    Consumer-Centric Information Systems: A Literature Review and Avenues for Further Research

    While consumer centricity has been extensively discussed as a concept of organizational transformation in the marketing domain, there is little research on its operationalization as a characteristic of information systems and associated antecedents.We review the marketing literature to understand generic organizational objectives of consumer centricity which are then generalized as characteristics of consumer-centric information systems. In a second step, we draw on socio-technical theory to conceptualize antecedents of consumer centricity as capabilities to align social and technical system components.Our research contributes to the body of knowledge by theoretically deriving an operationalization and antecedents of consumer centricity in IS research. This paper lays the foundation for a structured review of IS literature to theorize on component alignment capabilities as antecedents of consumer centricity. It further is the basis for case study research to construct a nomological network for consumer-centric information systems

    Tracking specialized T cell subsets Following Immunization Based on Fluorescent Reporter Protein

    The intestine relies upon T regulatory and effector cells to regulate immune response to multiple antigens. A full understanding of this phenomenon would be significant in the treatment of food intolerance and inflammatory bowel diseases (IBDs). The role of Retinoic Acid (RA) in T-cell migration to the gut is well documented. However, the distribution of tissues where this exposure to RA occurs has not been extensively mapped. In order to determine this, the cre-lox system was used to engineer a RA-responsive reporter gene that expresses the fluorescent protein tdTomato following RA exposure. The tissues were then imaged and analyzed using histo-cytometry to determine distribution of cells with RA exposure. RA exposure in various tissue microenvironments was characterized using flow cytometry, PCR, and confocal microscopy imaging to determine the changes in lymphoid expression of tdTomato during immune activation. It was found that intestinal and lymphoid tissues had greater concentrations of cells with prior RA exposure, particularly the Peyer’s Patch, MLN, and Spleen. The preliminary results of these experiments indicate that immune activation leads to a higher density of tdTomato expressing cells in the intestine and lymphoid tissues, but lower in peripheral organs. These results indicate that immunization causes T-cells to be drawn out of peripheral tissues and into gut-associated lymphoid tissues. It is worth looking into the composition of these T-cells as compared to the base population

    Relationships between correlated spikes, oxygen and LFP in the resting-state primate

    Resting-state functional MRI (rsfMRI) provides a view of human brain organization based on correlation patterns of blood oxygen level dependent (BOLD) signals recorded across the whole brain. The neural basis of resting-state BOLD fluctuations and their correlation remains poorly understood. We simultaneously recorded oxygen level, spikes, and local field potential (LFP) at multiple sites in awake, resting monkeys. Following a spike, the average local oxygen and LFP voltage responses each resemble a task-driven BOLD response, with LFP preceding oxygen by 0.5 s. Between sites, features of the long-range correlation patterns of oxygen, LFP, and spikes are similar to features seen in rsfMRI. Most of the variance shared between sites lies in the infraslow frequency band (0.01-0.1 Hz) and in the infraslow envelope of higher-frequency bands (e.g. gamma LFP). While gamma LFP and infraslow LFP are both strong correlates of local oxygen, infraslow LFP explains significantly more of the variance shared between correlated oxygen signals than any other electrophysiological signal. Together these findings are consistent with a causal relationship between infraslow LFP and long-range oxygen correlations in the resting state

    Extracellular Vesicle Characteristics in β-thalassemia as Potential Biomarkers for Spleen Functional Status and Ineffective Erythropoiesis

    β-thalassemia major (β-TM) is a therapeutically challenging chronic disease in which ineffective erythropoiesis is a main pathophysiological factor. Extracellular vesicles (EVs) are membrane-enclosed vesicles released by cells into biological fluids; they are involved in intercellular communication and in multiple physiological and pathological processes. The chaperone heat-shock protein 70 (HSP70), which is released from cells via EVs, aggravates ineffective erythropoiesis in β-TM. We propose that β-TM EVs may show specific signatures, reflecting disease mechanisms, stages and severity. Our study aims were to define EV profiles in β-TM patients, investigate the influence of hypersplenism and splenectomy on EV features, and explore the association of circulating EVs with ineffective erythropoiesis and iron-overload parameters. We characterized circulating EVs in 35 transfusion-dependent β-thalassemia patients and 35 controls using several techniques. Nanoparticle-tracking analysis revealed increased EV concentration in patients vs. controls (P = 0.0036), with smaller EV counts and sizes in patients with hypersplenism. Flow cytometry analysis showed lower levels of RBC and monocyte EVs in patients vs. controls. RBC-EV levels correlated with patient hematocrit, reflecting degree of anemia. The procoagulant potential of the EVs evaluated by flow cytometry revealed lower levels of endothelial protein C receptor-labeled EVs in patients vs. controls, and increased tissue factor-to-tissue factor pathway inhibitor-labeled EV ratio in splenectomized patients, suggesting a hypercoagulable state. Protein content, evaluated in EV pellets, showed increased levels of HSP70 in patients (P = 0.0018), inversely correlated with transfusion requirement and hemoglobin levels, and positively correlated with reticulocyte, erythropoietin and lactate dehydrogenase levels. This first description of EVs in patients with hypersplenism reveals the spleen’s importance in EV physiology and clearance. Circulating EV-HSP70 levels were associated with markers of ineffective erythropoiesis, hemolysis and hematological disease severity. EV analysis in β-TM—reflecting spleen status, hypercoagulability state and ineffective erythropoiesis—may serve as a biomarker of disease dynamics, supporting both anticipation of the risk of complications and optimizing treatment

    Survival of adults with acute lymphoblastic leukemia in Germany and the United States.

    BACKGROUND: Adulthood acute lymphoblastic leukemia (ALL) is a rare disease. In contrast to childhood ALL, survival for adults with ALL is poor. Recently, new protocols, including use of pediatric protocols in young adults, have improved survival in clinical trials. Here, we examine population level survival in Germany and the United States (US) to gain insight into the extent to which changes in clinical trials have translated into better survival on the population level. METHODS: Data were extracted from the Surveillance, Epidemiology, and End Results database in the US and 11 cancer registries in Germany. Patients age 15-69 diagnosed with ALL were included. Period analysis was used to estimate 5-year relative survival (RS). RESULTS: Overall 5-year RS was estimated at 43.4% for Germany and 35.5% for the US (p = 0.004), with a decrease in survival with increasing age. Survival was higher in Germany than the US for men (43.6% versus 37.7%, p = 0.002) but not for women (42.4% versus 40.3%, p\u3e0.1). Five-year RS estimates increased in Germany and the US between 2002 and 2006 by 11.8 and 7.3 percent units, respectively (p = 0.02 and 0.04, respectively). CONCLUSIONS: Survival for adults with ALL continues to be low compared with that for children, but a substantial increase in 5-year survival estimates was seen from 2002 to 2006 in both Germany and the US. The reasons for the survival differences between both countries require clarification
