Sistema di filologia computazionale per testi demotici
A project for a Demotic Inscriptions on Ostraka Database is being carried out in collaboration between ILC/CNR (Pisa), the Department of Electronic Engineering (Calabria University) and the Department of the Ancient World History (Egyptological section, Pisa University). The aim of the project is to analyse the digital colour images of demotic texts on ostraka (Medinet Madi, in Fayyum region) with the aid of computational tools. The module described in the paper is a neural component able to learn the graphical features of each demotic symbol, which has been previously segmented in the images thanks to a semiautomatic procedure. A specific neural network tries to recognize the text written in the images linking the symbols segmented within the ostraka images database to the correspondent symbols available on a virtual keyboard. The graphical interface is particularly useful for teaching and research activities on this type of archaeological documentation
Integrating Optical Wireless Communication Into an Optical Bifocal Metrology for Aerospace
Recently an innovative bifocal optical metrology method was proposed for space applications (e.g., rendez-vous and docking), based on unmodulated white LEDs. Here we design, realize, and test a solution that upgrades the metrology to include a digital communication feature, with no modification of the optical elements of the original system: indeed, the scheme exploits the same optical sources that are needed for metrology, which are now also working as optical antennas as their intensity is now modulated. At the receiver side, the conventional camera is now sided by a common photodiode. The system provides unidirectional data communication at 10 kbit/s speed. It is designed to support manoeuvres up to 400 m distance. The lab tests confirm the effectiveness of the proposed solutions, showing correct data transfer without any noticeable degradation of the metrology system
In silico identification of new putative pathogenic variants in the NEU1 sialidase gene affecting enzyme function and subcellular localization
The NEU1 gene is the first identified member of the human sialidases, glycohydrolitic enzymes that remove the terminal sialic acid from oligosaccharide chains. Mutations in NEU1 gene are causative of sialidosis (MIM 256550), a severe lysosomal storage disorder showing autosomal recessive mode of inheritance. Sialidosis has been classified into two subtypes: sialidosis type I, a normomorphic, late-onset form, and sialidosis type II, a more severe neonatal or early-onset form. A total of 50 causative mutations are reported in HGMD database, most of which are missense variants. To further characterize the NEU1 gene and identify new functionally relevant protein isoforms, we decided to study its genetic variability in the human population using the data generated by two large sequencing projects: the 1000 Genomes Project (1000G) and the NHLBI GO Exome Sequencing Project (ESP). Together these two datasets comprise a cohort of 7595 sequenced individuals, making it possible to identify rare variants and dissect population specific ones. By integrating this approach with biochemical and cellular studies, we were able to identify new rare missense and frameshift alleles in NEU1 gene. Among the 9 candidate variants tested, only two resulted in significantly lower levels of sialidase activity (pC and c.700G>A. These two mutations give rise to the amino acid substitutions p.V217A and p.D234N, respectively. NEU1 variants including either of these two amino acid changes have 44% and 25% residual sialidase activity when compared to the wild-type enzyme, reduced protein levels and altered subcellular localization. Thus they may represent new, putative pathological mutations resulting in sialidosis type I. The in silico approach used in this study has enabled the identification of previously unknown NEU1 functional alleles that are widespread in the population and could be tested in future functional studies
Effects of the current financial and economic crisis on the rural landscape as well as the agri-food sector in Europe and Central Asia
This paper reviews the expected effects of the current financial crisis and subsequent recession on the rural landscape, in particular the agri-food sector in Europe and Central Asia (ECA) on the basis of the structure of the rural economy and of different organisations and institutions. Empirical evidence suggests that the crisis has hit the ECA region the hardest. Agriculture contributes about 9% to gross domestic product (GDP) for the ECA region as a whole with 16% of the population being employed in the agricultural sector. As far as the impact of the financial crisis on the agri-food sector is concerned, there are a few interconnected issues: (1) reduction in income elastic food demand and commodity price decline, (2) loss of employment and earnings of rural people working in urban centres, implying also costly labour reallocation, (3) rising rural poverty originating mainly from lack of opportunities in the non-farm sector and a sizable decline of international remittances, (4) tightening of agricultural credit markets, and the (5) collapse of sectoral government support programs and social safety-net measures in many countries. The paper reveals how the crisis hit farming and broader agri-business differently in general and in the ECA sub-regions
