This graduate recital in voice presents a wide scope of the soprano recital repertoire and explores the relationship between music and literature throughout the centuries. Mozart\u27s Bella mia fiamma… Resta, o cara, K. 528 represents the Italian Classical period; the text is based on the Roman myth of the goddess Ceres. Strauss\u27 Drei Lieder der Ophelia, Op. 67 represents German Lieder from the late-Romantic period; the set presents a German translation of Ophelia\u27s mad scene from Shakespeare\u27s Hamlet. Argento\u27s Six Elizabethan Songs represent 20th-century music, but Argento draws the text from various poets of the Elizabethan era. Selected songs by Franz Liszt are representative of the French mélodie from the Romantic period; they set the text of the Romantic poet Victor Hugo. Rachmaninoff\u27s Six Songs, Op. 38 represent the post-Romantic (nearly Impressionist) period and explore Russian Symbolist poetry

Geosynthetic landfill cap stability: comparison of limit equilibrium, computational limit analysis and finite-element analyses

The stability of the veneer cover soil (landfill cap) is an important issue in landfill design. Incorrect design of the landfill cap can lead to failure, which may result in the veneer cover soil sliding on an underlying geosynthetic layer, or in tension failure of the geosynthetic itself. Previous limit equilibrium (LE) analyses of veneer cover layer stability presented in the literature have generally considered whole-slope failure. In this paper, modified LE equations are proposed that (a) encompass more critical cases of localised slope failure for specific cases, and (b) are calibrated against two other methods: 2-D computational limit analysis (CLA) using LimitState:GEO and 2-D elasto-plastic finite-element (FE) analysis using PLAXIS. The scenarios examined encompass a cover of uniform thickness, a buttressed cover, a cover of tapered thickness, the effects of seepage forces, and the effects of construction equipment. It is shown that the LE method provides a reasonable estimate of veneer cover layer stability for most cases examined, although it is in general non-conservative, relative to the CLA and FE analyses. Local failure was found to be critical in the case of the construction equipment, buttress and horizontal seepage scenarios. In the latter case the LE equations previously presented in the literature significantly overestimate stability compared with the LE, CLA and FE analyses considered in this paper

The role of an extended medial column arthrodesis for Charcot midfoot neuroarthropathy

The etiology of diabetic Charcot neuroarthropathy involving the midfoot often includes an inciting traumatic event or repetitive micro-trauma from an uncompensated biomechanical imbalance that potentiates an incompletely understood pathway leading to a rocker-bottom foot deformity and ulceration. In the setting of a severe Charcot foot fracture and/or dislocation with obvious osseous instability, diagnostic delay can potentiate the limb-threatening sequelae of infected midfoot ulcerations in this patient population. In this article, the authors discuss the thought process as well as the advantages of performing an extended medial column arthrodesis for selected Charcot midfoot deformities

Application of Ni(II)-Assisted Peptide Bond Hydrolysis to Non-Enzymatic Affinity Tag Removal

In this study, we demonstrate a non-enzymatic method for hydrolytic peptide bond cleavage, applied to the removal of an affinity tag from a recombinant fusion protein, SPI2-SRHWAP-His6. This method is based on a highly specific Ni(II) reaction with (S/T)XHZ peptide sequences. It can be applied for the protein attached to an affinity column or to the unbound protein in solution. We studied the effect of pH, temperature and Ni(II) concentration on the efficacy of cleavage and developed an analytical protocol, which provides active protein with a 90% yield and ∼100% purity. The method works well in the presence of non-ionic detergents, DTT and GuHCl, therefore providing a viable alternative for currently used techniques

Determination of antihypertensive and anti-arrhythmic activity of new 5,5-dimethylhydantoin derivatives

