    Giardia and Cryptosporidium and public health: the epidemiological scenario from the Italian perspective

    Giardia and Cryptosporidium spp. are protozoa that cause human and animal disease worldwide and often exhibit zoonotic transmission. This review gives ample information concerning the epidemiology of these parasites in Italy, i.e. prevalence data in humans, farm and pet animals, shellfish and aquatic environment. Moreover, it reports genotyping results obtained from different isolates, with particular emphasis on the spread of host-specific and zoonotic species/genotypes of various origin, and on molecular data that make the Italian situation different from that of other countries. Finally, possible explanations are given for the infrequent reports of Giardia and Cryptosporidium spp. outbreaks, despite widespread faecal contamination by these parasites

    Optical Synthesis of Terahertz and Millimeter-Wave Frequencies with Discrete Mode Diode Lasers

    It is shown that optical synthesis of terahertz and millimeter-wave frequencies can be achieved using two-mode and mode-locked discrete mode diode lasers. These edge-emitting devices incorporate a spatially varying refractive index profile which is designed according to the spectral output desired of the laser. We first demonstrate a device which supports two primary modes simultaneously with high spectral purity. In this case sinusoidal modulation of the optical intensity at terahertz frequencies can be obtained. Cross saturation of the material gain in quantum well lasers prevents simultaneous lasing of two modes with spacings in the millimeter-wave region. We show finally that by mode-locking of devices that are designed to support a minimal set of four primary modes, we obtain a sinusoidal modulation of the optical intensity in this frequency region.Comment: 6 page

    Bistability and all-optical memory in dual-mode diode lasers with time-delayed optical feedback

    A proposal for an all-optical memory based on a bistability of single-mode states in a dual-mode diode laser with time-delayed optical feedback is presented. The system is modeled using a multimode extension of the Lang-Kobayashi equations with injected optical pulses. We uncover the bifurcation structure by deriving analytical expressions for the boundaries of the bistable region and demonstrate how the delay time in the external cavity determines an optimal pulse duration for efficient switching of the memory element. We also show the relevant role played by gain saturation and by the dual-mode solutions of the Lang-Kobayashi equations for the existence of the bistable regions. Our results demonstrate that feedback induced bistability can lead to significant performance improvements when compared to memory elements based on the injection locking bistability in dual-mode devices

    Implicit Extended Kalman Filter for Optical Terrain Relative Navigation Using Delayed Measurements

    The exploration of celestial bodies such as the Moon, Mars, or even smaller ones such as comets and asteroids, is the next frontier of space exploration. One of the most interesting and attractive purposes from the scientific point of view in this field, is the capability for a spacecraft to land on such bodies. Monocular cameras are widely adopted to perform this task due to their low cost and system complexity. Nevertheless, image-based algorithms for motion estimation range across different scales of complexities and computational loads. In this paper, a method to perform relative (or local) terrain navigation using frame-to-frame features correspondences and altimeter measurements is presented. The proposed image-based approach relies on the implementation of the implicit extended Kalman filter, which works using nonlinear dynamic models and corrections from measurements that are implicit functions of the state variables. In particular, here, the epipolar constraint, which is a geometric relationship between the feature point position vectors and the camera translation vector, is employed as the implicit measurement fused with altimeter updates. In realistic applications, the image processing routines require a certain amount of time to be executed. For this reason, the presented navigation system entails a fast cycle using altimeter measurements and a slow cycle with image-based updates. Moreover, the intrinsic delay of the feature matching execution is taken into account using a modified extrapolation method

    Canine leishmaniasis in Southern Italy: a role for nitric oxide released from activated macrophages in asymptomatic infection?

