    Comparison of α-amylase isoforms from the midgut of Cerambyx cerdo L. (Coleoptera: Cerambycidae) larvae developed in the wild and on an artificial diet

    α-Amylase isoforms of Cerambyx cerdo larvae from the wild (ML and SL) and reared in the laboratory (ADL) were compared. Three amylase isoforms were presented in the SL and ML extracts while two isoforms were presented in the ADL according to zymogram after isoelectric focusing (IEF). All C. cerdo amylase isoforms were acidic proteins (pI < 3.5). Seven amylase isoforms (ACC 1-7) from the midgut of C. cerdo larvae were found in the ML midgut extract, six in the SL extract, and four in the ADL extract according to native PAGE zymogram. The ADL amylase had the highest activity. All crude midgut extracts of C. cerdo larvae were fractionated on a Superose 12 HR column. The molecular mass of the ACC was estimated to be 34 kDa.Upoređene su izoforme α-amilaze larvi Cerambyx cerdo sakupljenih iz prirode (ML i SL) i gajenih na veštačkoj podlozi u laboratoriji (ADL). Zimogramskom detekcijom posle IEF-a po tri izoforme su detektovane u ML i SL ekstraktima, a u ADL dve izoforme. Sve amilazne izoforme iz C. cerdo su bile kisele (pI < 3.5). Zimogramskom detekcijom posle nativne elektroforeze sedam izoformi je detektovano u ML ekstraktu, šest u SL ekstraktu i četiri u ADL ekstraktu. Najveća amilazna aktivnost je detektovana u ADL ekstraktu. Svi ekstrakti srednjih creva larvi C. cerdo su frakcionisani na koloni Superose 12 HR.     Characterization of trypsin-like enzymes from the midgut of Morimus funereus (Coleoptera: Cerambycidae) larvae

    The pH along the midgut of M. funereus larvae had different values, being acidic in the anterior section and basic in the middle and posterior sections. Elastase- and chymotrypsin-like activities were highest in the middle, low in the anterior, and negligible in the posterior section of the midgut. Trypsin-like activities were detected along the whole midgut, with more than 90% of activity in the anterior section. The level of elastase- and chymotrypsin-like activity was very low compared to trypsin-like activity. In the anterior section of the midgut, two isoforms of trypsin-like enzymes were found, both being basic and almost completely inhibited by benzamidine.pH vrednost duž srednjeg creva larvi M. funereus se razlikuje, kisela je u regionu prednjeg dela, dok je u srednjem i zadnjem delu bazna. Elastazama i himotripsinima slična aktivnost je najveća u srednjem delu srednjeg creva dok je u prednjem delu detektovana mala vrednost, a u zadnjem delu zanemarljiva. Tripsinima slična aktivnost je detektovana duž celog srednjeg creva, s tim da se više od 90 % aktivnosti detektuje u prednjem delu srednjeg creva. Zastupljenost elastazama i himotripsinima sličnih endopeptidaza je zanemarljivo mala u poređenju sa zastupljenošću tripsinima sličnih enzima. U prednjem delu srednjeg creva nalaze se dve izoforme tripsinima sličnih enzima, sa baznim pI vrednostima, koje su skoro u potpunosti inhibirane benzamidinom.Projekat ministarstva br. 142026

    Creating a new academic governance in Poland : assumptions to Law HEI 2.0 (Ustawa 2.0)

    W Polsce trwają zaawansowane prace nad przebudową systemu nauki i szkolnictwa wyższego. Powszechne jest przekonanie środowiska akademickiego, ale również opinii społecznej, że istniejące rozwiązania nie wykorzystują w pełni możliwości kadry naukowo-dydaktycznej, studentów i potencjału polskich uczelni. Powszechnie znane są miary słabości, związane chociażby z pozycją uczelni polskich w rankingach międzynarodowych [Wróblewski 2013]. Również efekty edukacyjne, umasowienie kształcenia i niekorzystne zmiany w etosie akademickim napawają niepokojem [Sułkowski 2016]. Naprawy wymaga zatem zarówno poziom ładu systemowego szkolnictwa i nauki (governance), jak i zarządzanie uczelniami wyższymi w Polsce (management) [Antonowicz, Brdulak, Hulicka i in. 2016]. Strategicznym elementem tej zmiany ma być przyjęcie nowej ustawy regulującej system nauki i szkolnictwa wyższego. Na początku 2017 roku odbyły się prezentacje projektów "Założeń do ustawy 2.0", przygotowanych przez zespoły wyłonione w trybie konkursowym w 2016 roku. Rezultaty pracy to trzy koncepcje zmiany ładu legislacyjnego w polskiej nauce i szkolnictwie wyższym. Każdy z trzech projektów ma swoją dominującą logikę, jednak wszystkie próbują odpowiadać na podobne problemy polskiego systemu nauki i kształcenia wyższego, trafnie zidentyfikowane przez zespoły konkursowe na etapie diagnozy. Posługując się metodą porównawczą, pozwolę sobie poddać analizie propozycje Kolegów, zwracając uwagę przede wszystkim na rozwiązania mające poprawić sytuację polskich uczelni i całego systemu szkolnictwa.In Poland we are working on a modification of higher education system because the current solutions are not satisfactory for Polish education potential, as well as for people engaged in this field and, last but not least, students. Law HEI 2.0 (Ustawa 2.0) is supposed to be the strategic element of change. Polish academics prepared projects to the new regulation and three of them have been chosen. The article is a critical analysis of these proposals and it brings attention to solutions that might improve a situation of Polish higher education and the whole system of education

    Different proteolytic activities using natural and synthetic substrates were analyzed in the midgut of Cerambyx cerdo larvae. Significant leucyl-aminopeptidase and elastase-like activities, as well as some trypsin-like activities were detected.Korišćenjem prirodnih i sintetičkih supstrata analizirane su različite proteolitičke aktivnosti u srednjem crevu larve Cerambyx cerdo.     Synthesis, structural characterization, DFT calculations and antiproliferative evaluation of novel spirohydantoin derivatives containing a substituted benzyl moiety

    Two series of cycloalkanespiro-5-hydantoins, namely cyclohexanespiro-5-hydantoins and cycloheptanespiro-5-hydantoins with a 4-substituted benzyl or a 2-(4-substituted phenyl)-2-oxoethyl group at N3 position, were synthesized and their effects on proliferation of human colon (HCT-116), leukemia (K562) and breast (MDA-MB-231) cancer cell lines were tested. For comparison, we also described the 5,5-diphenylhydantoin analogues. The structural features of the investigated compounds were characterized by elemental analysis, FT-IR, UV–Vis, 1 H and 13 C NMR spectroscopy and X-ray crystallography. Regarding their structure–activity relationships, it was shown that the substitution on the benzyl moiety with the methoxy, chloro or bromo group potentiated the antiproliferative activity relative to the parent compounds, while an increase in the size of the cycloalkyl group resulted mostly in a decrease of the antiproliferative activity. The single crystal X-ray analysis revealed the existence of dimers and chains formed by the N–H⋯O hydrogen bonds. The analysis of the molecular descriptors of Lipinski demonstrated that all investigated compounds obeyed the rule of five. To further understand their geometry and electronic structure, DFT calculations with B3LYP method using 6-311++G(d,p) basic set were performed. In this context, the UV–Vis spectra of the investigated compounds were analyzed in detail, whereby the predicted absorption spectra from DFT calculation matched the experimentally obtained ones, with a good correlation.
