internacional decide no intervenir en la España de Franco, ello condena
al exilio perpetuo a varias decenas de miles de anónimos ciudadanos

Software-based Integrating Charge-Coupled Devices Signal Processing Using Correlated Double Sampling Technique
En este artículo se analiza un método para reconstruir una imagen de la señal de salida digitalizada de una cámara integradora de dispositivos acoplados por carga (escaneo lento) basada en software mediante el uso muestreo doble correlacionado y uso eficiente de los recursos de procesamiento para tiempos de procesamiento cortos.
El software desarrollado es enteramente escrito en C++ para una ejecución de alta velocidad y puede trabajar con datos adquiridos por diferentes tipos de dispositivos de detección con pequeños cambios. Este trabajo permite implementar una adquisición de bajo costo sistema para dispositivos de carga acoplada o proporcionar un desarrollo herramienta adecuada para probar filtros digitales, diferentes técnicas para píxeles computación o análisis de señales de dispositivos de carga acoplada.CONACYT – Consejo Nacional de Ciencia y TecnologíaPROCIENCI

Soluble platelet-endothelial cell adhesion molecule-1, a biomarker of ventilator-induced lung injury
Introduction: Endothelial cell injury is an important component of acute lung injury. Platelet-endothelial cell adhesion molecule-1 (PECAM1) is a transmembrane protein that connects endothelial cells to one another and can be detected as a soluble, truncated protein (sPECAM1) in serum. We hypothesized that injurious mechanical ventilation (MV) leads to shedding of PECAM1 from lung endothelial cells resulting in increasing sPECAM1 levels in the systemic circulation. Methods: We studied 36 Sprague–Dawley rats in two prospective, randomized, controlled studies (healthy and septic) using established animal models of ventilator-induced lung injury. Animals (n = 6 in each group) were randomized to spontaneous breathing or two MV strategies: low tidal volume (VT) (6 ml/kg) and high-VT (20 ml/kg) on 2 cmH2O of positive end-expiratory pressure (PEEP). In low-VT septic animals, 10 cmH2O of PEEP was applied. We performed pulmonary histological and physiological evaluation and measured lung PECAM1 protein content and serum sPECAM1 levels after four hours ventilation period. Results: High-VT MV caused severe lung injury in healthy and septic animals, and decreased lung PECAM1 protein content (P < 0.001). Animals on high-VT had a four- to six-fold increase of mean sPECAM1 serum levels than the unventilated counterpart (35.4 ± 10.4 versus 5.6 ± 1.7 ng/ml in healthy rats; 156.8 ± 47.6 versus 35.6 ± 12.6 ng/ml in septic rats) (P < 0.0001). Low-VT MV prevented these changes. Levels of sPECAM1 in healthy animals on high-VT MV paralleled the sPECAM1 levels of non-ventilated septic animals. Conclusions: Our findings suggest that circulating sPECAM1 may represent a promising biomarker for the detection and monitoring of ventilator-induced lung injury
Efficacy and safety of universal (TCRKO) ARI-0001 CAR-T cells for the treatment of B-cell lymphoma
Autologous T cells expressing the Chimeric Antigen Receptor (CAR) have been
approved as advanced therapy medicinal products (ATMPs) against several
hematological malignancies. However, the generation of patient-specific CART
products delays treatment and precludes standardization. Allogeneic off-theshelf
CAR-T cells are an alternative to simplify this complex and timeconsuming
process. Here we investigated safety and efficacy of knocking out
the TCR molecule in ARI-0001 CAR-T cells, a second generation aCD19 CAR
approved by the Spanish Agency of Medicines and Medical Devices (AEMPS)
under the Hospital Exemption for treatment of patients older than 25 years with
Relapsed/Refractory acute B cell lymphoblastic leukemia (B-ALL). We first
analyzed the efficacy and safety issues that arise during disruption of the TCR
gene using CRISPR/Cas9. We have shown that edition of TRAC locus in T cells
using CRISPR as ribonuleorproteins allows a highly efficient TCR disruption
(over 80%) without significant alterations on T cells phenotype and with an
increased percentage of energetic mitochondria. However, we also found that
efficient TCRKO can lead to on-target large and medium size deletions,
indicating a potential safety risk of this procedure that needs monitoring.
Importantly, TCR edition of ARI-0001 efficiently prevented allogeneic
responses and did not detectably alter their phenotype, while maintaining a
similar anti-tumor activity ex vivo and in vivo compared to unedited ARI-0001 CAR-T cells. In summary, we showed here that, although there are still some
risks of genotoxicity due to genome editing, disruption of the TCR is a feasible
strategy for the generation of functional allogeneic ARI-0001 CAR-T cells. We
propose to further validate this protocol for the treatment of patients that do

