    Development and application of test cases for comparing vertical ground heat exchanger models

    The main objective of this paper is to establish a set of test cases for analytical verifications and intermodel comparisons of ground heat exchanger (GHX) models used in building simulation programs. Several test cases are suggested. They range from steady-state heat rejection in a single borehole to varying hourly loads with large yearly thermal imbalance in multiple borehole configurations. The usefulness of the proposed test cases is illustrated by running them with different GHX models. This comparison exercise has shown that 1-D models compare favourably well with the more elaborate 3-D models for relatively small simulation periods. The cyclic heat rejection/collection test has revealed some small deficiency in the load aggregation scheme of a particular model. Finally, the use of the asymmetric (cooling-dominated) load profile test case for a bore field composed of 100 boreholes revealed that the borehole wall temperature predicted by two GHX models can differ by as much as 10oC after a 30 year simulation

    Egr-1 inhibits the expression of extracellular matrix genes in chondrocytes by TNFα-induced MEK/ERK signalling

    Introduction TNFα is increased in the synovial fluid of patients with rheumatoid arthritis and osteoarthritis. TNFα activates mitogen-activated kinase kinase (MEK)/extracellular regulated kinase (ERK) in chondrocytes; however, the overall functional relevance of MEK/ERK to TNFα-regulated gene expression in chondrocytes is unknown. Methods Chondrocytes were treated with TNFα with or without the MEK1/2 inhibitor U0126 for 24 hours. Microarray analysis and real-time PCR analyses were used to identify genes regulated by TNFα in a MEK1/2-dependent fashion. Promoter/ reporter, immunoblot, and electrophoretic mobility shift assays were used to identify transcription factors whose activity in response to TNFα was MEK1/2 dependent. Decoy oligodeoxynucleotides bearing consensus transcription factor binding sites were introduced into chondrocytes to determine the functionality of our results. Results Approximately 20% of the genes regulated by TNFα in chondrocytes were sensitive to U0126. Transcript regulation of the cartilage-selective matrix genes Col2a1, Agc1 and Hapln1, and of the matrix metalloproteinase genes Mmp-12 and Mmp-9, were U0126 sensitive – whereas regulation of the inflammatory gene macrophage Csf-1 was U0126 insensitive. TNFα-induced regulation of Sox9 and NFKB activity was also U0126 insensitive. Conversely, TNFα-increased early growth response 1 (Egr-1) DNA binding was U0126 sensitive. Transfection of chondrocytes with cognate Egr-1 oligodeoxynucleotides attenuated the ability of TNFα to suppress Col2a1, Agc1 or Hapln1 mRNA expression. Conclusions Our results suggest that MEK/ERK and Egr1 are required for TNFα-regulated catabolic and anabolic genes of the cartilage extracellular matrix, and hence may represent potential targets for drug intervention in osteoarthritis or rheumatoid arthritis

    Modélisation de l'évolution du couvert nival, sur le sous-bassin LG4 de la rivière La Grande dans le nord du Québec, à l'aide du modèle français CROCUS

    Au Québec, l'accumulation de la neige au sol prend une grande importance dans bon nombre d'activités humaines comme la production d'hydroélectricité. Face à cette importance de la neige, il devient fort intéressant de faire appel à des outils de modélisation dans le but de faire le suivi complet du manteau neigeux par la simulation des différents processus physiques agissant sur le couvert nival. En ce sens, la présente recherche vise à évaluer la capacité d'adaptation aux conditions environnementales du Québec, du modèle français multicouches CROCUS de suivi et de fonte du couvert nival, en vue de son utilisation ultérieure. Le choix du site d'étude s'accorde avec la présence de stations d'acquisition de données sur le couvert nival et la disponibilité d'un large éventail de données météorologiques essentielles aux modélisations.Suite au prétraitement des données d'entrée, le modèle CROCUS a pu être étalonné dans le but d'évaluer ses aptitudes à simuler l'évolution de l'épaisseur et de l'équivalent en eau du couvert nival, de même que les profils de densité et de température de ce couvert. Compte tenu des données disponibles, les résultats des périodes d'étalonnage et de vérification peuvent être considérés comme très bons et très encourageants pour des applications ultérieures de CROCUS au Québec.In Québec, snow accumulation on the ground is important for a large number of activities, including hydroelectric production. Snow being an important environmental aspect to look at for operations in cold regions, making use of adequate models able to simulate the various physical processes affecting the characteristics of the snowpack throughout the entire snow season becomes also quite important. So, the present project aims to evaluate the ability of the CROCUS French multilayer snow model to be adapted to Northern Quebec environmental conditions. The choice of the study area has been done on the basis of data availability on both the temporal evolution the snowpack at three automatic stations and the meteorological variables necessary as input to the model. Following preprocessing of input data, the CROCUS model has been calibrated in order to evaluate his capability to simulate the temporal evolution not only of the thickness and water equivalent of the snowpack, but also of the density and temperature profiles within the pack. Given the available data, the results for both the calibration and verification periods can be considered as good and very encouraging for future applications of the CROCUS model in Québec

