    Mosquitoes of Urban Oklahoma and Their Potential as Disease Vectors

    The mosquito diversity of Oklahoma was last evaluated in a series of surveys conducted between 2003 and 2006 and before that, the previous survey was in 1965. Prior surveys targeted specific questions including the evaluation of West Nile virus. However, a systematic survey of the cities in Oklahoma was not conducted. In this study, mosquito populations were surveyed approximately bi-weekly at six different cities, including four with military bases, in central and southeastern Oklahoma between May and September 2016. Three different traps were used: CDC Mini Light traps with lights removed and baited with dry ice, CDC Gravid traps baited with Bermuda grass-conditioned water, and BG-SentinelοΏ½ traps baited with BG lure. A total of 11,980 adult female mosquitoes representing 34 species were collected over 834 trapping events. Mosquito communities differed significantly among trap type, with CDC Mini Light traps collecting the most individuals. Mosquito communities also differed significantly among cities sampled. These results show that urban areas in Oklahoma do not possess a homogenous mosquito community, and each city should be evaluated individually (at least within the same climate region). Aedes aegypti, an important disease vector was recorded in Oklahoma for the first time since 1940. Eighty-eight specimens of Ae. aegypti were collected in the cities of Altus, Ardmore, Frederick, and Lawton. CDC Mini Light traps were more attractive to Ae. aegypti than other two trap types. The most abundant mosquito species: Ae. albopictus, Ae. sollicitans, Ae. triseriatus, Cx. pipiens, and Cx. tarsalis were tested for presence of Dirolfilaria immitis, the causative agent for dog heartworm, and other parasitic nematodes. One mosquito pool tested was PCR positive for D. immitis, from a Cx. pipiens pool. Six other nematodes were collected in pools from Ae. albopictus. The D. immitis positive pool was collected from Idabel, OK, while four other positive pools were collected from Ardmore, OK, and one from Midwest City, OK. The information generated from this study provides insight into mosquito populations and potential for nematode transmission in Oklahoma. With increases in human populations living in cities, movement of people and global climate change, the results of this study serves as a baseline and informs mosquito management strategies for southern Oklahoma.Entomology & Plant Pathology (MS

    Mosquito-borne parasites in the Great Plains: Searching for vectors of nematodes and avian malaria parasites

    Vector-borne diseases in the United States have recently increased as a result of the changing nature of vectors, hosts, reservoirs, parasite/pathogens, and the ecological and environmental conditions. While most focus has been on mosquito-borne pathogens affecting humans, little is known regarding parasites of companion animal, livestock and wildlife and their potential mosquito hosts in the United States. This study assessed the prevalence of mature infections of Dirofilaria immitis and avian malaria parasites (Haemosporida) within urban mosquito (Diptera, Culicidae) communities in Oklahoma. 2,620 pools consisting of 12,686 mosquitoes from 13 species collected over two summers were tested for the presence of filarioid and haemosporidian DNA. Dirofilaria immitis-infected mosquitoes were detected only in Aedes albopictus (MIR=0.18-0.22) and Culex pipiens complex (MIR=0.12) collected in cities in central and southern Oklahoma. Two other filarioid nematode species with 91-92% similarity with Onchocerca spp. and Mansonella spp. were also detected. Haemosporidian DNA was detected in 13 mosquito pools (0.9% of pools tested) from seven mosquito species out of 13 species tested. Plasmodium DNA in four species (Cx. coronator, Cx. pipiens complex, Cx. tarsalis, and Psorophora columbiae) had high homology with published sequences of avian Plasmodium species while DNA in four other species (Cx. nigripalpus, Ps. columbiae, Anopheles quadrimaculatus, and An. punctipennis) were closely related to Plasmodium species from deer. One pool of Cx. tarsalis was positive with a 100% sequence identity of Haemoproteus sacharovi. This study provides a baseline concerning the diversity of parasites in different mosquito species present in the southern Great Plains. These studies provide important information for understanding the factors of transmission involving the mosquito community, potential hosts, and different mosquito-borne parasites in this important region involved in livestock management and wildlife conservation.Peer reviewedEntomology and Plant Patholog

    Music therapy for end-of-life care: An updated systematic review.

    BACKGROUND: Music therapy during palliative and end-of-life care is well established and positive benefits for patients have been reported. AIM: Assess the effectiveness of music therapy versus standard care alone or standard care in combination with other therapies for improving psychological, physiological and social outcomes among adult patients in any palliative care setting. DATA SOURCES: In order to update an existing Cochrane systematic review, we searched MEDLINE, CINAHL, EMBASE, PsycINFO, CENTRAL, ClinicalTrials.gov register and Current Controlled Trials register to identify randomised or quasi-randomised controlled trials published between 2009 and April 2015. Nine electronic music therapy journals were searched from 2009 until April 2015, along with reference lists and contact was made with key experts in music therapy. Only studies published in English were eligible for inclusion. Two reviewers independently screened titles, abstracts, assessed relevant studies for eligibility, extracted data and judged risk of bias for included studies. Disagreements were resolved through discussion with a third reviewer. Data were synthesised in Revman using the random effects model. Heterogeneity was assessed using I(2). RESULTS: Three studies were included in the review. Findings suggest that music therapy may be effective for helping to reduce pain in palliative care patients (standard mean deviation = -0.42, 95% confidence interval = -0.68 to -0.17, p = 0.001). CONCLUSION: Available evidence did not support the use of music therapy to improve overall quality of life in palliative care. While this review suggests that music therapy may be effective for reducing pain, this is based on studies with a high risk of bias. Further high-quality research is required

