    Ce mémoire de création littéraire se divise en deux parties. La première propose une série de chansons intitulée Chansons Fleuve. Ce sont huit pièces créées autour de l'idée de l'intime, dont j'ai composé les paroles et la musique1. Elles ont été enregistrées sur un support audio qui accompagne la copie papier de ce mémoire. La seconde partie consiste en une analyse des rapports qu'entretiennent le texte et la musique d'"Arbre à fruits, arbre à fruits" de Marie-Jo Thério, avec l'intime. À travers les paroles de la pièce, les marques de l'intime sont présentes comme contenu (l'essence de l'intime, les thématiques) et comme forme (les relations, les situations, le temps, l'espace). Dans l'enregistrement de la voix et l'interprétation, sont véhiculées plusieurs émotions qui installent un climat d'intimité, appuient le texte et contribuent à rapprocher le public de l'artiste

    MMP-2 expression in uveal melanoma : differential activation status dictated by the cellular environment

    Purpose: MMPs are recognized to play a major role in tumor progression and metastasis of many forms of cancers. The purpose of this study was to compare the expression and activity of MMP-2 in uveal melanoma cell lines grown either in vitro on plastic culture plates or in vivo as tumors produced in chick embryos. Methods: The chick chorioallantoic membrane (CAM) model was used to evaluate the tumorigenic potential of uveal melanoma cell lines derived either from the primary uveal melanoma tumor isolated from three different patients (cell lines SP6.5, SP8.0, and TP31) or from a metastatic lesion derived from the liver of a patient diagnosed with uveal melanoma (cell line H79). The presence of MMP-2 in the vicinity of the tumor cells was determined by immunofluorescence analyses. Gelatin zymography was used for the detection of latent and activated forms of MMP-2 in uveal melanoma cell lines when grown in vitro on plastic, or in the solid tumors these cell lines produced in vivo on the CAM of the chick embryo. The gelatinase activity was quantified by densitometric analyses and the active/(active+pro-form) ratio was calculated as the MMP-2 activation ratio. Western blot analyses were performed to confirm the zymographic profile. Results: Only the inactive form of MMP-2 was expressed and secreted in vitro by all uveal melanoma cell lines, higher levels being found for the liver-derived H79 cell line whereas SP8.0 only expressed MMP-2 to a very low level. On the other hand, all solid tumors produced in the CAM from these cell lines expressed and secreted, although to varying levels (SP6.5 and SP8.0, TP31 and H79), primarily the active form of MMP-2. Gelatinolytic activities of active MMP-2 were significantly higher in uveal melanoma tissues than in the non-neoplastic CAM, as revealed by the measurement of the activation ratio. The immunolocalization of MMP-2 revealed that all cell lines were MMP-2-positive although a reduced and more diffuse staining was observed for H79 and SP6.5 than in SP8.0 and TP31 cells. Conclusions: These results suggest the activation of proMMP-2 as an important event in the process of uveal melanoma progression. An elevated active to inactive MMP-2 ratio in the tumor environment of uveal melanoma suggests that a potential MMP-2 activity could be related to the progression of this type of cancer

    Preparation for an Half-Ironmantm Triathlon amongst Amateur Athlete: Finishing rate and physiological adaptation.

    International Journal of Exercise Science 13(6): 766-777, 2020. Long distance triathlon has gained in popularity amongst the general population. Coaches establish training programs based upon their knowledge, personal experience and on current training principles. The goal was to observe the effect of a triathlon training program for a half Ironman event in neophyte amateur athletes. A specific triathlon training program was followed from February to June 2016 by a group preparing for their first half ironman. Out of the 32 participants (19 Males and 13 Females; mean age of 39 ± 9.9 years old; body weight of 72.7 ± 13.4 kg and a height of 171.5 ± 10.2 cm), only one did not complete the event. A mean training volume of 410 ± 201 min per week led to a mean finishing time of 6 hours 28 minutes. The training program significantly increased the maximal oxygen consumption (45.9 ± 8.2 to 48.6 ± 7.5 ml/kg/min, p =0.002) and the maximal power output (293.1 ± 63.7 to 307.8 ± 58.7 W, p \u3c 0.001). The absolute oxygen consumption and power output at both ventilatory thresholds also significantly increased (VT1: 2.2 ± 0.4 to 2.5 ± 0.5 L, p = 0.001; 157.8 ± 41.8 to 176.7 ± 41.1 W p = 0.009 and VT2: 2.9 ± 0.4 to 3.0 ± 0.4 L, p = 0.017; 229.3 ± 62.0 to 244.8± 55.2 W, p = 0.022 ). A significant diminution of waist circumference was observed (83.2 ± 10.0 to 81.8 ± 9.5 cm, p = 0.032) with no significant changes in body weight. Thus, a 24-week specific training program appears to be safe and efficient for amateur athletes aiming to finish their first half- Ironman event

    A MAP6-Related Protein Is Present in Protozoa and Is Involved in Flagellum Motility

    In vertebrates the microtubule-associated proteins MAP6 and MAP6d1 stabilize cold-resistant microtubules. Cilia and flagella have cold-stable microtubules but MAP6 proteins have not been identified in these organelles. Here, we describe TbSAXO as the first MAP6-related protein to be identified in a protozoan, Trypanosoma brucei. Using a heterologous expression system, we show that TbSAXO is a microtubule stabilizing protein. Furthermore we identify the domains of the protein responsible for microtubule binding and stabilizing and show that they share homologies with the microtubule-stabilizing Mn domains of the MAP6 proteins. We demonstrate, in the flagellated parasite, that TbSAXO is an axonemal protein that plays a role in flagellum motility. Lastly we provide evidence that TbSAXO belongs to a group of MAP6-related proteins (SAXO proteins) present only in ciliated or flagellated organisms ranging from protozoa to mammals. We discuss the potential roles of the SAXO proteins in cilia and flagella function