    <p>Abstract</p> <p>Background</p> <p>Human and canine leishmaniasis (CanL) by <it>Leishmania infantum </it>is endemic in Italy, with a high percentage of infected asymptomatic animals. However, the immune response mechanisms underlying the clinical presentation of CanL have not been fully investigated. Among leishmanicidal molecules produced by activated macrophages, nitric oxide (NO) produced by an inducible NO synthase seems to play an important protective role, but no conclusive data are available. Therefore, NO released by cultured macrophages from dogs with natural <it>Leishmania </it>infection living in an endemic area for CanL was evaluated.</p> <p>Methods</p> <p>On the basis of one year's clinical and laboratory follow-up, 22 dogs infected by <it>Leishmania infantum </it>were identified and grouped as: asymptomatic dogs (n = 13) and dogs with symptoms of leishmaniasis (n = 9). Each animal was bled twice at 4-month intervals and macrophage and lymphocyte cultures were obtained from peripheral blood mononuclear cells. Supernatants of <it>L. infantum</it>-infected macrophage cultures, with or without addition of autologous lymphocytes, were assayed for NO production by Griess reaction for nitrites.</p> <p>Results</p> <p>In the first months of the infection the levels of NO in supernatants of <it>Leishmania</it>-infected macrophages were higher in symptomatic than in asymptomatic dogs, but they were significantly increased in the latter group eight months after the diagnosis of infection. Furthermore, NO release significantly decreased in the presence of autologous lymphocytes in both groups of animals.</p> <p>Conclusion</p> <p>These results suggest that NO may be involved in the long-term protection of dogs against natural <it>Leishmania </it>infection and in the clinical presentation of canine leishmaniasis in the Mediterranean area.</p

    Point-to-point overlay of a 100Gb/s DP-QPSK channel in LR-PONs for urban and rural areas

    The continuing growth in information demand from fixed and mobile end-users, coupled with the need to deliver this content in an economically viable manner, is driving new innovations in access networks. In particular, it is becoming increasingly important to find new ways to enable the coexistence of heterogeneous services types which may require different signal modulation formats over the same fiber infrastructure. For example, the same physical layer can potentially be used to deliver shared 10Gb/s services to residential customers, dedicated point-to-point (P2P) 100Gb/s services to business customers, and wireless fronthaul, in a highly cost-effective manner. In this converged scenario, the performance of phase modulated signals can be heavily affected by nonlinear crosstalk from co-propagating on-off-keying (OOK) channels. In this paper, the overlay of a 100G P2P dual-polarization quadrature phase-shift keying (DP-QPSK) channel in a long-reach passive optical network (LR-PON) in the presence of co-propagating 10Gb/s OOK neighboring channels is studied for two different PON topologies. The first LR-PON topology is particularly suited for densely populated areas while the second is aimed at rural, sparsely populated areas. The experimental results indicate that with an emulated load of 40 channels the urban architecture can support up to 100km span and 512 users, while the rural architecture can support up to 120km span and 1024 users. Finally, a system model is developed to predict the system performance and system margins for configurations different from the experimental setups and to carry out design optimization that could in principle lead to even more efficient and robust schemes

    Giardia and Cryptosporidium in inflowing water and harvested shellfish in a Lagoon in Southern Italy

    Giardia and Cryptosporidium spp. are important enteric protozoan pathogens for humans and animals, and have been found to contaminate water as well as edible shellfish all over the world. This is the first study to simultaneously investigate the presence of Giardia and Cryptosporidium in inflowing water and harvested shellfish in a geographically closed environment (Varano Lagoon, Southern Italy). Samples of treated wastewater were collected each month - at the outlet from the treatment plant, and downstream at the inlet into the lagoon - from the channels flowing into the Lagoon, together with specimens of Ruditapes decussatus and Mytilus galloprovincialis from shellfish-farms on the same lagoon. Giardia cysts were found by immunofluorescence (IF) microscopy in 16 out of 21 samples of treated wastewater and in 7 out of 21 samples from downstream water channels, and viable cysts were also detected by a beta-giardin RT-PCR. G. duodenalis Assemblages A and B were identified by small ribosomal Subunit (18S-rDNA) and triosephosphate isomerase (tpi)-PCR, followed by sequencing. Cryptosporidium oocysts were found by IF in 5 out of 21 wastewater samples, and in 8 out of 21 samples from water channels. Molecular analysis identified the zoonotic species Cryptosporidium parvum by oocyst wall protein (COWP)-PCR and sequencing. Higher concentrations of Giardia cysts than Cryptosporidium oocysts were registered in almost all wastewater and water samples. IF and molecular testing of shellfish gave negative results for both protozoa. Wastewaters carrying Giardia and Cryptosporidium (oo)cysts are discharged into the Lagoon; however, the shellfish harvested in the same environment were found to be unaffected, thus suggesting that physical, ecological and climatic conditions may prevent contamination of harvested shellfish.
