    Report on the September 2011 Meeting of the Next Generation Safegaurds Professional Network

    The Next Generation Safeguards Professional Network (NGSPN) was established in 2009 by Oak Ridge National Laboratory targeted towards the engagement of young professionals employed in safeguards across the many national laboratories. NGSPN focuses on providing a mechanism for young safeguards professionals to connect and foster professional relationships, facilitating knowledge transfer between current safeguards experts and the next generation of experts, and acting as an entity to represent the interests of the international community of young and mid-career safeguards professionals. This is accomplished in part with a yearly meeting held at a national laboratory site. In 2011, this meeting was held at Pacific Northwest National Laboratory. This report documents the events and results of that meeting

    Increasing altruistic and cooperative behaviour with simple moral nudges

    The conflict between pro-self and pro-social behaviour is at the core of many key problems of our time, as, for example, the reduction of air pollution and the redistribution of scarce resources. For the well-being of our societies, it is thus crucial to find mechanisms to promote pro-social choices over egoistic ones. Particularly important, because cheap and easy to implement, are those mechanisms that can change people's behaviour without forbidding any options or significantly changing their economic incentives, the so-called "nudges". Previous research has found that moral nudges (e.g., making norms salient) can promote pro-social behaviour. However, little is known about whether their effect persists over time and spills across context. This question is key in light of research showing that pro-social actions are often followed by selfish actions, thus suggesting that some moral manipulations may backfire. Here we present a class of simple moral nudges that have a great positive impact on pro-sociality. In Studies 1-4 (total N = 1,400), we use economic games to demonstrate that asking subjects to self-report "what they think is the morally right thing to do" does not only increase pro-sociality in the choice immediately after, but also in subsequent choices, and even when the social context changes. In Study 5, we explore whether moral nudges promote charity donations to humanitarian organisations in a large (N = 1,800) crowdfunding campaign.     Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival

    Six new host records and an updated list of wild hosts for Neobenedenia melleni (MacCallum) (Monogenea: Capsalidae) are provided. We report specimens of N. melleni from the skin of a whitefin sharksucker (Echeneis neucratoides Zuieuw [Echeneidae]) caught off MayagĂĽez, Puerto Rico; from the skin of a mosquitofish (Gambusia xanthosoma Greenfield [Poeciliidae)) caught in Little Salt Creek, Grand Cayman Island, British West Indies; from a freshwater immersion bath of red grouper (Epinephelus morio (Valenciennes) [Serranidae] caught in the Gulf of Mexico off Sarasota, Florida, U.S.A.; from the skin of a garden eel (Heteroconger hassi (Klausewitz and Eibl-Eibesfeldt) [Congridae]) in the Toledo Zoo, Toledo, Ohio, U.S.A.; from the skin of a raccoon butterftyfish (Chaetodon lunula (Cuvier) [Chaetodontidae]) in the Fort Wayne Children\u27s Zoo, Fort Wayne, Indiana, U.S.A.; and from the gill cavity of a red snapper (Lutjanus campechanus (Poey) [Lutjanidae]) in holding facilities at the Gulf Coast Research Laboratory, Ocean Springs, Mississippi, U.S.A. Neobenedenia melleni had not been reported previously from a suspected wild host in the Gulf of Mexico (i.e., E. morio) or from a member of Echeneidae, Atheriniformes, or Anguilliformes. Published host records indicate that N. melleni exhibits a relatively low degree of host specificity among captive and wild hosts; in nature, N. melleni infests predominantly shallow-water or reef teleosts
