    LPS Regulates SOCS2 Transcription in a Type I Interferon Dependent Autocrine-Paracrine Loop

    Recent studies suggest that SOCS2 is involved in the regulation of TLR signaling. In this study, we found that the expression of SOCS2 is regulated in human monocyte-derived DC by ligands stimulating TLR2, 3, 4, 5, 8 and 9 signaling. SOCS2 induction by LPS was dependent on the type I IFN regulated transcription factors IRF1 and IRF3 as shown by using silencing RNAs for IRFs. Blocking endogenous type I IFN signaling, by neutralizing antibodies to the receptor IFNAR2, abolished SOCS2 mRNA expression after TLR4 stimulation. Transcription factors STAT3, 5 and 6 displayed putative binding sites in the promoter regions of the human SOCS2 gene. Subsequent silencing experiments further supported that STAT3 and STAT5 are involved in LPS induced SOCS2 regulation. In mice we show that SOCS2 mRNA induction is 45% lower in bone marrow derived macrophages derived from MyD88−/− mice, and do not increase in BMMs from IRF3−/− mice after BCG infection. In conclusion, our results suggest that TLR4 signaling indirectly increases SOCS2 in late phase mainly via the production of endogenous type I IFN, and that subsequent IFN receptor signaling activates SOCS2 via STAT3 and STAT5

    GPS Multipath Analysis Using Fresnel Zones

    GNSS (Global Navigation Satellite Systems) multipath has been subject to scientific research for decades and although numerous methods and techniques have already been developed to mitigate this effect, it is still one of the accuracy-limiting factors in many GNSS applications. Since multipath is highly dependent on the individual antenna environment, there is still a need for new methods and further investigations to increase the understanding of this systematic effect. In this paper, the concept of Fresnel zones is applied to two different aspects of multipath. First, Fresnel zones are determined for the line-of-sight transmission between satellite and receiver. By comparing the boundary of the Fresnel zones to an obstruction adaptive elevation mask, potentially diffracted signals can be identified and excluded from the position estimation process. Both the percentage of epochs with fixed ambiguities and the positioning accuracy can be increased by the proposed method. Second, Fresnel zones are used to analyze the multipath induced by a horizontal and spatially-limited reflector. The comparison of simulated and real signal-to-noise (SNR) observations reveals a relationship between the percentage of the overlap of the Fresnel zone and reflector and the occurrence of multipath. It is found that an overlap of 50% is sufficient to induce multipath effects. This is of special interest, since this does not confirm theoretical assumptions of the multipath theory

    Increasing Spatio-Temporal Resolution for Monitoring Alpine Solifluction Using Terrestrial Laser Scanners and 3D Vector Fields

    This article investigates the usage of terrestrial laser scanner (TLS) point clouds for monitoring the gradual movements of soil masses due to freeze–thaw activity and water saturation, commonly referred to as solifluction. Solifluction is a geomorphic process which is characteristic for hillslopes in (high-)mountain areas, primarily alpine periglacial areas and the arctic. The movement can reach millimetre-to-centimetre per year velocities, remaining well below the typical displacement mangitudes of other frequently monitored natural objects, such as landslides and glaciers. Hence, a better understanding of solifluction processes requires increased spatial and temporal resolution with relatively high measurement accuracy. To that end, we developed a workflow for TLS point cloud processing, providing a 3D vector field that can capture soil mass displacement due to solifluction with high fidelity. This is based on the common image-processing techniques of feature detection and tracking. The developed workflow is tested on a study area placed in Hohe Tauern range of the Austrian Alps with a prominent assemblage of solifluction lobes. The derived displacements were compared with the established geomonitoring approach with total station and signalized markers and point cloud deformation monitoring approaches. The comparison indicated that the achieved results were in the same accuracy range as the established methods, with an advantage of notably higher spatial resolution.     Absence of Immunoglobulin Class Switch in Primary Lymphomas of the Central Nervous System

    Primary lymphomas of the central nervous system (PCNSLs) were investigated for their capacity to perform further maturation steps. We studied a series of 11 PCNSLs derived from immunocompetent patients for immunoglobulin (Ig) class switch recombination (CSR) by performing reverse transcriptase-polymerase chain reaction (RT-PCR) for transcripts of Ig constant region gene segments (IGHC). This analysis revealed exclusive transcription of IgM and IgD mRNA in the absence of IgG, IgA, or IgE transcription. This finding was corroborated at the protein level by the immunohistochemical demonstration of IgM on the surface of the tumor cells. The unexpected lack of CSR may be due to internal switch Ό region deletions, which were detected in 7 of 11 cases. We also found that expression of activation-induced cytidine deaminase (AID), which is required for CSR and somatic hypermutation, was detectable by RT-PCR in 4 of 10 cases and by immunohistochemistry in one of three cases analyzed. This may indicate that ongoing somatic mutation, which is often observed in PCNSL, could be due to sustained AID expression in a fraction of cases and that intraclonal V gene diversity may occur in other cases at an earlier phase of tumor clone expansion, when AID may have been expressed

    Multiplex qPCR assay for simultaneous quantification of CYP51‐S524T and SdhC‐H152R substitutions in European populations of Zymoseptoria tritici

    Demethylation inhibitor (DMI) and succinate dehydrogenase inhibitor (SDHI) fungicides are currently relied upon for the control of septoria tritici blotch (STB) in European wheat fields. However, multiple mutations have occurred over time in the genes encoding the targeted proteins that have led to a practical loss of fungicide efficacies. Among the different amino acid substitutions in Zymoseptoria tritici associated with resistance to these fungicides, S524T in CYP51 (DMI target) and H152R in SdhC (SDHI target) are regarded as conferring the highest resistance factors to DMI and SDHI, respectively. To facilitate further studies on the monitoring and selection of these substitutions in Z. tritici populations, a multiplex allele-specific quantitative PCR (qPCR) assay allowing for estimation of both allele frequencies in bulk DNA matrices was developed. The assay was then used on complex DNA samples originating from a spore trap network set up in Belgium, Denmark, Sweden, and Ireland in 2017 and 2018, as well as on leaf samples with symptoms. The S524T allele was present in all field samples and its proportion was significantly higher in Ireland than in Belgium, whereas the proportion of H152R was only sporadically present in both countries. The frequency of S524T varied greatly in the airborne inoculum of all four countries; however, the H152R allele was never detected in the airborne inoculum. The method developed in this study can be readily adopted by other laboratories and used for multiple applications including resistance monitoring in field populations of Z. tritici