    New records of Aedes aegypti in southern Oklahoma, 2016

    Aedes aegypti is an important subtropical vector species and is predicted to have a limited year-round distribution in the southern United States. Collection of the species has not been officially verified in Oklahoma since 1940. Adult mosquitoes were collected in 42 sites across 7 different cities in Oklahoma using 3 different mosquito traps between May and September 2016. Between July and September 2016, 88 Ae. aegypti adults were collected at 18 different sites in 4 different cities across southern Oklahoma. Centers for Disease Control and Prevention mini light traps baited with CO2 attracted the highest numbers of Ae. aegypti individuals compared to Biogents (BG)-Sentinel traps baited with Biogents (BG)-lure and octenol and Centers for Disease Control and Prevention gravid traps baited with Bermuda grass-infused water. The discovery of Ae. aegypti mosquitoes within urban/exurban areas in Oklahoma is important from an ecological as well as a public health perspective.Peer reviewedEntomology and Plant Patholog

    Aquatic biosurvey of the Lovell River on UNH land

    We assessed the physical, chemical and biological conditions at two sites along the Lovell River on University of New Hampshire (UNH) -owned conservation land. The discharge was 4.4 m3 s-1 at Site 1 and 5.7 m3 s -1 downstream at Site 2. Canopy coverage ranged from 8-25%. Canopy was dominated by Eastern Hemlock (79-84%). Much of the stream was strewn with large boulders and the substrate consisted of rocks of highly variable sizes ( 3-549 cm dia.). Specific conductivity (22.1-23.3 Β΅S), pH (6.4) and temperature (7.9-8.3 Β°C) varied little between sites. Macro-invertebrate bio-indices indicated either excellent water quality with no apparent organic pollution (3.0/10) or good water quality with possible slight organic pollution (4.4/10)

    Literature Triage and Indexing in the Mouse Genome Informatics (MGI) Group

    The Mouse Genome Informatics (MGI; "http://www.informatics.jax.org":http://www.informatics.jax.org) group is comprised of several collaborating projects including the Mouse Genome Database (MGD) Project, the Gene Expression Database (GXD) Project, the Mouse Tumor Biology (MTB) Database Project, and the Gene Ontology (GO) Project. Literature identification and collection is performed cooperatively amongst the groups.

In recent years many institutional libraries have transitioned from a focus largely on print holdings to one of electronic access to journals. This change has necessitated adaptation on the part of the MGI curatorial group. Whereas the majority of journals covered by the group used to be surveyed in paper form, those journals are now surveyed electronically. Approximately 160 journals have been identified as those most relevant to the various database groups. Each curator in the group has the responsibility of scanning several journals for articles relevant to any of the database projects. Articles chosen via this process are marked as to their potential significance for various projects. Each article is catalogued in a Master Bibliography section of the MGI database system and annotated to the database sections for which it has been identified as relevant. A secondary triage process allows curators from each group to scan the chosen articles and mark ones desired for their project if such annotation has been missed on the initial scan.

Once articles have been identified for each database project a variety of processes are implemented to further categorize and index data from those articles. For example, the Alleles and Phenotype section of the MGD database indexes each article marked for MGD and in this indexing process they identify each mouse gene and allele examined in the article. The GXD database indexing process has a different focus. In this case articles are indexed with regard to the stage of development used in the study as well as the assay technique used. In each case the indexing gives an overview of the data held in the article and assists in the more extensive curation performed in the following step of the curation process. Indexing also provides each group with valuable information used to prioritize and streamline the overall curation process.

    Extensive innate immune gene activation accompanies brain aging, increasing vulnerability to cognitive decline and neurodegeneration: a microarray study

    BACKGROUND: This study undertakes a systematic and comprehensive analysis of brain gene expression profiles of immune/inflammation-related genes in aging and Alzheimer’s disease (AD). METHODS: In a well-powered microarray study of young (20 to 59 years), aged (60 to 99 years), and AD (74 to 95 years) cases, gene responses were assessed in the hippocampus, entorhinal cortex, superior frontal gyrus, and post-central gyrus. RESULTS: Several novel concepts emerge. First, immune/inflammation-related genes showed major changes in gene expression over the course of cognitively normal aging, with the extent of gene response far greater in aging than in AD. Of the 759 immune-related probesets interrogated on the microarray, approximately 40% were significantly altered in the SFG, PCG and HC with increasing age, with the majority upregulated (64 to 86%). In contrast, far fewer immune/inflammation genes were significantly changed in the transition to AD (approximately 6% of immune-related probesets), with gene responses primarily restricted to the SFG and HC. Second, relatively few significant changes in immune/inflammation genes were detected in the EC either in aging or AD, although many genes in the EC showed similar trends in responses as in the other brain regions. Third, immune/inflammation genes undergo gender-specific patterns of response in aging and AD, with the most pronounced differences emerging in aging. Finally, there was widespread upregulation of genes reflecting activation of microglia and perivascular macrophages in the aging brain, coupled with a downregulation of select factors (TOLLIP, fractalkine) that when present curtail microglial/macrophage activation. Notably, essentially all pathways of the innate immune system were upregulated in aging, including numerous complement components, genes involved in toll-like receptor signaling and inflammasome signaling, as well as genes coding for immunoglobulin (Fc) receptors and human leukocyte antigens I and II. CONCLUSIONS: Unexpectedly, the extent of innate immune gene upregulation in AD was modest relative to the robust response apparent in the aged brain, consistent with the emerging idea of a critical involvement of inflammation in the earliest stages, perhaps even in the preclinical stage, of AD. Ultimately, our data suggest that an important strategy to maintain cognitive health and resilience involves reducing chronic innate immune activation that should be initiated in late midlife
