Measuring language lateralisation with different language tasks: a systematic review
Language lateralisation refers to the phenomenon in which one hemisphere (typically
the left) shows greater involvement in language functions than the other. Measurement
of laterality is of interest both to researchers investigating the neural organisation of
the language system and to clinicians needing to establish an individual’s hemispheric
dominance for language prior to surgery, as in patients with intractable epilepsy.
Recently, there has been increasing awareness of the possibility that different language
processes may develop hemispheric lateralisation independently, and to varying
degrees. However, it is not always clear whether differences in laterality across language
tasks with fMRI are reflective of meaningful variation in hemispheric lateralisation,
or simply of trivial methodological differences between paradigms. This systematic
review aims to assess different language tasks in terms of the strength, reliability and
robustness of the laterality measurements they yield with fMRI, to look at variability
that is both dependent and independent of aspects of study design, such as the baseline
task, region of interest, and modality of the stimuli. Recommendations are made that
can be used to guide task design; however, this review predominantly highlights that
the current high level of methodological variability in language paradigms prevents
conclusions as to how different language functions may lateralise independently. We
conclude with suggestions for future research using tasks that engage distinct aspects
of language functioning, whilst being closely matched on non-linguistic aspects of task
design (e.g., stimuli, task timings etc); such research could produce more reliable and
conclusive insights into language lateralisation. This systematic review was registered

Defining conservation units with enhanced molecular tools to reveal fine scale structuring among Mediterranean green turtle rookeries

Genetic tools are widely used for this purpose, but the results can be limited by the resolution of the genetic markers in relation to the species and geographic scale. Here, we investigated natal philopatry in green turtles (Chelonia mydas) from four rookeries within close geographic proximity (~200 km) on the Mediterranean island of Cyprus. We genotyped hypervariable mtSTRs, a mtDNA control region sequence (CR) and 13 microsatellite loci to genetically characterise 479 green turtles using markers with different modes of inheritance. We demonstrated matrilineal stock structure for the first time among Mediterranean green turtle rookeries. This result contradicts previous regional assessments and supports a growing body of evidence that green turtles exhibit a more precise level of natal site fidelity than has commonly been recognised. The microsatellites detected weak male philopatry with significant stock structure among three of the six pairwise comparisons. The absence of Atlantic CR haplotypes and mtSTRs amongst these robust sample sizes reaffirms the reproductive isolation of Mediterranean green turtles and supports their status as a subpopulation. A power analysis effectively demonstrated that the mtDNA genetic markers previously employed to evaluate regional stock identity were confounded by an insufficient resolution considering the recent colonisation of this region. Satellite tracking and stable isotope analysis highlight differential recruitment among foraging areas in green turtles

Available on open access from Inter Research via the DOI in this recordIdentifying links between breeding and non-breeding sites in migratory animals is an important step in understanding their ecology. Recognising the relative importance of foraging areas and ascertaining site-specific levels of recruitment can provide fundamental and applied insights. Here, satellite telemetry and the stable isotope ratios (δ13C, δ15N and δ34S) of 230 green turtles Chelonia mydas from a regionally important rookery in northern Cyprus were employed to evaluate the relative importance of 4 foraging areas. A preliminary analysis of stable isotope ratios suggested that a major foraging area had been missed through satellite telemetry as a large proportion of turtles had isotope ratios that did not correspond to sites previously identified. Stable isotope ratios were then employed to select 5 turtles to be fitted with platform terminal transmitters in 2015. All 5 turtles were subsequently tracked to the same location, Lake Bardawil in Egypt. Serially collected tissue samples from 45 females, ranging over 2 to 4 breeding seasons, suggested that foraging site fidelity was very common, with 82% of females exhibiting extremely high temporal consistency in isotope ratios. Quantifying fidelity allowed an evaluation of foraging area-specific contributions to each breeding cohort over the past 2 decades and demonstrated that recruitment was unequal among sites, and dynamic over time, with Egypt now currently the major contributor to the nesting aggregation. This work demonstrates the utility of stable isotope analysis to elucidate the spatial ecology of cryptic taxa and illustrates how more robust baselines can be assembled against which to measure the success of future marine conservation initiatives.P.J.B. is funded by a National Environment

Assessment of a novel, capsid-modified adenovirus with an improved vascular gene transfer profile