    U(1) spin liquids and valence bond solids in a large-N three-dimensional Heisenberg model

    We study possible quantum ground states of the Sp(N) generalized Heisenberg model on a cubic lattice with nearest-neighbor and next-nearest-neighbor exchange interactions. The phase diagram is obtained in the large-N limit and fluctuation effects are considered via appropriate gauge theories. In particular, we find three U(1) spin liquid phases with different short-range magnetic correlations. These phases are characterized by deconfined gapped spinons, gapped monopoles, and gapless ``photons''. As N becomes smaller, a confinement transition from these phases to valence bond solids (VBS) may occur. This transition is studied by using duality and analyzing the resulting theory of monopoles coupled to a non-compact dual gauge field; the condensation of the monopoles leads to VBS phases. We determine the resulting VBS phases emerging from two of the three spin liquid states. On the other hand, the spin liquid state near J_1 \approx J_2 appears to be more stable against monopole condensation and could be a promising candidate for a spin liquid state in real systems.Comment: revtex file 12 pages, 17 figure

    RF and THz Identification Using a New Generation of Chipless RFID Tags

    This article presents two chipless RFID approaches where data are reading using electromagnetic waves and where the medium encoding the data is completely passive. The former approach rests on the use of RF waves (more precisely the ultra-wide band UWB). The tags developed for this application are comparable with very specific, planar, conductive, radar targets where the relation between the tag geometry and its electromagnetic signature is perfectly known and is used to encode the data. The principle of operation as well as the realization process of the RF tags presented in this paper is similar to those already reported in the literature. However, contrary to the majority of chipless RFID tags, these labels do not present an antenna function dissociated from the circuit part where the data are stored. Here, functions such as the receiver, the treatment and the emitter of the signal are closely dependent. The data storage capacity of the RF chipless tags is proportional to of the used frequency bandwidth. As radio spectrum is regulated, the number of possible encoding bits is thus strongly limited with this technology. This is the reason why we introduce a new family of tags radically different from the preceding one, where data is encoded in volume thanks to a multilayer structure operating in the THz domain. These two approaches although different are complementary and allow to increase significantly the data storage capacity of the chipless tags. Simulation and experimental results are reported in this paper for both configurations. We demonstrate a coding capacity of 3.3 bit/cm2 for RFID chipless tags and a potential 10 bits coding capacity in the THz domain

    Regulation of Sox9 activity by crosstalk with nuclear factor-κB and retinoic acid receptors

    Introduction: Sox9 and p300 cooperate to induce expression of cartilage-specific matrix proteins, including type II collagen, aggrecan and link protein. Tumour necrosis factor (TNF)-alpha, found in arthritic joints, activates nuclear factor-kappaB (NF-kappaB), whereas retinoic acid receptors (RARs) are activated by retinoid agonists, including all-trans retinoic acid (atRA). Like Sox9, the activity of NF-kappaB and RARs depends upon their association with p300. Separately, both TNF-alpha and atRA suppress cartilage matrix gene expression. We investigated how TNF-alpha and atRA alter the expression of cartilage matrix genes. Methods: Primary cultures of rat chondrocytes were treated with TNF-alpha and/or atRA for 24 hours. Levels of transcripts encoding cartilage matrix proteins were determined by Northern blot analyses and quantitative real-time PCR. Nuclear protein levels, DNA binding and functional activity of transcription factors were assessed by immunoblotting, electrophoretic mobility shift assays and reporter assays, respectively. Results: Together, TNF-alpha and atRA diminished transcript levels of cartilage matrix proteins and Sox9 activity more than each factor alone. However, neither agent altered nuclear levels of Sox9, and TNF-alpha did not affect protein binding to the Col2a1 48-base-pair minimal enhancer sequence. The effect of TNF-alpha, but not that of atRA, on Sox9 activity was dependent on NF-kappaB activation. Furthermore, atRA reduced NF-kappaB activity and DNA binding. To address the role of p300, we over-expressed constitutively active mitogen-activated protein kinase kinase kinase (caMEKK)1 to increase p300 acetylase activity. caMEKK1 enhanced basal NF-kappaB activity and atRA-induced RAR activity. Over-expression of caMEKK1 also enhanced basal Sox9 activity and suppressed the inhibitory effects of TNF-alpha and atRA on Sox9 function. In addition, over-expression of p300 restored Sox9 activity suppressed by TNF-alpha and atRA to normal levels. Conclusion: NF-kappaB and RARs converge to reduce Sox9 activity and cartilage matrix gene expression, probably by limiting the availability of p300. This process may be critical for the loss of cartilage matrix synthesis in inflammatory joint diseases. Therefore, agents that increase p300 levels or activity in chondrocytes may be useful therapeutically

    Mise en place d’une base de données pour une modélisation hydrologique distribuée du bassin versant du Bandama (Côte d’Ivoire) : apport d’un modèle numérique d’altitude, de la télédétection et du SIG Physitel

