    Variables determining the commercial maturity of castilla blackberry (Rubus Glaucus Benth)

La mora de Castilla (Rubus glaucus Benth), considerada un fruto no climat?rico altamente perecedero, evidencia en su recolecci?n heterogeneidad en forma, tama?o, color, peso y composici?n, que genera altas p?rdidas durante las etapas de cosecha, poscosecha y comercializaci?n, lo que, en consecuencia, afecta vol?menes en el mercado y produce p?rdidas econ?micas. Por lo anterior, el objetivo de este estudio fue establecer el grado de madurez de cosecha adecuado para la comercializaci?n de la mora proveniente del ca??n del Combeima, para lo cual se recurri? a la evaluaci?n de par?metros f?sicos, qu?micos y fisiol?gicos en producto con grado de madurez 4, 5 y 6 seg?n la NTC 4106. Los resultados obtenidos evidencian diferencias significativas en el estado de madurez 4 respecto del peso fresco que se reduce en promedio 12 %, el m?nimo contenido de s?lidos solubles (6.93 ? 0.12 ?Bx) que indicen en el ?ndice de madurez y la superior intensidad respiratoria durante su almacenamiento refrigerado, aspectos que reducen la calidad y vida ?til de este fruto. Se concluy? que el estado de madurez recomendable para la cosecha y comercializaci?n del producto evaluado es el 5, teniendo en cuenta que en este se alcanza la adecuada acumulaci?n de ?cidos, s?lidos solubles, tama?o y forma caracter?stica del fruto, sin diferencias significativas en peso al compararse con la madurez comercial actual (GM 6).ABSTRACT. The maturity stage is considered a factor decisive in the marketing of horticultural products, mainly because its direct related with the quality and postharvest shelf life. Castilla blackberry (Rubus glaucus Benth) is considered a non-climateric highly perishable fruit, presented in its harvest heterogeneity, in shape, size, color, weight and composition, generating high losses during the harvest, post-harvest and marketing that in consequence affect the market stock and economic losses. Therefore, the aim of this study was establish the proper harvest maturity for marketing of blackberry from Canyon Combeima, which was assement of physical, chemical and physiological with maturity level 4, 5 and 6 according to standard Icontec 4106. The results showed significant differences in maturity stage 4 respect to fresh weight was reduced 12 %, minimum soluble solids content (6.93 ? 0.12 ?Bx) influencing maturity index and higher respiratory rate during refrigerated storage, both of which reduce the quality and shelf life of this fruit. It was concluded that the state of maturity harvest and marketing recommended of the product evaluated is five, considering that in that state it reached adequate accumulation of acids, soluble solids, size and characteristic form of the fruit, without significant differences in weight when compared with the actual commercial maturity (GM 6)

    Numerical dynamic analysis of stiffened plates under blast loading

    Using the general purpose finite element package Abaqus, an investigation has been carried out to examine the dynamic response of steel stiffened plates subjected to uniform blast loading. The main objective of this study is to determine the dynamic response of the stiffened plates considering the effect of stiffener configurations. Several parameters, such as boundary conditions, mesh dependency and strain rate, have been considered in this study. Special emphasis is focused on the evaluation of midpoint displacements and energy of models. The modeling techniques were described in details. The numerical results provide better insight into the effect of stiffener configurations on the nonlinear dynamic response of the stiffened plates subjected to uniform blast loading

    A characteristics framework for Semantic Information Systems Standards

    Semantic Information Systems (IS) Standards play a critical role in the development of the networked economy. While their importance is undoubted by all stakeholders—such as businesses, policy makers, researchers, developers—the current state of research leaves a number of questions unaddressed. Terminological confusion exists around the notions of “business semantics”, “business-to-business interoperability”, and “interoperability standards” amongst others. And, moreover, a comprehensive understanding about the characteristics of Semantic IS Standards is missing. The paper addresses this gap in literature by developing a characteristics framework for Semantic IS Standards. Two case studies are used to check the applicability of the framework in a “real-life” context. The framework lays the foundation for future research in an important field of the IS discipline and supports practitioners in their efforts to analyze, compare, and evaluate Semantic IS Standard

    Efficacy and safety of the anti-IL-12/23 p40 monoclonal antibody, ustekinumab, in patients with active psoriatic arthritis despite conventional non-biological and biological anti-tumour necrosis factor therapy: 6-month and 1-year results of the phase 3, multicentre, double-blind, placebo-controlled, randomised PSUMMIT 2 trial