<p>Background: Cardiovascular disorders, including coronary artery bypass graft failure and in-stent restenosis remain significant opportunities for the advancement of novel therapeutics that target neointimal hyperplasia, a characteristic of both pathologies. Gene therapy may provide a successful approach to improve the clinical outcome of these conditions, but would benefit from the development of more efficient vectors for vascular gene delivery. The aim of this study was to assess whether a novel genetically engineered Adenovirus could be utilised to produce enhanced levels of vascular gene expression.</p>
<p>Methods: Vascular transduction capacity was assessed in primary human saphenous vein smooth muscle and endothelial cells using vectors expressing the LacZ reporter gene. The therapeutic capacity of the vectors was compared by measuring smooth muscle cell metabolic activity and migration following infection with vectors that over-express the candidate therapeutic gene tissue inhibitor of matrix metalloproteinase-3 (TIMP-3).</p>
<p>Results: Compared to Adenovirus serotype 5 (Ad5), the novel vector Ad5T*F35++ demonstrated improved binding and transduction of human vascular cells. Ad5T*F35++ mediated expression of TIMP-3 reduced smooth muscle cell metabolic activity and migration in vitro. We also demonstrated that in human serum samples pre-existing neutralising antibodies to Ad5T*F35++ were less prevalent than Ad5 neutralising antibodies.</p>
<p>Conclusions: We have developed a novel vector with improved vascular transduction and improved resistance to human serum neutralisation. This may provide a novel vector platform for human vascular gene transfer.</p>
Dental plaque as a biofilm and a microbial community – implications for health and disease
Dental plaque is a structurally- and functionally-organized biofilm. Plaque forms in an ordered way and has a diverse microbial composition that, in health, remains relatively stable over time (microbial homeostasis). The predominant species from diseased sites are different from those found in healthy sites, although the putative pathogens can often be detected in low numbers at normal sites. In dental caries, there is a shift toward community dominance by acidogenic and acid-tolerating species such as mutans streptococci and lactobacilli, although other species with relevant traits may be involved. Strategies to control caries could include inhibition of biofilm development (e.g. prevention of attachment of cariogenic bacteria, manipulation of cell signaling mechanisms, delivery of effective antimicrobials, etc.), or enhancement of the host defenses. Additionally, these more conventional approaches could be augmented by interference with the factors that enable the cariogenic bacteria to escape from the normal homeostatic mechanisms that restrict their growth in plaque and out compete the organisms associated with health. Evidence suggests that regular conditions of low pH in plaque select for mutans streptococci and lactobacilli. Therefore, the suppression of sugar catabolism and acid production by the use of metabolic inhibitors and non-fermentable artificial sweeteners in snacks, or the stimulation of saliva flow, could assist in the maintenance of homeostasis in plaque. Arguments will be presented that an appreciation of ecological principles will enable a more holistic approach to be taken in caries control
In vitro and in vivo evaluation of human adenovirus type 49 as a vector for therapeutic applications
The human adenovirus phylogenetic tree is split across seven species (A–G). Species D adenoviruses offer potential advantages for gene therapy applications, with low rates of pre-exist-ing immunity detected across screened populations. However, many aspects of the basic virology of species D—such as their cellular tropism, receptor usage, and in vivo biodistribution profile— remain unknown. Here, we have characterized human adenovirus type 49 (HAdV-D49)—a rela-tively understudied species D member. We report that HAdV-D49 does not appear to use a single pathway to gain cell entry, but appears able to interact with various surface molecules for entry. As such, HAdV-D49 can transduce a broad range of cell types in vitro, with variable engagement of blood coagulation FX. Interestingly, when comparing in vivo biodistribution to adenovirus type 5, HAdV-D49 vectors show reduced liver targeting, whilst maintaining transduction of lung and spleen. Overall, this presents HAdV-D49 as a robust viral vector platform for ex vivo manipulation of human cells, and for in vivo applications where the therapeutic goal is to target the lung or gain access to immune cells in the spleen, whilst avoiding liver interactions, such as intravascular vaccine applications
Study protocol: evaluation of a parenting and stress management programme: a randomised controlled trial of Triple P discussion groups and stress control
<br>Background: Children displaying psychosocial problems are at an increased risk of negative developmental outcomes. Parenting practices are closely linked with child development and behaviour, and parenting programmes have been recommended in the treatment of child psychosocial problems. However, parental mental health also needs to be addressed when delivering parenting programmes as it is linked with parenting practices, child outcomes, and treatment outcomes of parenting programmes. This paper describes the protocol of a study examining the effects of a combined intervention of a parenting programme and a cognitive behavioural intervention for mental health problems.</br>