Acceleration of the DNA methylation clock among lynch syndrome‑associated mutation carriers

aging and disease. The purpose of this study is to assess whether or not individuals carrying Lynch Syndromeassociated
mutations are affected in their rate of biological aging, as measured by the epigenetic clock.
Methods: Genome-wide bisulfite DNA sequencing data were generated using DNA from CD4 + T-cells obtained
from peripheral blood using 27 patient samples from Lynch syndrome families. Horvath’s DNAm age model based on
penalized linear regression was applied to estimate DNAm age from patient samples with distinct clinical and genetic
characteristics to investigate cancer mutation-related aging effects.
Results: Both Lynch mutation carriers and controls exhibited high variability in their estimated DNAm age, but
regression analysis showed steeper slope for the Lynch mutation carriers. Remarkably, six Lynch Syndrome-associated
mutation carriers showed a strong correlation to the control group, and two sisters carrying Lynch Syndrome-associated
mutations, with no significant difference in lifestyle and similar chronological age, were assigned very different
DNAm age.
Conclusions: Future studies will be required to explore, in larger patient populations, whether specific epigenetic
age acceleration is predictive of time-to-cancer development, treatment response, and survival. Epigenetic clock
DNAm metrics may be affected by the presence of cancer mutations in the germline, and thus show promise of
potential clinical utility for stratified surveillance strategies based on the relative risk for imminent emergence of

Faja milonítica El Cortijo: puesta en valor de su patrimonio geominero y propuesta de un nuevo sitio de interés geológico en Tandil, Buenos Aires
El basamento del Sistema de Tandilia reúne a las rocas más antiguas de la Argentina, las cuales guardan una valiosa información de los procesos geológicos ocurridos hace aproximadamente 2.200 Ma. Entre estas rocas ígneo-metamórficas (Complejo Buenos Aires) se encuentran las que integran la Formación El Cortijo, que contrastan notoriamente con el resto por sus características mineralógicas, metamórficas y estructurales. Esta Formación está constituida por rocas típicas de una cuenca oceánica desarrollada durante el lapso Neoarqueano - Paleoproterozoico, fuertemente milonitizadas. Aflora en las cercanías de la ciudad de Tandil, a lo largo de una faja de 3 kilómetros con rumbo este-oeste (aquí denominada Faja milonítica El Cortijo), a la que se accede por la Ruta Nacional 226. Tandil es una ciudad reconocida por su origen minero, actualmente con una fuerte impronta turística y una infraestructura óptima para ofrecer diversos circuitos geomineros. Esta contribución tiene como objetivo presentar un nuevo sitio de interés geológico en Tandil, que junto con los otros circuitos geoturísticos urbanos, manifieste el valor geológico de la región. Esto conduciría a su protección y podría impulsar la creación de un futuro Parque Geológico, producto de la integración de todos los geositios conocidos además del aquí propuesto. El trabajo realizado incluyó un relevamiento del área mediante imágenes satelitales y actividades de campo que permitieron seleccionar un conjunto de canteras labradas en la faja milonítica que constituye el geositio propuesto. Este sector de la provincia de Buenos Aires reúne todos los atributos necesarios para establecer un sitio de interés geológico como el sugerido, considerando sus riquezas paisajísticas y geológicas relacionadas con la evolución geológica y tectónica del basamento de Tandilia
Lentiviral vectors for inducible, transactivator-free advanced therapy medicinal products: Application to CAR-T cells
Controlling transgene expression through an externally
administered inductor is envisioned as a potent strategy
to improve safety and efficacy of gene therapy approaches.
Generally, inducible ON systems require a chimeric transcription
factor (transactivator) that becomes activated by
an inductor, which is not optimal for clinical translation
due to their toxicity. We generated previously the first
all-in-one, transactivator-free, doxycycline (Dox)-responsive
(Lent-On-Plus or LOP) lentiviral vectors (LVs) able to control
transgene expression in human stem cells. Here, we
have generated new versions of the LOP LVs and have
analyzed their applicability for the generation of inducible
advanced therapy medicinal products (ATMPs) with special
focus on primary human T cells. We have shown that, contrary
to all other cell types analyzed, an Is2 insulator must
be inserted into the 30 long terminal repeat of the LOP
LVs in order to control transgene expression in human
primary T cells. Importantly, inducible primary T cells
generated by the LOPIs2 LVs are responsive to ultralow
doses of Dox and have no changes in phenotype or function
compared with untransduced T cells. We validated
the LOPIs2 system by generating inducible CAR-T cells
that selectively kill CD19+ cells in the presence of Dox.
In summary, we describe here the first transactivatorfree,
all-one-one system capable of generating Dox-inducible

