Characteristics of the safety climate in teams with world-class safety performance on construction projects in South Africa
Accidents and incidents in the construction environment are not reduced or eliminated effectively, despite numerous efforts made to improve health and safety in the industry. An extensive field of research has been conducted on how teams in the construction environment interact to deliver a project successfully in terms of cost, quality and time. Previous research exists on how team dynamics interface with safety, but is found to be of different focus or markets than this study. This study aims to determine the characteristics of the safety climate that exists in construction teams in South Africa in terms of world-class safety performance, when compared to teams with poor safety performance. An adaptation of the modified safety pyramid (Zohar, 2010) provides a useful conceptual model to investigate the link between safety performance and safety climate related to construction teams. By combining observations from literature and predominantly adapting the Safety Climate Questionnaire (SCQ), a number of directly measurable factors are identified that can be correlated with safety performance and utilised in this investigation to identify the unique characteristics of safety climate in construction teams with world-class safety performance. The characteristics of the safety climate in construction teams were divided into eight groups, namely work pressure, incident investigation, adequacy of procedures, communication and training, relationships, personal protective equipment, spares, and safety. The framework developed in previous studies is used for assessing the responses of safety users to that of safety performance observations. It is found that relationships within an organisation are a major contributory factor. in safety performance, but also the way in which the safety climate is enforced in an inclusive management style through proper procedure, training and communication. This also supports the results of Cohen (1977). 
Feasibility of a Research Protocol to Investigate the Effect of the TherapressureTM Program Using Salivary Cortisol
Background: There is an absence of high quality research to support the use of the Therapressure ProgramTM. This pilot study aimed at developing appropriate research protocols to investigate the effectiveness of the Therapressure ProgramTM on the stress response in children with sensory overresponsivity.
Method: A one-group pre-test/post-test repeated measures design was conducted using a convenience sample. Six children (6-8 years of age) with sensory overresponsivity received 14 consecutive days of the Therapressure ProgramTM by their parents at home. Parents concurrently collected salivary cortisol samples from their children.
Results: Children with sensory overresponsivity displayed both hyper- and hypo-cortisolism at baseline. All of the children’s cortisol levels shifted toward a normative range after intervention. Aspects have been identified related to the data collection protocol.
Conclusion: When testing children with sensory overresponsivity who are constantly activating their stress response system, we raise awareness of the need to check for both hyper- and hypo-cortisolism during statistical analysis. Preliminary pilot data may also show modulation of sympathetic arousal following the intervention. Further research is warranted and recommendations are made related to data collection protocols
A molecular map of mesenchymal tumors
BACKGROUND: Bone and soft tissue tumors represent a diverse group of neoplasms thought to derive from cells of the mesenchyme or neural crest. Histological diagnosis is challenging due to the poor or heterogenous differentiation of many tumors, resulting in uncertainty over prognosis and appropriate therapy. RESULTS: We have undertaken a broad and comprehensive study of the gene expression profile of 96 tumors with representatives of all mesenchymal tissues, including several problem diagnostic groups. Using machine learning methods adapted to this problem we identify molecular fingerprints for most tumors, which are pathognomonic (decisive) and biologically revealing. CONCLUSION: We demonstrate the utility of gene expression profiles and machine learning for a complex clinical problem, and identify putative origins for certain mesenchymal tumors

Targeting of Fumarate Hydratase from Mycobacterium tuberculosis Using Allosteric Inhibitors with a Dimeric-Binding Mode.
With the growing worldwide prevalence of antibiotic-resistant strains of tuberculosis (TB), new targets are urgently required for the development of treatments with novel modes of action. Fumarate hydratase (fumarase), a vulnerable component of the citric acid cycle in Mycobacterium tuberculosis (Mtb), is a metabolic target that could satisfy this unmet demand. A key challenge in the targeting of Mtb fumarase is its similarity to the human homolog, which shares an identical active site. A potential solution to this selectivity problem was previously found in a high-throughput screening hit that binds in a nonconserved allosteric site. In this work, a structure-activity relationship study was carried out with the determination of further structural biology on the lead series, affording derivatives with sub-micromolar inhibition. Further, the screening of this series against Mtb in vitro identified compounds with potent minimum inhibitory concentrations
Biblical Theology of Life in the Old Testament
Life is a primary theme in Scripture, expressed in the rich diversity of the various books, corpora and genres of Scripture. Much has been published on what Scripture teaches about life and death. To date, however, no comprehensive biblical theology in which the concept of life is traced throughout the different books and corpora of the Old and New Testament has been published. It is this lacuna that this book aims to fill, assuming that such an approach can provide a valuable contribution to the theological discourse on life and related concepts. The primary aim of this book is to give an indication of the different nuances of the concept of life in the various books and corpora of the Old and New Testament by providing the reader with a book-by-book overview of the concept of life in Scripture. The secondary aim is to give an indication of the overall use and function of the concept of life in the Old Testament, the New Testament, and Scripture as a whole. The latter is provided by using the findings of the book-by-book overview of the concept of life in Scripture to draw the lines together

Development of Inhibitors against Mycobacterium abscessus tRNA (m1G37) Methyltransferase (TrmD) Using Fragment-Based Approaches.
Mycobacterium abscessus (Mab) is a rapidly growing species of multidrug-resistant nontuberculous mycobacteria that has emerged as a growing threat to individuals with cystic fibrosis and other pre-existing chronic lung diseases. Mab pulmonary infections are difficult, or sometimes impossible, to treat and result in accelerated lung function decline and premature death. There is therefore an urgent need to develop novel antibiotics with improved efficacy. tRNA (m1G37) methyltransferase (TrmD) is a promising target for novel antibiotics. It is essential in Mab and other mycobacteria, improving reading frame maintenance on the ribosome to prevent frameshift errors. In this work, a fragment-based approach was employed with the merging of two fragments bound to the active site, followed by structure-guided elaboration to design potent nanomolar inhibitors against Mab TrmD. Several of these compounds exhibit promising activity against mycobacterial species, including Mycobacterium tuberculosis and Mycobacterium leprae in addition to Mab, supporting the use of TrmD as a target for the development of antimycobacterial compounds
KSHV-Initiated Notch Activation Leads to Membrane-Type-1 Matrix Metalloproteinase-Dependent Lymphatic Endothelial-to-Mesenchymal Transition
SummaryKaposi sarcoma (KS), an angioproliferative disease associated with Kaposi sarcoma herpesvirus (KSHV) infection, harbors a diversity of cell types ranging from endothelial to mesenchymal cells of unclear origin. We developed a three-dimensional cell model for KSHV infection and used it to demonstrate that KSHV induces transcriptional reprogramming of lymphatic endothelial cells to mesenchymal cells via endothelial-to-mesenchymal transition (EndMT). KSHV-induced EndMT was initiated by the viral proteins vFLIP and vGPCR through Notch pathway activation, leading to gain of membrane-type-1 matrix metalloproteinase (MT1-MMP)-dependent invasive properties and concomitant changes in viral gene expression. Mesenchymal markers and MT1-MMP were found codistributed with a KSHV marker in the same cells from primary KS biopsies. Our data explain the heterogeneity of cell types within KS lesions and suggest that KSHV-induced EndMT may contribute to KS development by giving rise to infected, invasive cells while providing the virus a permissive cellular microenvironment for efficient spread