    Le fleuve Bandama, l’un des plus importants fleuves de la Côte d’Ivoire (Afrique de l’Ouest) est le seul dont le bassin versant est entièrement situé sur le territoire ivoirien. Il prend sa source dans le Nord du pays etcoule globalement dans une direction Nord-Sud. Son bassin versant est à cheval sur les deux principales formations végétales caractéristiques des régions tropicales : la savane (Nord) et la forêt (Sud). Le déplacement du front forêt-savane, causé par les coupes anarchiques de bois, les extensions de plantations industrielles ont eu des impacts sur le régime des écoulements (de surface et souterrains) des eaux dans cet hydrosystème. De plus, les aménagements hydrauliques (construction des barrages hydro-électriques de Kossou et Taabo) et hydro-agricoles (construction d’environ 250 barrages pour l’irrigation) ont encore desimpacts très importants sur le fonctionnement et les caractéristiques hydrogéologiques du bassin. Pour mieux comprendre le fonctionnement de cet hydrosystème et dans le contexte d’une gestion durable desressources en eau, la modélisation hydrologique du bassin versant du Bandama s’avère nécessaire. L’objectif de cette étude est de fournir les paramètres physiques du bassin versant du Bandama à partir dumodèle numérique d’Altitude (MNA) et de la télédétection, utilisables à des fins de modélisation hydrologique distribuée. La démarche adoptée comporte deux (2) étapes. La première étape consiste en la cartographie des descripteurs physiques du bassin qui influencent les processus hydrologiques tels que la topographie, le réseau hydrographique, la couverture végétale et le type de sol. A partir du module PHYSITEL et d’un Modèle Numérique d’Altitude (MNA), les cartes des pentes et des orientations des facettes sont dérivées et ensuite couplées au réseau hydrographique pour la caractérisation de la structure interne d’écoulement du bassin et la détermination d’Unités Hydrologiques Relativement Homogènes (UHRH : petits sous bassins versants utilisés comme unités de simulations des processus hydrologiques). La classification supervisée des images satellitaires MODIS permet de produire des cartes d’occupation du sol. La carte des types de sols est extraite de la carte pédologique de la Côte d’Ivoire. La seconde étape consiste au croisement des cartes thématiques établies. La combinaison de ces cartes thématiques permet de déterminer le type de sol dominant et les occupations du sol par UHRH.Mots-clés : hydrosystème, modélisation hydrologique, MNA, télédétection, PHYSITEL, UHRH

    Material and structural behaviour of PMMA from low temperatures to over the glass transition: Quasi-static and dynamic loading

    This work aims at characterizing the mechanical behaviour of polymethyl-methacrylate (PMMA) under high velocity impact conditions over a wide range of testing temperatures. To this end, the mechanical response at uniaxial compression is studied for both quasi-static and dynamic conditions covering testing temperatures below, at and above glass transition. A pseudo-brittle to ductile transition in the failure of PMMA is observed at a threshold that depends on testing temperature and strain rate. This analysis allows for the interpretation of the perforation impact tests and to explain the principal deformation and failure mechanisms. To complete the study, the Richeton model to predict yielding is revisited. Finally, we provide a new constitutive model for finite deformations to further identify the deformation mechanisms governing the mechanical behaviour of PMMA and the influence of temperature and strain rate on them.D.     The induction of CCN2 by TGFβ1 involves Ets-1

    CCN2 is encoded by an immediate-early gene induced in mesenchymal cells during the formation of blood vessels, bone and connective tissue. It plays key roles in cell adhesion and migration, as well as matrix remodeling. CCN2 is overexpressed in fibrosis, arthritis and cancer; thus, an understanding of how to control CCN2 expression is likely to have importance in developing therapies to combat these pathologies. Previously, we found that the promoter sequence GAGGAATG is important for Ccn2 gene regulation in NIH 3T3 fibroblasts. In this report, we show that this sequence mediates activation of the CCN2 promoter by the ETS family of transcription factors. Endogenous Ets-1 binds this element of the CCN2 promoter, and dominant negative Ets-1 and specific Ets-1 small interfering RNA block induction of CCN2 expression by TGFβ. In the absence of added TGFβ1, Ets-1, but not the related fli-1, synergizes with Smad 3 to activate the CCN2 promoter. Whereas the ability of transfected Ets-1 to activate the CCN2 promoter is dependent on protein kinase C (PKC), Ets-1 in the presence of co-transfected Smad3 does not require PKC, suggesting that the presence of Smad3 bypasses the requirement of Ets-1 for PKC to activate target promoter activity. Our results are consistent with the notion that Smad3 and Ets-1 cooperate in the induction of the CCN2 promoter by TGFβ1. Antagonizing Ets-1 might be of benefit in attenuating CCN2 expression in fibrosis, arthritis and cancer, and may be useful in modulating the outcome of these disorders