<br>Methods: The effects of a combined intervention of Triple P Discussion Groups and Stress Control will be examined using a randomised controlled trial design. Parents with a child aged 3?8?years will be recruited to take part in the study. After obtaining informed consent and pre-intervention measures, participants will be randomly assigned to either an intervention or a waitlist condition. The two primary outcomes for this study are change in dysfunctional/ineffective parenting practices and change in symptoms of depression, anxiety, and stress. Secondary outcomes are child behaviour problems, parenting experiences, parental self-efficacy, family relationships, and positive parental mental health. Demographic information, participant satisfaction with the intervention, and treatment fidelity data will also be collected. Data will be collected at pre-intervention, mid-intervention, post-intervention, and 3-month follow-up.</br>
<br>Discussion: The aim of this paper is to describe the study protocol of a randomised controlled trial evaluating the effects of a combined intervention of Triple P Discussion Groups and Stress Control in comparison to a waitlist condition. This study is important because it will provide evidence about the effects of this combined intervention for parents with 3?8?year old children. The results of the study could be used to inform policy about parenting support and support for parents with mental health problems. Does the carer support needs assessment tool cover the established support needs of carers of patients with chronic obstructive pulmonary disease? A systematic literature search and narrative review
Background: Informal carers play a key supportive role for patients with chronic obstructive pulmonary disease. However, caring can have a considerable impact on health and wellbeing. Carers may have unidentified support needs that could be a target for intervention. Literature on the support needs of informal carers has not been fully synthesised, and our knowledge of the comprehensiveness of the Carer Support Needs Assessment Tool for these individuals is limited. Aim: To explore whether the Carer Support Needs Assessment Tool covers the support needs of carers of patients with chronic obstructive pulmonary disease identified in published literature. Design: English language studies were identified against predetermined inclusion/exclusion criteria through database searching. Further studies were identified through searching reference lists and citations of included papers. Papers were critically appraised and data extracted and synthesised by two reviewers. Identified needs were mapped to Carer Support Needs Assessment Tool questions. Data sources: MEDLINE, CINAHL, EMBASE, CDSR, ASSIA, PsycINFO and Scopus databases (Jan 1997–Dec 2017). Results: Twenty-four studies were included. Results suggest that carers have support needs in a range of domains including physical, social, psychological and spiritual. Many of these needs are unmet. Particular areas of concern relate to prolonged social isolation, accessing services, emotional support and information needs. Findings also suggest amendment of the Carer Support Needs Assessment Tool may be required relating to difficulties within relationship management. Conclusion: Evidence suggests that carers of patients with chronic obstructive pulmonary disease would benefit from identification and response to their support needs by healthcare professionals but to enable this, the Carer Support Needs Assessment Tool requires an additional question. Future planned work will explore this with carers of patients with chronic obstructive pulmonary disease
Predicting predator–prey interactions in terrestrial endotherms using random forest
Species interactions play a fundamental role in ecosystems. However, few ecological communities have complete data describing such interactions, which is an obstacle to understanding how ecosystems function and respond to perturbations. Because it is often impractical to collect empirical data for all interactions in a community, various methods have been developed to infer interactions. Machine learning is increasingly being used for making interaction predictions, with random forest being one of the most frequently used of these methods. However, performance of random forest in inferring predator-prey interactions in terrestrial vertebrates and its sensitivity to training data quality remain untested. We examined predator–prey interactions in two diverse, primarily terrestrial vertebrate classes: birds and mammals. Combining data from a global interaction dataset and a specific community (Simpson Desert, Australia), we tested how well random forest predicted predator–prey interactions for mammals and birds using species' ecomorphological and phylogenetic traits. We also tested how variation in training data quality – manipulated by removing records and switching interaction records to non-interactions – affected model performance. We found that random forest could predict predator–prey interactions for birds and mammals using ecomorphological or phylogenetic traits, correctly predicting up to 88 and 67% of interactions and non-interactions in the global and community-specific datasets, respectively. These predictions were accurate even when there were no records in the training data for focal species. In contrast, false non-interactions for focal predators in training data strongly degraded model performance. Our results demonstrate that random forest can identify predator–prey interactions for birds and mammals that have few or no interaction records. Furthermore, our study provides guidance on how to prepare training data to optimise machine learning classifiers for predicting species interactions, which could help ecologists 1) address knowledge gaps and explore network-related questions in data-poor situations, and 2) predict interactions for range-expanding species