Predictors of Response to Exclusive Enteral Nutrition in Newly Diagnosed Crohn´s Disease in Children: PRESENCE Study from SEGHNP
Exclusive enteral nutrition (EEN) has been shown to be more effective than corticosteroids in achieving mucosal healing in children with Crohn´s disease (CD) without the adverse effects of these drugs. The aims of this study were to determine the efficacy of EEN in terms of inducing clinical remission in children newly diagnosed with CD, to describe the predictive factors of response to EEN and the need for treatment with biological agents during the first 12 months of the disease. We conducted an observational retrospective multicentre study that included paediatric patients newly diagnosed with CD between 2014–2016 who underwent EEN. Two hundred and twenty-two patients (140 males) from 35 paediatric centres were included, with a mean age at diagnosis of 11.6 ± 2.5 years. The median EEN duration was 8 weeks (IQR 6.6–8.5), and 184 of the patients (83%) achieved clinical remission (weighted paediatric Crohn’s Disease activity index [wPCDAI] 15 mg/L and ileal involvement tended to respond better to EEN. EEN administered for 6–8 weeks is effective for inducing clinical remission. Due to the high response rate in our series, EEN should be used as the first-line therapy in luminal paediatric Crohn’s disease regardless of the location of disease and disease activityS
Clonal chromosomal mosaicism and loss of chromosome Y in elderly men increase vulnerability for SARS-CoV-2
The pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2, COVID-19) had an estimated overall case fatality ratio of 1.38% (pre-vaccination), being 53% higher in males and increasing exponentially with age. Among 9578 individuals diagnosed with COVID-19 in the SCOURGE study, we found 133 cases (1.42%) with detectable clonal mosaicism for chromosome alterations (mCA) and 226 males (5.08%) with acquired loss of chromosome Y (LOY). Individuals with clonal mosaic events (mCA and/or LOY) showed a 54% increase in the risk of COVID-19 lethality. LOY is associated with transcriptomic biomarkers of immune dysfunction, pro-coagulation activity and cardiovascular risk. Interferon-induced genes involved in the initial immune response to SARS-CoV-2 are also down-regulated in LOY. Thus, mCA and LOY underlie at least part of the sex-biased severity and mortality of COVID-19 in aging patients. Given its potential therapeutic and prognostic relevance, evaluation of clonal mosaicism should be implemented as biomarker of COVID-19 severity in elderly people.
