    Efectos de los procesos de ahumado sobre el valor nutricional y la composición de ácidos grasos de la lucioperca (Sander lucioperca)

    This study aimed to estimate the nutritional quality of Sander lucioperca fillets and alterations in lipid quality following hot and cold smoking processes. Our results revealed that the total fat content of zander fillets was 1.86 g/100g. Polyunsaturated fatty acids (PUFAs) were higher than saturated and monounsaturated fatty acids in fresh tissue samples. Arachidonic, docosahexaenoic and eicosapentaenoic acids were the most abundant PUFAs. Differences in the fatty acid compositions of the smoked and fresh fillet were significant, with a decrease in (PUFAs) in the proportion of total fatty acids. The fatty acid profile of the neutral lipids was unchanged after the cold smoking process, whereas PUFAs decreased significantly during both smoking processes, especially the hot smoking process. Our findings showed a partial alteration of polar lipids. Both smoking treatments produced lipoperoxidation and lipid oxidation in the fillets. Other investigational smoking conditions should be tested to reduce such oxidation and hydrolysis in fillets, which could be susceptible to off-flavors and off-odors.Este estudio tuvo como objetivo estimar la calidad nutricional de los filetes de lucioperca y las alteraciones en la calidad de los lípidos tras el proceso de ahumado en caliente y en frío. Nuestros resultados revelaron que el contenido total de grasa de los filetes de lucioperca fue de 1.86 g/100 g. Los ácidos grasos poliinsaturados (PUFA) fueron más altos que los ácidos grasos saturados y monoinsaturados en muestras de tejido fresco. Los ácidos grasos araquidónico, docosahexaenoico y eicosapentaenoico fueron los PUFA más abundantes. Las diferencias en la composición de ácidos grasos del filete ahumado y fresco fueron significativas, con una disminución de la proporción (PUFA) en los ácidos grasos totales. El perfil de ácidos grasos de los lípidos neutros se mantuvo sin cambios en el proceso de ahumado en frío, mientras que los PUFA disminuyeron significativamente durante ambos procesos de ahumado, especialmente en el proceso de ahumado en caliente. Nuestros resultados mostraron una alteración parcial de los lípidos polares. Ambos tratamientos de ahumado indicaron lipo-peroxidación y oxidación de lípidos en los filetes. Otras condiciones de ahumado se deben probar en investigación para reducir dicha oxidación e hidrólisis en los filetes, que podrían ser responsables de sabores y olores desagradables

    Two novel frame shift, recurrent and de novo mutations in the ITGB2 (CD18) gene causing leukocyte adhesion deficiency in a highly inbred North African population

    We have identified four different mutations causing leukocyte adhesion deficiency (LAD) in the ITGB2 gene of patients from a highly inbred population. Two were novel single-bp deletions (1497delG and 1920delG) causing frame shift and the two others were the missense mutations G284S and R593C. In our study, the G284S was a recurrent mutation while the R593C occurred de novo. We have also characterized a novel Xba1 polymorphic site located at the 5′ end of the ITGB2 locus. Family studies showed that the 1497delG mutation segregated with this marker and the intragenic AvaII polymorphic marker, suggesting the presence of a founder effect. The observation of a heterogeneous spectrum including de novo and recurrent mutations causing LAD in a highly inbred population is rather unexpected. In view of the literature published on the molecular genetics of LAD and considering the ethnic origin of the patients studied, our findings confirm the heterogeneity of the mutations causing LAD and point out potential mutational hot spots in the ITGB2 gene

    Composición bioquímica y potencial antioxidante del pepino del mar Mediterráneo comestible Holothuria tubulosa

