    Substrate Moisture Content Effects on Growth and Shelf Life of Angelonia angustifolia

    Wilting during shelf life is a major cause of postharvest shrink for bedding plants shipped long distances from production greenhouses to retail outlets. The objective of this research was to determine if irrigation at lower, constant substrate moisture content (SMC) during greenhouse production would be a feasible way to acclimate plants for reduced shrinkage during shelf life, while potentially conserving irrigation water. Rooted plugs of Angelonia angustifolia 'Angelface Blue' were grown in greenhouse production until a marketable stage in substrates irrigated at SMC levels of 10, 20, 30, and 40% using a controlled irrigation system. At the end of the greenhouse production stage, plants were irrigated to container capacity and subjected to a simulated shipping environment, in shipping boxes in the dark for two days. After shipping, plants were placed back in the greenhouse and watered minimally to simulate a retail environment. Data was taken at the end of each stage i.e. greenhouse production, simulated shipping, and simulated retail. Parameters measured at the end of the production stage were fresh and senesced flower number, stem number, pre-dawn and mid-day water potential, SPAD meter readings (Experiment 2), and plant height and node number segmented into vegetative, flowering, and bud area. Plant quality was observed and rated. At the end of the simulated retail stage, the same data was taken, along with fresh and dry shoot and root weight. Results indicated that as SMC decreased from 40 to 10%, plants were shorter in height, but had proportional flowering sections (Experiment 1) or more compact flowering sections (Experiment 2). The volume of water received by the 40% SMC plants was three times greater (Experiment 1) and 12 times greater (Experiment 2) than the 20% SMC plants during greenhouse production, and two times greater (Experiment 1) and nine time greater (Experiment 2) during simulated retail. Additionally, the 40% SMC plants used 15 liters (Experiment 1) and 38 liters (Experiment 2) of water during greenhouse production compared to the 20% SMC plants using only three liters in both experiments. During simulated retail the 40% SMC plants used six liters (Experiment 1) and nine liters (Experiment 2) of water while the 20% SMC plants used five liters (Experiment 1) and three liters (Experiment 2) of water. During production, mid-day water potentials decreased as the SMC levels decreased, but at the end of the simulated retail (Experiment 1), the mid-day water potentials were all the same, suggesting plants that were drought stressed during production area were acclimated to lower water levels experienced in retail settings. Overall, the 20% SMC treatment produced the best postharvest quality plant due to reduced plant height without detrimental effects on flowering. The results demonstrate that while conserving water, controlled irrigation at a medium-low SMC can produce high quality plants that have equal shelf life to those that are irrigated at high levels

    Maintaining independence in individuals with dementia at home after a fall:a protocol for the UK pilot cluster randomised controlled trial MAINTAIN

    Introduction: Individuals with dementia face an increased risk of falls. Falls can cause a decline in the individual’s overall functionality. All types of falls, including those that do not result in injury, can lead to psychosocial consequences, such as diminished confidence and a fear of falling. Projections indicate a rising trend in dementia diagnoses, implying an increase in fall incidents. Yet, there is a lack of evidence to support interventions for people living with dementia who have fallen. Our objective is to test the feasibility of a falls intervention trial for people with dementia. Method and analysis: This is a UK-based two-arm pilot cluster randomised controlled trial. In this study, six collaborating sites, which form the clusters, will be randomly allocated to either the intervention arm or the control arm (receiving treatment as usual) at a 1:1 ratio. During the 6 month recruitment phase, each cluster will enrol 10 dyads, comprising 10 individuals with dementia and their respective carers, leading to a total sample size of 60 dyads. The primary outcomes are the feasibility parameters for a full trial (ie, percentage consented, follow-up rate and cost framework). Secondary outcomes include activities of daily living, quality of life, fall efficacy, mobility, goal attainment, cognitive status, occurrence of falls, carer burden and healthcare service utilisation. Outcome measures will be collected at baseline and 28 weeks, with an additional assessment scheduled at 12 weeks for the healthcare service utilisation questionnaire. An embedded process evaluation, consisting of interviews and observations with participants and healthcare professionals, will explore how the intervention operates and the fidelity of study processes. Ethics and dissemination: The study was approved by the NHS and local authority research governance and research ethics committees (NHS REC reference: 23/WA/0126). The results will be shared at meetings and conferences and will be published in peer-reviewed journals. Trial registration number: ISRCTN16413728