    Objective: Assess ustekinumab efficacy (week 24/week 52) and safety (week 16/week 24/week 60) in patients with active psoriatic arthritis (PsA) despite treatment with conventional and/or biological anti-tumour necrosis factor (TNF) agents. Methods: In this phase 3, multicentre, placebo-controlled trial, 312 adults with active PsA were randomised (stratified by site, weight (≤100 kg/>100 kg), methotrexate use) to ustekinumab 45 mg or 90 mg at week 0, week 4, q12 weeks or placebo at week 0, week 4, week 16 and crossover to ustekinumab 45 mg at week 24, week 28 and week 40. At week 16, patients with <5% improvement in tender/swollen joint counts entered blinded early escape (placebo→45 mg, 45 mg→90 mg, 90 mg→90 mg). The primary endpoint was ≥20% improvement in American College of Rheumatology (ACR20) criteria at week 24. Secondary endpoints included week 24 Health Assessment Questionnaire-Disability Index (HAQ-DI) improvement, ACR50, ACR70 and ≥75% improvement in Psoriasis Area and Severity Index (PASI75). Efficacy was assessed in all patients, anti-TNF-naïve (n=132) patients and anti-TNF-experienced (n=180) patients. Results: More ustekinumab-treated (43.8% combined) than placebo-treated (20.2%) patients achieved ACR20 at week 24 (p<0.001). Significant treatment differences were observed for week 24 HAQ-DI improvement (p<0.001), ACR50 (p≤0.05) and PASI75 (p<0.001); all benefits were sustained through week 52. Among patients previously treated with ≥1 TNF inhibitor, sustained ustekinumab efficacy was also observed (week 24 combined vs placebo: ACR20 35.6% vs 14.5%, PASI75 47.1% vs 2.0%, median HAQ-DI change −0.13 vs 0.0; week 52 ustekinumab-treated: ACR20 38.9%, PASI75 43.4%, median HAQ-DI change −0.13). No unexpected adverse events were observed through week 60. Conclusions: The interleukin-12/23 inhibitor ustekinumab (45/90 mg q12 weeks) yielded significant and sustained improvements in PsA signs/symptoms in a diverse population of patients with active PsA, including anti-TNF-experienced PsA patients

    Real world study of sacituzumab govitecan in metastatic triple-negative breast cancer in the United Kingdom

    \ua9 The Author(s) 2024.Background: Treatment options for pre-treated patients with metastatic triple-negative breast cancer (mTNBC) remain limited. This is the first study to assess the real-world safety and efficacy of sacituzumab govitecan (SG) in the UK. Methods: Data was retrospectively collected from 16 tertiary UK cancer centres. Pts had a diagnosis of mTNBC, received at least two prior lines of treatment (with at least one being in the metastatic setting) and received at least one dose of SG. Results: 132 pts were included. Median age was 56 years (28–91). All patients were ECOG performance status (PS) 0-3 (PS0; 39, PS1; 76, PS2; 16, PS3;1). 75% (99/132) of pts had visceral metastases including 18% (24/132) of pts with CNS disease. Median PFS (mPFS) was 5.2 months (95% CI 4.5–6.6) with a median OS (mOS) of 8.7 months (95% CI 6.8-NA). The most common adverse events (AEs) were fatigue (all grade; 82%, G3/4; 14%), neutropenia (all grade; 55%, G3/4; 29%), diarrhoea (all grade; 58%, G3/4, 15%), and nausea (all grade; 38%, G3/4; 3%). SG dose reduction was required in 54% of pts. Conclusion: This study supports significant anti-tumour activity in heavily pre-treated pts with mTNBC. Toxicity data aligns with clinical trial experience

    Real world, multicentre patterns of treatment and survival in metastatic renal cell carcinoma with the UK Renal Oncology Collaborative (UK ROC): Is it time to look favourably on first-line immunotherapy containing combinations in all IMDC groups?

