Characteristic value determination from small samples
The paper deals with the characteristic value determination from relatively small samples. When the distribution and its parameters of a random variable are known, the characteristic value is deterministic quantity. However, in practical problems the parameters of distribution are unknown and can only be estimated from random samples. Therefore the characteristic value is by itself a random variable. The estimates of characteristic values are strongly dependant on the distribution of random variable. In the paper we show the analytical solution for characteristic value determination from random samples of normal and lognormal random variables. The confirmation of analytical results is accomplished by the use of computer simulations. For Gumbel, and Weibull distribution the characteristic value estimates are obtained numerically by combination of simulations and bisection method. In the paper the numerical results are presented for 5% characteristic values with 75% confidence interval, which is in accord with the majority of European building standards. The proposed approach is demonstrated on the data of experimentally obtained bending strengths of finger jointed wooden beams. (C) 2006 Elsevier Ltd. All rights reserved
Optimization-based modeling of Lombard speech articulation:Supraglottal characteristics
This paper shows that a highly simplified model of speech production based on the optimization of articulatory effort versus intelligibility can account for some observed articulatory consequences of signal-to-noise ratio. Simulations of static vowels in the presence of various background noise levels show that the model predicts articulatory and acoustic modifications of the type observed in Lombard speech. These features were obtained only when the constraint applied to articulatory effort decreases as the level of background noise increases. These results support the hypothesis that Lombard speech is listener oriented and speakers adapt their articulation in noisy environments.</p
Optimal control of speech with context-dependent articulatory targets
This paper presents a computational implementation of phonetic planning which consists of choosing the position of articulatory targets which satisfy conflicting linguistic and extra-linguistic requirements. We present a minimal model that considers intelligibility and least effort as task requirements. To achieve the context-dependent variability of targets, our model approximates intelligibility as a function of target phoneme recognition probability given a vector of articulatory parameters. Preliminary experiments show that our minimal computational model of phonetic planning is able to predict two types of hypoarticulation by adjusting the weight assigned to effort: vowel centralization and stop consonant lenition.Peer reviewe

Diverse diazotrophs are present on sinking particles in the North Pacific Subtropical Gyre.
Sinking particles transport carbon and nutrients from the surface ocean into the deep sea and are considered hot spots for bacterial diversity and activity. In the oligotrophic oceans, nitrogen (N2)-fixing organisms (diazotrophs) are an important source of new N but the extent to which these organisms are present and exported on sinking particles is not well known. Sinking particles were collected every 6âh over a 2-day period using net traps deployed at 150âm in the North Pacific Subtropical Gyre. The bacterial community and composition of diazotrophs associated with individual and bulk sinking particles was assessed using 16S rRNA and nifH gene amplicon sequencing. The bacterial community composition in bulk particles remained remarkably consistent throughout time and space while large variations of individually picked particles were observed. This difference suggests that unique biogeochemical conditions within individual particles may offer distinct ecological niches for specialized bacterial taxa. Compared to surrounding seawater, particle samples were enriched in different size classes of globally significant N2-fixing cyanobacteria including Trichodesmium, symbionts of diatoms, and the unicellular cyanobacteria Crocosphaera and UCYN-A. The particles also contained nifH gene sequences of diverse non-cyanobacterial diazotrophs suggesting that particles could be loci for N2 fixation by heterotrophic bacteria. The results demonstrate that diverse diazotrophs were present on particles and that new N may thereby be directly exported from surface waters on sinking particles
Identification of Exosite-Targeting Inhibitors of Anthrax Lethal Factor by High-Throughput Screening
SummaryProtease inhibitor discovery has focused almost exclusively on compounds that bind to the active site. Inhibitors targeting protease exosites, regions outside of the active site that influence catalysis, offer potential advantages of increased specificity but are difficult to systematically discover. Here, we describe an assay suitable for detecting exosite-targeting inhibitors of the metalloproteinase anthrax lethal factor (LF) based on cleavage of a full-length mitogen-activated protein kinase kinase (MKK) substrate. We used this assay to screen a small-molecule library and then subjected hits to a secondary screen to exclude compounds that efficiently blocked cleavage of a peptide substrate. We identified a compound that preferentially inhibited cleavage of MKKs compared with peptide substrates and could suppress LF-induced macrophage cytolysis. This approach should be generally applicable to the discovery of exosite-targeting inhibitors of many additional proteases