    Irregular wave runup statistics on plane beaches: application of a Boussinesq-type model incorporating a generating-absorbing sponge layer and second-order wave generation

    Efficient absorption of reflected waves at the offshore boundary is a prerequisite for the accurate physical or theoretical modelling of long-duration irregular wave runup statistics at uniform, gently sloped beaches. This paper presents an implementation of the method suggested by Zhang et al. (2014) to achieve reflected wave absorption and simultaneous generation and propagation of incident waves in an existing numerical wave flume incorporating a moving boundary wavemaker. A generating–absorbing layer is incorporated within this 1DH hybrid Boussinesq-nonlinear shallow water equation model such that inshore-travelling incident waves, encompassing bound-wave structure approximately correct to second order, propagate unhindered while offshore-travelling reflected waves are absorbed. Once validated, the method is used to compile random wave runup statistics on uniform beach slopes broadly representative of dissipative, intermediate, and reflective beaches. Analyses of the individual runup time series, ensemble statistics and comparison to an empirical formula based on experimental runup data suggest that the main aspects of runup observed in the field are properly represented by the model. Existence of an upper limit on maximum runup is investigated using a simple extreme-value statistical analysis. Spectral saturation is examined by considering ensemble-averaged swash spectra for three representative beach slopes subject to incident waves with two different offshore significant wave heights. All spectra show f^−4 roll-off at high frequencies in agreement with many previous field studies. The effect is also investigated of the swash motions preceding one particular extreme runup event on the eventual maximum runup elevation

    Somatically acquired hypomethylation of IGF2 in breast and colorectal cancer

    The imprinted insulin-like growth factor 2 (IGF2) gene is expressed predominantly from the paternal allele. Loss of imprinting (LOI) associated with hypomethylation at the promoter proximal sequence (DMR0) of the IGF2 gene was proposed as a predisposing constitutive risk biomarker for colorectal cancer. We used pyrosequencing to assess whether IGF2 DMR0 methylation is either present constitutively prior to cancer or whether it is acquired tissue-specifically after the onset of cancer. DNA samples from tumour tissues and matched non-tumour tissues from 22 breast and 42 colorectal cancer patients as well as peripheral blood samples obtained from colorectal cancer patients [SEARCH (n=case 192, controls 96)], breast cancer patients [ABC (n=case 364, controls 96)] and the European Prospective Investigation of Cancer [EPIC-Norfolk (n=breast 228, colorectal 225, controls 895)] were analysed. The EPIC samples were collected 2–5 years prior to diagnosis of breast or colorectal cancer. IGF2 DMR0 methylation levels in tumours were lower than matched non-tumour tissue. Hypomethylation of DMR0 was detected in breast (33%) and colorectal (80%) tumour tissues with a higher frequency than LOI indicating that methylation levels are a better indicator of cancer than LOI. In the EPIC population, the prevalence of IGF2 DMR0 hypomethylation was 9.5% and this correlated with increased age not cancer risk. Thus, IGF2 DMR0 hypomethylation occurs as an acquired tissue-specific somatic event rather than a constitutive innate epimutation. These results indicate that IGF2 DMR0 hypomethylation has diagnostic potential for colon cancer rather than value as a surrogate biomarker for constitutive LOI

    Ethnic differences in blood lipids and dietary intake between UK children of black African, black Caribbean, South Asian, and white European origin: the Child Heart and Health Study in England (CHASE).