    The sea cucumber or holothurian is a marine species which has been prized in some Asian coun­tries for its nutritional qualities. The purpose of this work was to study the biochemical composition and free radical scavenging and antioxidant activities of Holothuria tubulosa tegument from the Bizerta lagoon in north­ern Tunisia. The obtained data demonstrated that the extract of sea cucumber teguments exhibited high bio­chemical levels (such as moisture 80.77%, protein 7.07%, lipids 10.21%, energy value 13.64 Kcal/g ww), and an important nutritional value (including n-3/n-6: 2.11, EPA+DHA: 20.96, AI: 1.38 and TI: 0.54). High anti­oxidant activities were recorded in the integument by the radical scavenging tests of ABTS and DPPH as well as by the total antioxidant capacity and the FRAP in comparison with the BHT standard. Our results showed that H. Tubulosa tegument has high nutritional value with high antioxidant activities and could be considered a nutraceutical product.El pepino de mar o la holoturia es una especie marina apreciada en algunos países asiáti­cos por sus cualidades nutricionales. El propósito de este trabajo fue estudiar la composición bioquímica y las actividades antioxidantes y de eliminación de radicales libres del tegumento de Holothuria tubulosa de la laguna de Bizerta, en el norte de Túnez. Los datos obtenidos demuestran que el extracto de tegumentos de pepino de mar mostró altos niveles bioquímicos (como humedad 80,77%, proteína 7,07%, lípidos 10,21%, valor energético 13,64 Kcal/gww) y un valor nutricional importante (incluyendo n-3/ n-6: 2,11, EPA+DHA: 20,96, AI: 1,38 y TI: 0,54). Se registraron altas actividades antioxidantes en el tegumento mediante las pruebas de eliminación de radicales de ABTS y DPPH, así como por la capacidad antioxidante total y el FRAP, y esto, en comparación con el estándar BHT. Nuestros resultados mostraron que el tegumento de H. Tubulosa tiene un valor nutricional importante con una alta actividad antioxidante y podría considerarse un producto nutracéutico

    Losartan activates sirtuin 1 in rat reduced-size orthotopic liver transplantation

Aim: To investigate a possible association between losartan and sirtuin 1 (SIRT1) in reduced-size orthotopic liver transplantation (ROLT) in rats. Methods: Livers of male Sprague-Dawley rats (200-250 g) were preserved in University of Wisconsin preservation solution for 1 h at 4°C prior to ROLT. In an additional group, an antagonist of angiotensin II type 1 receptor (AT1R), losartan, was orally administered (5 mg/kg) 24 h and 1 h before the surgical procedure to both the donors and the recipients. Transaminase (as an indicator of liver injury), SIRT1 activity, and nicotinamide adenine dinucleotide (NAD+, a co-factor necessary for SIRT1 activity) levels were determined by biochemical methods. Protein expression of SIRT1, acetylated FoxO1 (ac-FoxO1), NAMPT (the precursor of NAD+), heat shock proteins (HSP70, HO-1) expression, endoplasmic reticulum stress (GRP78, IRE1α, p-eIF2) and apoptosis (caspase 12 and caspase 3) parameters were determined by Western blot. Possible alterations in protein expression of mitogen activated protein kinases (MAPK), such as p-p38 and p-ERK, were also evaluated. Furthermore, the SIRT3 protein expression and mRNA levels were examined. Results: The present study demonstrated that losartan administration led to diminished liver injury when compared to ROLT group, as evidenced by the significant decreases in alanine aminotransferase (358.3 133.44 vs 206 33.61, P + (0.87 0.22 vs 1.195 0.144, P < 0.05) the co-factor necessary for SIRT1 activity, as well as with decreases in ac-FoxO1 expression. Losartan treatment also provoked significant attenuation of endoplasmic reticulum stress parameters (GRP78, IRE1α, p-eIF2) which was consistent with reduced levels of both caspase 12 and caspase 3. Furthermore, losartan administration stimulated HSP70 protein expression and attenuated HO-1 expression. However, no changes were observed in protein or mRNA expression of SIRT3. Finally, the protein expression pattern of p-ERK and p-p38 were not altered upon losartan administration.     The effects of cold and hot smoking and the addition of&nbsp;Dunaliella salina&nbsp;polyphenol extract on the biochemical quality and shelf-life of&nbsp;Sander lucioperca&nbsp;fillets after storage for 90 days at 0-4 °C were examined. The results showed a significant increase in protein, lipid, free fatty acid, and 1,1-diphenyl-2-picrylhydrazyl contents, and a decrease in peroxide and thiobarbituric acid reactive substances, and volatile base nitrogen levels in cold (CSF) and hot (HSF) smoked fillets covered with or without extract and stored for 1, 20, and 90 days compared to fresh fillets (FF). Saturated and monounsaturated fatty acids exhibited a significant increase in FF and CSF and HSF covered with or without extract. The total polyunsaturated fatty acids revealed a significant decrease in FF and CSF and HSF with or without extract. Therefore, cold and hot smoking and polyphenol extract improved the biochemical quality and storage shelf-life of fillets for 90 days at 0-4 °C.Se examinaron los efectos del ahumado en frío y en caliente y la adición de extracto de polifenoles de&nbsp;Dunaliella salina&nbsp;sobre la calidad bioquímica y la vida útil de filetes de&nbsp;Sander lucioperca&nbsp;almacenados durante 90 días a 0-4 °C. Los resultados mostraron un aumento significativo en los contenidos de proteínas, lípidos, ácidos grasos libres y 1,1-difenil-2-picrilhidrazilo, y una disminución en las sustancias reactivas de peróxido y ácido tiobarbitúrico, y los niveles de nitrógeno básico volátil en frío (LCR) y caliente (HSF) de filetes ahumados cubiertos con o sin extracto y almacenados durante 1, 20 y 90 días en comparación con los filetes frescos (FF). Los ácidos grasos saturados y monoinsaturados exhibieron un aumento significativo en FF y LCR y HSF cubiertos con o sin extracto. Los ácidos grasos poliinsaturados totales revelaron una disminución significativa en FF y CSF y HSF con o sin extracto. Por lo tanto, el ahumado en frío y en caliente y el extracto de polifenoles mejoraron la calidad bioquímica y la vida útil durante el almacenamiento de los filetes durante 90 días a 0-4 °C