Cancer Medicine published by John Wiley & Sons Ltd.Introduction: Clinical trials show improved progression-free survival (PFS) and overall survival (OS) in first-line metastatic renal cell carcinoma (mRCC) patients with immunotherapy containing systemic anti-cancer therapies (SACT). However, in the favourable international metastatic renal cell cancer database consortium (IMDC) group there is no trial evidence for OS benefit despite clear PFS improvement when comparing anti-VEGF tyrosine kinase inhibitor (TKI) monotherapy and (immunotherapy and TKI) IO/TKI combinations. Objective: To assess the impact of first-line SACT choice on the clinical outcomes of PFS and OS in mRCC. To evaluate this impact of initial SACT for allcomers and the favourable IMDC group. Methods: A multicentre retrospective review of patients who started SACT for mRCC (01/01/2018–30/06/2021) at 17 UK NHS trusts. Patient demographics and IMDC group were analysed. Survival data were compared using Kaplan–Meier curves, and the statistical significance of differences in outcome between the groups was assessed with the log-rank test. Univariable and multivariable Cox proportional hazard modelling estimate the hazard ratios (HRs) for survival outcomes associated with IMDC and treatment subtype. Results: One thousand three hundred and nineteen patients were identified with a median age of 64. 294 (22.3%), 695 (52.7%) and 321 (24.3%) were IMDC group favourable, intermediate and poor, respectively. 311 (23.6%), 197 (14.9%) and 778 (59%) patients received checkpoint inhibitor and anti-CTLA4 monoclonal antibody (IO/IO), IO/TKI and TKI first-line SACT across all IMDC groups. Significant PFS improvement favouring IO/TKI versus TKI was demonstrated in allcomers HR = 0.61. In the favourable risk group, Log rank testing demonstrated a significant benefit for IO/TKI over TKI for PFS (HR = 0.60, 95% CI [0.39, 0.91]) and OS (HR = 0.42, 95% CI [0.18, 0.99]). Conclusion: In this real-world evidence cohort, we have shown OS and PFS benefit with IO/TKI versus TKI in the favourable IMDC risk group. This has not been previously reported from trial outcomes and would support use of front-line IO/TKI in mRCC favourable risk patients

    Do semantic standards lack quality? : a survey among 34 semantic standards

    The adoption of standards to improve interoperability in the automotive, aerospace, shipbuilding and other sectors could save billions. While interoperability standards have been created for a number of industries, problems persist, suggesting a lack of quality of the standards themselves. The issue of semantic standard quality is not often addressed. In this research we take a closer look at the quality of semantics standards, development processes, and survey the current state of the quality of semantic standards by means of a questionnaire that was sent to standards developers. This survey looked at 34 semantic standards, and it shows that the quality of semantic standards for inter-organizational interoperability can be improved. Improved standards may advance interoperability in networked business. Improvement of semantic standards requires transparency of their quality. Although many semantic standard development organisations already have quality assurance in place, this research shows that they could benefit from a quality measuring instrument

    Evidence of Alternative Splicing as a Regulatory Mechanism for Kissr2 in Pejerrey Fish

    Kisspeptin receptors are G-Protein-Coupled Receptors that regulate GnRH synthesis and release in vertebrates. Here, we report the gene structure of two kisspeptin receptors (kissr2 and kissr3) in pejerrey fish. Genomic analysis exposed a gene structure with 5 exons and 4 introns for kissr2 and 6 exons and 5 introns for kissr3. Two alternative variants for both genes, named kissr2_v1 and _v2, and kissr3_v1 and v2, were revealed by gene expression analyses of several tissues. For both receptors, these variants were originated by alternative splicing retaining intron 3 and intron 4 for kissr2_v2 and kissr3_v2, respectively. In the case of kissr2, the intron retention introduced two stop codons leading to a putatively truncated protein whereas for kissr3, the intron retention produced a reading shift leading to a stop codon in exon 5. Modeling and structural analysis of Kissr2 and Kissr3 spliced variants revealed that truncation of the proteins may lead to non-functional proteins, as the structural elements missing are critical for receptor function. To understand the functional significance of splicing variants, the expression pattern for kissr2 was characterized on fish subjected to different diets. Fasting induced an up-regulation of kissr2_v1 in the hypothalamus, a brain region implicated in control of reproduction and food intake, with no expression of kissr2_v2. On the other hand, fasting did not elicit differential expression in testes and habenula. These results suggest that alternative splicing may play a role in regulating Kissr2 function in pejerrey

    Electromagnetic Correlates of Musical Expertise in Processing of Tone Patterns

    Using magnetoencephalography (MEG), we investigated the influence of long term musical training on the processing of partly imagined tone patterns (imagery condition) compared to the same perceived patterns (perceptual condition). The magnetic counterpart of the mismatch negativity (MMNm) was recorded and compared between musicians and non-musicians in order to assess the effect of musical training on the detection of deviants to tone patterns. The results indicated a clear MMNm in the perceptual condition as well as in a simple pitch oddball (control) condition in both groups. However, there was no significant mismatch response in either group in the imagery condition despite above chance behavioral performance in the task of detecting deviant tones. The latency and the laterality of the MMNm in the perceptual condition differed significantly between groups, with an earlier MMNm in musicians, especially in the left hemisphere. In contrast the MMNm amplitudes did not differ significantly between groups. The behavioral results revealed a clear effect of long-term musical training in both experimental conditions. The obtained results represent new evidence that the processing of tone patterns is faster and more strongly lateralized in musically trained subjects, which is consistent with other findings in different paradigms of enhanced auditory neural system functioning due to long-term musical training