    BACKGROUND: Ischemic heart disease (IHD) rates are lower in UK black Africans and black Caribbeans and higher in South Asians when compared with white Europeans. Ethnic differences in lipid concentrations may play a part in these differences. OBJECTIVE: The objective was to investigate blood lipid and dietary patterns in UK children from different ethnic groups. DESIGN: This was a cross-sectional study in 2026 UK children (including 285 black Africans, 188 black Caribbeans, 534 South Asians, and 512 white Europeans) attending primary schools in London, Birmingham, and Leicester. We measured fasting blood lipid concentrations and collected 24-h dietary recalls. RESULTS: In comparison with white Europeans, black African children had lower total cholesterol (-0.14 mmol/L; 95% CI: -0.25, -0.04 mmol/L), LDL-cholesterol (-0.10 mmol/L; 95% CI: -0.20, -0.01 mmol/L), and triglyceride concentrations (proportional difference: -0.11 mmol/L; 95% CI: -0.16, -0.06 mmol/L); HDL-cholesterol concentrations were similar. Lower saturated fat intakes (-1.4%; 95% CI: -1.9%, -0.9%) explained the differences between total and LDL cholesterol. Black Caribbean children had total, LDL-cholesterol, HDL-cholesterol, and triglyceride concentrations similar to those for white Europeans, with slightly lower saturated fat intakes. South Asian children had total and LDL-cholesterol concentrations similar to those for white Europeans, lower HDL-cholesterol concentrations (-0.7 mmol/L; 95% CI: -0.11, -0.03 mmol/L), and elevated triglyceride concentrations (proportional difference: 0.14 mmol/L; 95% CI: 0.09, 0.20 mmol/L); higher polyunsaturated and monounsaturated fat intakes did not explain these lipid differences. CONCLUSIONS: Only black African children had a blood lipid profile and associated dietary pattern likely to protect against future IHD. The loss of historically lower LDL-cholesterol concentrations among UK black Caribbeans and South Asians may have important adverse consequences for future IHD risk in these groups

    Bulky DNA adducts, 4-aminobiphenyl-haemoglobin adducts and diet in the European Prospective Investigation into Cancer and Nutrition (EPIC) prospective study

    In contrast to some extensively examined food mutagens, for example, aflatoxins, N-nitrosamines and heterocyclic amines, some other food contaminants, in particular polycyclic aromatic hydrocarbons (PAH) and other aromatic compounds, have received less attention. Therefore, exploring the relationships between dietary habits and the levels of biomarkers related to exposure to aromatic compounds is highly relevant. We have investigated in the European Prospective Investigation into Cancer and Nutrition (EPIC) cohort the association between dietary items (food groups and nutrients) and aromatic DNA adducts and 4-aminobiphenyl-Hb adducts. Both types of adducts are biomarkers of carcinogen exposure and possibly of cancer risk, and were measured, respectively, in leucocytes and erythrocytes of 1086 (DNA adducts) and 190 (Hb adducts) non-smokers. An inverse. statistically significant, association has been found between DNA adduct levels and dietary fibre intake (P=0.02), vitamin E (P =0.04) and alcohol (P=0.03) but not with other nutrients or food groups. Also, an inverse association between fibre and fruit intake, and BMI and 4-aminobiphenyl-Hb adducts (P=0.03, 0.04, and 0.03 respectively) was observed. After multivariate regression analysis these inverse correlations remained statistically significant, except for the correlation adducts v. fruit intake. The present study suggests that fibre intake in the usual range can modify the level of DNA or Hb aromatic adducts, but Such role seems to be quantitatively modest. Fibres could reduce the formation of DNA adducts in different manners, by diluting potential food mutagens and carcinogens in the gastrointestinal tract, by speeding their transit through the colon and by binding carcinogenic substances

    Activation of a cGAS-STING-mediated immune response predicts response to neoadjuvant chemotherapy in early breast cancer.

    BACKGROUND: The DNA-damage immune-response (DDIR) signature is an immune-driven gene expression signature retrospectively validated as predicting response to anthracycline-based therapy. This feasibility study prospectively evaluates the use of this assay to predict neoadjuvant chemotherapy response in early breast cancer. METHODS: This feasibility study assessed the integration of a novel biomarker into clinical workflows. Tumour samples were collected from patients receiving standard of care neoadjuvant chemotherapy (FEC + /-taxane and anti-HER2 therapy as appropriate) at baseline, mid- and post-chemotherapy. Baseline DDIR signature scores were correlated with pathological treatment response. RNA sequencing was used to assess chemotherapy/response-related changes in biologically linked gene signatures. RESULTS: DDIR signature reports were available within 14 days for 97.8% of 46 patients (13 TNBC, 16 HER2 + ve, 27 ER + HER2-ve). Positive scores predicted response to treatment (odds ratio 4.67 for RCB 0-1 disease (95% CI 1.13-15.09, P = 0.032)). DDIR positivity correlated with immune infiltration and upregulated immune-checkpoint gene expression. CONCLUSIONS: This study validates the DDIR signature as predictive of response to neoadjuvant chemotherapy which can be integrated into clinical workflows, potentially identifying a subgroup with high sensitivity to anthracycline chemotherapy. Transcriptomic data suggest induction with anthracycline-containing regimens in immune restricted, "cold" tumours may be effective for immune priming.