    A Multi-Key with Partially Homomorphic Encryption Scheme for Low-End Devices Ensuring Data Integrity †

    In today’s hyperconnected world, the Internet of Things and Cloud Computing complement each other in several areas. Cloud Computing provides IoT systems with an efficient and flexible environment that supports application requirements such as real-time control/monitoring, scalability, fault tolerance, and numerous security services. Hardware and software limitations of IoT devices can be mitigated using the massive on-demand cloud resources. However, IoT cloud-based solutions pose some security and privacy concerns, specifically when an untrusted cloud is used. This calls for strong encryption schemes that allow operations on data in an encrypted format without compromising the encryption. This paper presents an asymmetric multi-key and partially homomorphic encryption scheme. The scheme provides the addition operation by encrypting each decimal digit of the given integer number separately using a special key. In addition, data integrity processes are performed when an untrusted third party performs homomorphic operations on encrypted data. The proposed work considers the most widely known issues like the encrypted data size, slow operations at the hardware level, and high computing costs at the provider level. The size of generated ciphertext is almost equal to the size of the plaintext, and order-preserving is ensured using an asymmetrical encryption version

    Sirtuin 1 in rat orthotopic liver transplantation: An IGL-1 preservation solution approach

AIM: To investigate the possible involvement of Sirtuin 1 (SIRT1) in rat orthotopic liver transplantation (OLT), when Institute Georges Lopez 1 (IGL-1) preservation solution is enriched with trimetazidine (TMZ). METHODS: Male Sprague-Dawley rats were used as donors and recipients. Livers were stored in IGL-1 preservation solution for 8h at 4 °C, and then underwent OLT according to Kamada's cuff technique without arterialization. In another group, livers were stored in IGL-1 preservation solution supplemented with TMZ, at 10-6 mol/L, for 8 h at 4 °C and then underwent OLT. Rats were sacrificed 24 h after reperfusion, and liver and plasma samples were collected. Liver injury (transaminase levels), mitochondrial damage (glutamate dehydrogenase activity) oxidative stress (malondialdehyde levels), and nicotinamide adenine dinucleotide (NAD+), the co-factor necessary for SIRT1 activity, were determined by biochemical methods. SIRT1 and its substrates (ac-FoxO1, ac-p53), the precursor of NAD+, nicotinamide phosphoribosyltransferase (NAMPT), as well as the phosphorylation of adenosine monophosphate activated protein kinase (AMPK), p-mTOR, p-p70S6K (direct substrate of mTOR), autophagy parameters (beclin-1, LC3B) and MAP kinases (p-p38 and p-ERK) were determined by Western blot. RESULTS: Liver grafts preserved in IGL-1 solution enriched with TMZ presented reduced liver injury and mitochondrial damage compared with those preserved in IGL-1 solution alone. In addition, livers preserved in IGL-1 + TMZ presented reduced levels of oxidative stress. This was consistent with enhanced SIRT1 protein expression and elevated SIRT1 activity, as indicated by decreased acetylation of p53 and FoxO1. The elevated SIRT1 activity in presence of TMZ can be attributed to the enhanced NAMPT protein and NAD+/NADH levels. Up-regulation of SIRT1 was consistent with activation of AMPK and inhibition of phosphorylation of mTOR and its direct substrate (p-p70S6K). As a consequence, autophagy mediators (beclin-1 and LC3B) were overexpressed. Furthermore, MAP kinases were regulated in livers preserved with IGL-1 + TMZ, as they were characterized by enhanced p-ERK and decreased p-p38 protein expression.     ¿En qué medida la grasa de la merluza y la calidad de su aceite están influenciados por el parásito Lernaeocera lusci?

