    Soil contamination following an industrial accident : towards efficient investigations and assessment

    International audienceWhen an industrial accident occurs, e.g. the explosion or the fire of a chemical facility, soil investigations and subsequent risk mitigation generally need to be decided and performed rapidly. This requires specific organisation and tools: Procedures for an immediate and coordinated intervention of relevant actors: industrials, administrations for industrial facilities, emergency and health, local authorities, environmental consultants and laboratories, NGOs. Models and input data on emission, atmospheric transfer and deposition on soil, for an accidental source; investigation plans and adequate soil quality references, guidelines... But the European Seveso legislation, and its application in France and probably in Europe, is focused on the prevention of immediate impacts on health and constructions; and the Integrated Pollution Prevention and Control (IPPC) legislation deals with the chronic impacts of operating facilities. Thus, post-accidental impacts of industrial accidents are hardly dealt with, the specific organisation and tools are lacking, and when accidents occur, the industrials and administrations concerned are largely unprepared for managing their delayed impacts, first of them on soils

    Scour potential at Laouzas Dam

    Laouzas Dam, owned and operated by Electricité de France (EDF), is a 52 m high double-curvature arch dam in a large valley located on the VÚbre River in the Languedoc-Roussillon Midi Pyrénées region of France. The dam is founded on heavily fractured granite and migmatite rock. It houses a surface spillway equipped with 3 radial gates. The radial gates are positioned such that their gate lip is systematically located about 1m upstream of the spillway crest itself. This particular design generates aerated jets that do not behave like normal falling jets. Despite the absence of severe spillages since dam construction in 1961-1965, the ones observed in the past have allowed creating locally a 5 m deep scour hole along the right hand side of the rock mass. The granite and migmatite have significant unconfined compressive strength. The rock mass fracturing is composed of 3-4 joint sets and shows desquamation joints affecting both abutments. EDF has commissioned a numerical study to assess scour potential downstream of the dam as well as the positive influence of rock anchors, by using the Comprehensive Scour Model (CSM)

    The data and estimation methods presented in this article are associated with the research article, "Cut-off values of serum potassium and core temperature at hospital admission for extracorporeal rewarming of avalanche victims in cardiac arrest: a retrospective multi-centre study" [1]. In this article we estimate recommended cut-off values for in-hospital triage with respect to extracorporeal rewarming. With only 6 survivors of 103 patients collected over a period of 20 years the ability to estimate reliable threshold values is limited. In addition, because the number of avalanche victims is also limited, a significantly larger dataset is unlikely to be obtained. We have therefore adapted two non-parametric estimation methods (bootstrapping and exact binomial distribution) to our specific needs and performed a simulations to confirm validity and reliability

    HIF2α reduces growth rate but promotes angiogenesis in a mouse model of neuroblastoma

    <p>Abstract</p> <p>Background</p> <p>HIF2α/EPAS1 is a hypoxia-inducible transcription factor involved in catecholamine homeostasis, vascular remodelling, physiological angiogenesis and adipogenesis. It is overexpressed in many cancerous tissues, but its exact role in tumour progression remains to be clarified.</p> <p>Methods</p> <p>In order to better establish its function in tumourigenesis and tumour angiogenesis, we have stably transfected mouse neuroblastoma N1E-115 cells with the native form of HIF2α or with its dominant negative mutant, HIF2α (1–485) and studied their phenotype <it>in vitro </it>and <it>in vivo</it>.</p> <p>Results</p> <p><it>In vitro </it>studies reveal that HIF2α induces neuroblastoma cells hypertrophy and decreases their proliferation rate, while its inactivation by the HIF2α (1–485) mutant leads to a reduced cell size, associated with an accelerated proliferation. However, our <it>in vivo </it>experiments show that subcutaneous injection of cells overexpressing HIF2α into syngenic mice, leads to the formation of tumour nodules that grow slower than controls, but that are well structured and highly vascularized. In contrast, HIF2α (1–485)-expressing neuroblastomas grow fast, but are poorly vascularized and quickly tend to extended necrosis.</p> <p>Conclusion</p> <p>Together, our data reveal an unexpected combination between an antiproliferative and a pro-angiogenic function of HIF2α that actually seems to be favourable to the establishment of neuroblastomas <it>in vivo</it>.</p

    HIF-1α determines the metastatic potential of gastric cancer cells

    Gastric adenocarcinoma is characterised by rapid emergence of systemic metastases, resulting in poor prognosis due to vanished curative treatment options. Better understanding of the molecular basis of gastric cancer spread is needed to design innovative treatments. The transcription factor HIF-1α (hypoxia-inducible factor 1α) is frequently overexpressed in human gastric cancer, and inhibition of HIF-1α has proven antitumour efficacy in rodent models, whereas the relevance of HIF-1α for the metastatic phenotype of gastric adenocarcinoma remains elusive. Therefore, we have conducted a comprehensive analysis of the role of HIF-1α for pivotal metastasis-associated processes of human gastric cancer. Immunhistochemistry for HIF-1α showed specific staining at the invading tumour edge in 90% of human gastric cancer samples, whereas normal gastric tissue was negative and only a minority of early gastric cancers (T1 tumours) showed specific staining. Hypoxia-inducible factor 1α-deficient cells showed a significant reduction of migratory, invasive and adhesive properties in vitro. Furthermore, the HIF-1α-inhibitor 2-methoxy-estradiol significantly reduced metastatic properties of gastric cancer cells. The accentuated expression at the invading edge together with the in vitro requirement of HIF-1α for migration, invasion and adherence argues for a pivotal role of HIF-1α in local invasion and, ultimately, systemic tumour spread. These results warrant the exploration of HIF-1α-inhibiting substances in clinical treatment studies of advanced gastric cancer