Choroby układu krążenia stanowią jedną z wiodących chorób cywilizacyjnych XXI wieku, dlatego tak ważne stało się poznanie mechanizmów elektrofizjologicznych leżących u podstaw zaburzeń rytmu serca, ułatwiając tym samym wybór optymalnej metody leczenia umiarawiającego. Znajomość charakterystyki farmakologicznej, mechanizmów działania, a także objawów ubocznych leków antyarytmicznych przyczyniła się do dynamicznego rozwoju możliwości terapeutycznych w leczeniu zaburzeń rytmu serca.Celem niniejszej pracy było przebadanie nowych pochodnych 5,5-dimetylohydantoiny pod względem właściwości hipotensyjnych, elektrofizjologicznych oraz przeciwarytmicznych. Oceniono ich powinowactwo do receptorów α1-adrenergicznych, aktywność hipotensyjną, wpływ na prawidłowy elektrokardiogram szczurów oraz aktywność przeciwarytmiczną w modelu arytmii adrenalinowej. W celu porównania działania badanych struktur jako związku referencyjnego użyto dostępnego komercyjnie leku α1-adrenolitycznego – urapidylu. Badania farmakologiczne wykazały, że wszystkie pochodne 5,5-dimetylohydantoiny posiadają powinowactwo do receptorów α-adrenergicznych, przy czym najsilniej wypierał 3H-prazosynę w korze mózgowej szczura związek MG-15. Spośród z trzech przebadanych substancji w modelu arytmii adrenalinowej, związek MG-15 najsilniej zapobiegał przedwczesnym pobudzeniom komorowym i nadkomorowym, natomiast związki OJ-1 i OJ-8 nie chroniły zwierząt przed wystąpieniem zaburzeń poadrenalinowych - bradykardii, bigeminii, bloków przedsionkowo-komorowych, ekstrasystoli oraz nie wpływały na śmiertelność zwierząt. Oceniając wpływ arylopiperazynowych pochodnych hydantoiny na ciśnienie tętnicze u normotensyjnego szczura, po podaniu dożylnym, stwierdzono, że najsilniejsze działanie hipotensyjne (zbliżone do związku odniesienia) wykazywał związek MG-15, natomiast pochodne hydantoiny oznaczone symbolami OJ-1 oraz OJ-8 nie wykazywały efektu hipotensyjnego u normotensyjnych szczurów. Uzyskane wyniki badań farmakologicznych wskazują, że wśród 3 nowych arylopiperazynowych pochodnych hydantoiny, wyselekcjonowano 1 związek o działaniu hipotensyjnym i przeciwarytmicznym oraz wykazującym powinowactwo do receptorów α1-adrenergicznych. Mechanizm działania zarówno hipotensyjnego, jak i przeciwarytmicznego tego związku można wiązać z blokadą receptora α1-adrenergicznego.Diseases of the circulatory system are one of the leading civilization diseases of the 21st century, which is why it became so important to learn the electrophysiological mechanisms underlying the arrhythmias, thus facilitating the selection of the optimal method of normalizing treatment. Knowledge of pharmacological characteristics, mechanisms of action, as well as side effects of anti-arrhythmic drugs has contributed to the dynamic development of therapeutic options in the treatment of cardiac arrhythmias.The aim of this thesis was to test new derivatives of 5,5-dimethylhydantoin in terms of hypotensive, electrophysiological and antiarrhythmic properties. Their affinity for α-adrenergic receptors, hypotensive activity, effects on normal rat’s electrocardiogram and anti-arrhythmic activity in the adrenaline arrhythmia model were evaluated. In order to compare the effect of the examined structures as a reference compound, was used a commercially available α1-adrenolytic drug – urapidil.Pharmacological research have shown that all 5,5-dimethylhydantoin derivatives have affinity for α1-adrenergic receptors, with the strongest supersession of 3H-prazosin in the rat’s cerebral cortex of the MG-15 compound. From the three substances tested in the adrenaline arrhythmia model, the MG-15 compound was the most effective at preventing premature ventricular and supraventricular stimuli, while the OJ-1 and OJ-8 compounds did not protect animals from post-adrenaline disorders (bradycardia, bigeminia, atrio-ventricular blocks, extrasystoles) and did not affect animal mortality.Assessing the effect of arylpiperazine derivatives of hydantoin on blood pressure in a normotensive rat after intravenous administration, it was proved that the strongest hypotensive effect (similar to the reference compound) was exhibited by MG-15, while hydantoin derivatives designated OJ-1 and OJ-8 were not showed a hypotensive effect in normotensive rats.The obtained results of pharmacological studies indicate, that among the 3 new arylpiperazine derivatives of hydantoin, selected one compound with antihypertensive and anti-arrhythmic activity and with affinity for α1-adrenergic receptors. The mechanism of action of both hypotensive and antiarrhythmic activity of this compound can be associated with blockade of the α1-adrenergic receptor

(Mehrfach-) Adressierungen in politischen Talkshows

Der vorliegende Beitrag befasst sich mit der (Mehrfach-) Adressierung von Gesprächsbeiträgen in politischen Fernsehdiskussionen aus interaktiver Sicht. Ausgehend von einer Reflexion des Adressaten- bzw. Rezipientenbegriffs wird mit den Mitteln der Gesprächsanalyse untersucht, wie Diskutanten Adressierungen realisieren und inwiefern sich anhand dessen Mehrfachadressierungen herleiten lassen. Ausgangspunkt der Überlegungen ist das Hinterfragen zweier als gesichert geltender Annahmen: (1) Die Annahme, mediale Gespräche seien per se mehrfachadressiert, weil sie inszenierte Kommunikation sind, und (2) die Annahme, Mehrfachadressierung ließe sich auf die Teilnehmerkategorie (z. B. Politiker einer Partei X) reduzieren, statt den Kontext ,mediales Gesprächʻ relevant zu setzen, der für alle Beteiligten gilt

Guarantees of the protection of unborn life in Polish law and in the law and doctrine of the Catholic Church.

Praca dotyczy gwarancji ochrony życia poczętego w prawie polskim oraz w prawie i doktrynie Kościoła katolickiego. Pierwszy rozdział zawiera odniesienia zarówno do ustawodawstwa polskiego, jak i międzynarodowego. Rozdział drugi obrazuje problem definicji życia oraz aborcji, zaś w rozdziale trzecim opisano jak kształtowało się stanowisko wobec aborcji na przestrzeni wieków. Ostatni rozdział to stosunek Kościoła katolickiego wobec nienarodzonych.This thesis presents the guarantee of protection of an unborn life in the Polish law and the law of the Catholic Church. The first chapter includes references both to the Polish legislation and the international law. The second chapter illustrates the problem of the definition of life and abortion, while the third chapter presents how the stance against abortion has evolved over the centuries. The last chapter describes the position of the Catholic Church on the unborn children