    The copepod Lernaeosera lusci is a common ectoparasite of the hake Merluccius merluccius, which constitutes its definitive host. The present study is the first to investigate the impact of L. lusci and its infection intensity on the fatty acid (FA) composition and oil quality of its host. The results revealed a substantial decrease in the total FA content, which dropped by 50, 70 and 83% when fish was infected by one (L1), two (L2) and three (L3) parasites, respectively. The lipid profile showed a tendency for a significant decrease in the saturated fatty acid (SFA), monounsaturated fatty acid (MUFA) and polyunsaturated fatty acid (PUFA) levels in all parasitized groups and in a parasite load dependant manner (p < 0.05). The same tendency was observed for the most commonly analysed FA and in particular for essential FA, such as e arachidonic (ARA), eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids, which showed smaller amounts associated with L. lusci co-infection. From a nutritional standpoint, our findings revealed that L. lusci is able to cause severe deterioration in the nutritional quality of its host oil. In addition to the increase in the thrombogenicity index (IT) and the high atherogenicity index (IA) values, the low hypocholesterolemic / hypercholesterolemic fatty acid ratio (H/H) and the modest amount of DHA+EPA (20 and 10 mg/100g) recorded at heavy infection levels make the parasitized fish inadequate sources of these PUFA and unsuitable for consumers with special dietary needs.El copépodo Lernaeosera lusci es un ectoparásito común de la merluza Merluccius merluccius que constituye su huésped definitivo. El presente estudio es el primero en investigar el impacto de L. lusci, considerando la intensidad de la infección, la composición de ácidos grasos (FA) y la calidad del aceite de su huésped. Los resultados mostraron una disminución sustancial en el contenido total de FA que se redujo en 50, 70 y 83% cuando el pez se infecta con uno (L1), dos (L2) y tres (L3) parásitos, respectivamente. El perfil lipídico mostró una tendencia a una disminución significativa en los niveles de ácidos grasos saturados (SFA), ácidos grasos monoinsaturados (MUFA) y ácidos grasos poliinsaturados (PUFA) en todos los grupos parasitados y de una manera dependiente de la carga parasitaria (p < 0.05). Se observó la misma tendencia para la mayoría de los FA y, en particular, para los FA esenciales como los ácidos araquidónico (ARA), eicosapentaenoico (EPA) y docosahexaenoico (DHA), que mostraron cantidades reducidas asociadas con la coinfección por L. lusci. Desde un punto de vista nutricional, nuestro descubrimiento reveló que L. lusci es capaz de causar un deterioro severo de la calidad nutricional de su aceite huésped. Además del aumento del índice de trombogenicidad (IT) y valores de índice de aterogenicidad (IA) alto, una relación hipocolesterolemia/hipercolesterolemia (H/H) baja y una cantidad modesta de DHA + EPA (20 y 10 mg)/100 g) registrados en infecciones graves hacen que los peces parasitados sean fuentes inadecuadas de PUFA y no sean adecuados para consumidores con necesidades dietéticas especiales