Bypassing the Inertness of Aziridine/CO2 Systems to Access 5-Aryl-2-Oxazolidinones: Catalyst-Free Synthesis Under Ambient Conditions
The development of sustainable synthetic routes to access valuable oxazolidinones via CO2 fixation is an active research area, and the aziridine/carbon dioxide coupling has aroused a considerable interest. This reaction features a high activation barrier and thus requires a catalytic system, and may present some other critical issues. Here, the straightforward gram-scale synthesis of a series of 5-aryl-2-oxazolidinones was developed at ambient temperature and atmospheric CO2 pressure, in the absence of any catalyst/co-catalyst. The key to this innovative procedure consists in the direct transfer of the pre-formed amine/CO2 adduct (carbamate) to common aziridine precursors (dimethylsulfonium salts), replacing the classical sequential addition of amine (intermediate isolation of aziridine) and then CO2. The reaction mechanism was investigated by NMR spectroscopy and DFT calculations applied to model cases
Spatio-temporal analysis of prodelta dynamics by means of new satellite generation: the case of Po river by Landsat-8 data
Abstract This paper describes a procedure to perform spatio-temporal analysis of river plume dispersion in prodelta areas by multi-temporal Landsat-8-derived products for identifying zones sensitive to water discharge and for providing geostatistical patterns of turbidity linked to different meteo-marine forcings. In particular, we characterized the temporal and spatial variability of turbidity and sea surface temperature (SST) in the Po River prodelta (Northern Adriatic Sea, Italy) during the period 2013–2016. To perform this analysis, a two-pronged processing methodology was implemented and the resulting outputs were analysed through a series of statistical tools. A pixel-based spatial correlation analysis was carried out by comparing temporal curves of turbidity and SST hypercubes with in situ time series of wind speed and water discharge, providing correlation coefficient maps. A geostatistical analysis was performed to determine the spatial dependency of the turbidity datasets per each satellite image, providing maps of correlation and variograms. The results show a linear correlation between water discharge and turbidity variations in the points more affected by the buoyant plumes and along the southern coast of Po River delta. Better inverse correlation was found between turbidity and SST during floods rather than other periods. The correlation maps of wind speed with turbidity show different spatial patterns depending on local or basin-scale wind effects. Variogram maps identify different spatial anisotropy structures of turbidity in response to ambient conditions (i.e. strong Bora or Scirocco winds, floods). Since the implemented processing methodology is based on open source software and free satellite data, it represents a promising tool for the monitoring of maritime ecosystems and to address water quality analyses and the investigations of sediment dynamics in estuarine and coastal waters
Frequency Evaluation of the Interleukin-6 −174G>C Polymorphism and Homeostatic Iron Regulator (HFE) Mutations as Disease Modifiers in Patients Affected by Systemic Lupus Erythematosus and Rheumatoid Arthritis
Autoimmune diseases are generally characterized by a multifactorial etiology and are often associated with a genetic predisposition. Both iron metabolism and the inflammatory cytokine system have been shown to play a pivotal role in the dysregulation of the immune response in many different autoimmune conditions, rheumatologic diseases included. The purpose of this work was to analyze the frequency of mutations altering the expression of IL-6 or influencing iron metabolism in patients affected by autoimmune diseases such as Rheumatoid Arthritis (RA) and Systemic Lupus Erythematosus (SLE). In this study, 144 patients were enrolled: 77 and 67 patients were affected by RA and SLE, respectively. In these cohorts, the frequency of the IL-6 polymorphism −174G>C located in the IL-6 gene promoter was tested. Moreover, the frequencies of the three HFE gene variations associated with iron overload were analyzed: p.His63Asp, p.Ser65Cys and p.Cys282Tyr. The two mutations p.His63Asp and p.Ser65Cys in the HFE gene did not reach statistical significance in any of the comparisons, regardless of the statistical model, cohorts of patients and control populations analyzed. The frequencies of the p.Cys282Tyr mutation and the IL-6 polymorphism −174G>C were found to be overall significantly decreased in RA and SLE patients when the Dominant model and Allele contrast were adopted with both the Odds Ratio and Chi-square. Although further investigation is needed, the examination of the frequencies of the −174G>C IL-6 promoter polymorphism and HFE mutations may add some valuable information on the interplay linking iron metabolism, inflammation and immunity in autoimmune diseases such as SLE and RA
