its activities. These efforts aimed at keeping the relationship with the remote theatre audiences, by extending the presence of artists, theatres and performances in the online context. However, they also provided an opportunity to reflect at large on the digital transformations of performing arts. The following contribution reflects on how we are researching the online response of the theatrical sector from the perspective of sociology and media studies. The paper aims to contextualise the phenomenon within the processes of theatre mediatisation and digital transformation of liveness, and to the present what we think are the most urgent research questions in this direction. The first part of the article introduces the theoretical premises of the investigation. We present the frame of theatre mediatisation by analysing three interrelated processes: the mediatisation of dramaturgy through the concept of transmedia; the mediatisation of theatrical presence, with the debate on digital liveness, and the mediatisation of the theatrical relationship through social media. The second part will analyse some of the main online initiatives of the theatrical sector, observing how they fit into the previously introduced mediatisation processes. The third part will observe how users have responded to the initiatives presented on social media by some of the main Italian theatres. In the conclusions, we will discuss which research questions we consider crucial to connect the analysis of this critical moment to the main themes of sociological and media studies research on performing arts

    Grape berry responses to sequential flooding and heatwave events: a physiological, transcriptional, and metabolic overview

    Grapevine cultivation, such as the whole horticulture, is currently challenged by several factors, among which the extreme weather events occurring under the climate change scenario are the most relevant. Within this context, the present study aims at characterizing at the berry level the physiological response of Vitis vinifera cv. Sauvignon Blanc to sequential stresses simulated under a semi-controlled environment: flooding at bud-break followed by multiple summer stress (drought plus heatwave) occurring at pre-vèraison. Transcriptomic and metabolomic assessments were performed through RNASeq and NMR, respectively. A comprehensive hormone profiling was also carried out. Results pointed out a different response to the heatwave in the two situations. Flooding caused a developmental advance, determining a different physiological background in the berry, thus affecting its response to the summer stress at both transcriptional levels, with the upregulation of genes involved in oxidative stress responses, and metabolic level, with the increase in osmoprotectants, such as proline and other amino acids. In conclusion, sequential stress, including a flooding event at bud-break followed by a summer heatwave, may impact phenological development and berry ripening, with possible consequences on berry and wine quality. A berry physiological model is presented that may support the development of sustainable vineyard management solutions to improve the water use efficiency and adaptation capacity of actual viticultural systems to future scenario

    SeqDeχ : a sequence deconvolution tool for genome separation of endosymbionts from mixed sequencing samples

    In recent years, the advent of NGS technology has made genome sequencing much cheaper than in the past; the high parallelization capability and the possibility to sequence more than one organism at once have opened the door to processing whole symbiotic consortia. However, this approach needs the development of specific bioinformatics tools able to analyze these data. In this work, we describe SeqDex, a tool that starts from a preliminary assembly obtained from sequencing a mixture of DNA from different organisms, to identify the contigs coming from one organism of interest. SeqDex is a fully automated machine learning-based tool exploiting partial taxonomic affiliations and compositional analysis to predict the taxonomic affiliations of contigs in an assembly. In literature, there are few methods able to deconvolve host-symbiont datasets, and most of them heavily rely on user curation and are therefore time consuming. The problem has strong similarities with metagenomic studies, where mixed samples are sequenced and the bioinformatics challenge is trying to separate contigs on the basis of their source organism; however, in symbiotic systems, additional information can be exploited to improve the output. To assess the ability of SeqDex to deconvolve host-symbiont datasets, we compared it to state-of-the-art methods for metagenomic binning and for host-symbiont deconvolution on three study cases. The results point out the good performances of the presented tool that, in addition to the ease of use and customization potential, make SeqDex a useful tool for rapid identification of endosymbiont sequences

    Baseline serum TSH levels predict the absence of thyroid dysfunction in cancer patients treated with immunotherapy

    Purpose Immunotherapy against immune checkpoints has significantly improved survival both in metastatic and adjuvant setting in several types of cancers. Thyroid dysfunction is the most common endocrine adverse event reported. Patients who are at risk of developing thyroid dysfunction remain to be defined. We aimed to identify predictive factors for the develop- ment of thyroid dysfunction during immunotherapy. Methods This is a retrospective study including a total of 68 patients who were treated with immune checkpoint inhibi- tors (ICIs) for metastatic or unresectable advanced cancers. The majority of patients were treated with anti-PD1 drugs in monotherapy or in combination with anti-CTLA4 inhibitors. Thyroid function and anti-thyroid antibodies, before starting immunotherapy and during treatment, were evaluated. Thyroid ultrasound was also performed in a subgroup of patients at the time of enrolment in the study. Results Eleven out of 68 patients (16.1%) developed immune-related overt thyroid dysfunction. By ROC curve analysis, we found that a serum TSH cut-off of 1.72 mUI/l, at baseline, had a good diagnostic accuracy in identifying patients without overt thyroid dysfunction (NPV = 100%, p = 0.0029). At multivariate analysis, both TSH and positive anti-thyroid antibod- ies (ATAbs) levels, before ICIs treatment, were independently associated with the development of overt thyroid dysfunction during immunotherapy (p = 0.0001 and p = 0.009, respectively). Conclusions Pre-treatment serum TSH and ATAbs levels may help to identify patients at high risk for primary thyroid dysfunction. Our study suggests guidance for an appropriate timely screening and for a tailored management of thyroid dysfunctions in patients treated with ICIs
