    Prelamin A Influences a Program of Gene Expression In Regulation of Cell Cycle Control

    The A-type lamins are intermediate filament proteins that constitute a major part of the eukaryotic nuclear lamina—a tough, polymerized, mesh lining of the inner nuclear membrane, providing shape and structural integrity to the nucleus. Lamin A (LA) filaments also permeate the nucleoplasm, providing additional structural support, but also scaffolding numerous tethered molecules to stabilize, organize, and facilitate molecular interactions to accomplish critical functions of cellular metabolism. Over the past 2 decades, much attention has been focused on roles of LA in maintenance of nuclear structural integrity. Only since the late 1990s have scientists discovered the devastating effects of LA gene (LMNA) mutations, as they have associated hundreds of LMNA mutations to a large group of diseases, called laminopathies, with a broad spectrum of phenotypes, ranging from skeletal, muscular, and neurological defects, to defective lipid storage, to accelerated aging phenotypes in diseases called progerias. Recent advances demonstrate LA regulatory functions include cell signaling, cell cycle regulation, transcription, chromatin organization, viral egress, and DNA damage repair. Amidst the flurry of fascinating research, only recently have researchers begun to focus attention on the different isoforms that exist for LA, a precursor form among them. LA is initially synthesized as Prelamin A (PreA), and undergoes a series of modifications that truncate the protein to produce “mature” LA. Existence of the precursor form, and its complex maturation pathway, have puzzled researchers since their realization. With a pattern of expression related to cell cycle phase, we hypothesized a role for PreA in cell cycle control. To investigate, we have performed array studies to assess gene expression effects at the levels of transcript expression, protein expression, and phosphorylation modification status. Here, we present evidence for a PreA-mediated program of cell cycle regulatory gene and protein expression modulation. Implicated pathways include RB-E2F, p53, p27Kip1, FoxOs, p300, and the Cyclins, with additional evidence indicating a role for the Pin1 prolyl isomerase in mediating PreA regulation of the cell cycle

    Production of gaseous fuel by pyrolysis of municipal solid waste

    Pilot plant tests were conducted on a simulated solid waste which was a mixture of shredded newspaper, wood waste, polyethylene plastics, crushed glass, steel turnings, and water. Tests were conducted at 1400 F in a lead-bath pyrolyser. Cold feed was deaerated by compression and was dropped onto a moving hearth of molten lead before being transported to a sealed storage container. About 80 percent of the feed's organic content was converted to gaseous products which contain over 90 percent of the potential waste energy; 12 percent was converted to water; and 8 percent remained as partially pyrolyzed char and tars. Nearly half of the carbon in the feed is converted to benzene, toluene and medium-quality fuel gas, a potential credit of over $25 per ton of solid waste. The system was shown to require minimal preprocessing and less sorting then other methods

    Ion laser plasmas

    The typical noble gas ion laser plasma consists of a high-current-density glow discharge in a noble gas, in the presence of a magnetic field. Typical CW plasma conditions are current densities of 100 to 2000 A/cm^2, tube diameters of 1 to 10 mm, filling pressures of 0.1 to 1.0 torr, and an axial magnetic field of the order of 1000 G. Under these conditions the typical fractional ionization is about 2 percent and the electron temperature between 2 and 4 eV. Pulsed ion lasers typically use higher current densities and lower operating pressures. This paper discusses the properties of ion laser plasmas, in terms of both their external discharge parameters and their internal ion and excited state densities. The effect these properties have on laser operation is explained. Many interesting plasma effects, which are important in ion lasers, are given attention. Among these are discharge nonuniformity near tube constrictions, extremely high ion radial drift velocities, wall losses intermediate between ambipolar diffusion and free fall, gas pumping effects, and radiation trapping. The current status of ion laser technology is briefly reviewed

    Experimental studies on the impact properties of water ice

    Experimental studies on the impact of ice particles at very low velocity were continued. These measurements have applications in the dynamics of Saturn's rings. Initially data were obtained on the coefficient of restitution for ice spheres of one radius of curvature. The type of measurements were expanded to include restitution data for balls with a variety of surfaces as well as sticking forces between ice particles. Significant improvements were made to this experiment, the most important being the construction of a new apparatus. The new apparatus consists of a smaller version of the disk pendulum and a stainless steel, double-walled cryostat. The apparatus has proved to be a significant improvement over the old one. Measurements can now be made at temperatures near 90 K, comparable to the temperature of the environment of Saturn's rings, and with much greater temperature stability. It was found that a roughened contact surface or the presence of frost can cause a much larger change in the restitution measure than the geometrical effect of the radius of curvature

    Earth-like sand fluxes on Mars

    Strong and sustained winds on Mars have been considered rare, on the basis of surface meteorology measurements and global circulation models, raising the question of whether the abundant dunes and evidence for wind erosion seen on the planet are a current process. Recent studies showed sand activity, but could not determine whether entire dunes were moving—implying large sand fluxes—or whether more localized and surficial changes had occurred. Here we present measurements of the migration rate of sand ripples and dune lee fronts at the Nili Patera dune field. We show that the dunes are near steady state, with their entire volumes composed of mobile sand. The dunes have unexpectedly high sand fluxes, similar, for example, to those in Victoria Valley, Antarctica, implying that rates of landscape modification on Mars and Earth are similar
