    We report results of a study on processes of Organizational Learning (OL). Changes and suspensions of organizational rules are taken as indicators for OL. The empiric analysis is performed with personnel rules of a German bank. The aim of our study is to identify factors that have an impact on rule changes and suspensions and thereby on OL. These influence factors are, on the one hand, variables that reflect experiences of organizational members with rules and, on the other, variables which capture organizational and environmental shifts. An important finding of this study is that the processes of rule change and rule suspension follow quite different patterns. The changing of a rule is mainly influenced by experience variables. Two basic modes of experiential influences could be shown: habitualization and working one's way out of a failure trap. Experience variables also play a role in the process of rule suspension. The negative influence of rule age on the suspension process suggests that the habitualization of rules over time increases reluctance to abolish a rule. It could also be shown that the positive version age effect on the suspension rate, which indicated an obsolescence process during the existence of a rule version when no additional variables are controlled, is explained by ecological shifts. Organizational and environmental shifts hardly display any significant effects on the rate of rule change. On the other hand, these shifts represent distinct influences on rule suspension. When the environment of a rule changes the need for an abolishment of this rule rises. The attention towards rules which are no longer appropriate seems to be stimulated by these environmental and organizational shifts. These findings suggest that OL can consist of a type of learning which is rather independent of certain ecological triggers and it can also consist of a type of learning which is maintained in order to adapt to certain changes within and outside the o

    Virulence of malaria is associated with differential expression of Plasmodium falciparum var gene subgroups in a case-control study

    Plasmodium falciparum erythrocyte membrane protein 1 (PfEMP1) is a major pathogenicity factor in falciparum malaria that mediates cytoadherence. PfEMP1 is encoded by approximately 60 var genes per haploid genome. Most var genes are grouped into 3 subgroups: A, B, and C. Evidence is emerging that the specific expression of these subgroups has clinical significance. Using field samples from children from Papua New Guinea with severe, mild, and asymptomatic malaria, we compared proportions of transcripts of var groups, as determined by quantitative polymerase chain reaction. We found a significantly higher proportion of var group B transcripts in children with clinical malaria (mild and severe), whereas a large proportion of var group C transcripts was found in asymptomatic children. These data from naturally infected children clearly show that major differences exist in var gene expression between parasites causing clinical disease and those causing asymptomatic infections. Furthermore, parasites forming rosettes showed a significant up-regulation of var group A transcripts

    Short term cost effectiveness of a regional myocardial infarction network

    Aims: Myocardial infarction networks have been shown to improve guideline adherent therapy and outcomes in patients presenting with acute ST-elevation myocardial infarction (STEMI). Our objective was to assess the short term cost effectiveness of a network structure. Methods and results: Outcome data and reimbursement data for the index hospital stay were gathered in consecutive patients with acute STEMI (n=536) admitted to any of the hospitals in a 350.000 inhabitant rural network area during the years 2002 (n=185), 2005 (n=163) and 2008 (n=188). Network structure was established between 2002 and 2005 aiming for identical treatment of all acute STEMI patients during 24 h/7d a week with primary angioplasty. Patient baseline characteristics in the different years were quite comparable. From 2002 to 2005 regional hospital mortality in STEMI patients decreased from 16% to 9%. Lower mortality under network conditions was confirmed in 2008. Reimbursement data of different years were standardized to exclude effects not induced by the network. The mean initial costs per saved live during the index stay were 7727 with a 95%-confidence interval of -3.500 to 36.700 (referenced to the German reimbursement in 2005). Conclusion: The short term cost effectiveness of a myocardial infarction network organisation is within well accepted boundaries under conditions of the German reimbursement system

    Longitudinal Assessment of Plasmodium falciparum var Gene Transcription in Naturally Infected Asymptomatic Children in Papua New Guinea

    Sequestration and antigenic variation are essential for Plasmodium falciparum survival in vivo contributing to severe pathologic findings and, also, chronic infection. Both are conferred by P. falciparum erythrocyte membrane proteins encoded by ∼60 var genes. To study the dynamics of var gene expression, we conducted a 4-month longitudinal study of semi-immune children from Papua New Guinea. By use of magnetic bead- anchored reverse-transcription polymerase chain reaction analysis performed over 5 var regions, as well as cloning and sequencing, the longitudinal distribution of full-length var transcripts was analyzed. We identified a dynamic picture of var gene expression with rapid switches but with identical var transcripts recurring for up to 10 weeks. The number of var transcripts was correlated to the number of infections, with a mean of 1.7 var transcripts identified per sample and infecting strain. Analysis of 158 different Duffy binding-like 1α sequences confirmed the recombinogenic nature of var genes.     Novel Genotyping Tools for Investigating Transmission Dynamics of Plasmodium falciparum

    Background. Differentiation between gametocyte-producing Plasmodium falciparum clones depends on both high levels of stage-specific transcripts and high genetic diversity of the selected genotyping marker obtained by a high-resolution typing method. By analyzing consecutive samples of one host, the contribution of each infecting clone to transmission and the dynamics of gametocyte production in multiclone infections can be studied. Methods. We have evaluated capillary electrophoresis based differentiation of 6 length-polymorphic gametocyte genes. RNA and DNA of 25 µL whole blood from 46 individuals from Burkina Faso were simultaneously genotyped. Results. Highest discrimination power was achieved by pfs230 with 18 alleles, followed by pfg377 with 15 alleles. When assays were performed in parallel on RNA and DNA, 85.7% of all pfs230 samples and 59.5% of all pfg377 samples contained at least one matching genotype in DNA and RNA. Conclusions. The imperfect detection in both, DNA and RNA, was identified as major limitation for investigating transmission dynamics, owing primarily to the volume of blood processed and the incomplete representation of all clones in the sample tested. Abundant low-density gametocyte carriers impede clone detectability, which may be improved by analyzing larger volumes and detecting initially sequestered gametocyte clones in follow-up sample

    The European sea bass Dicentrarchus labrax genome puzzle: comparative BAC-mapping and low coverage shotgun sequencing

    Abstract Background Food supply from the ocean is constrained by the shortage of domesticated and selected fish. Development of genomic models of economically important fishes should assist with the removal of this bottleneck. European sea bass Dicentrarchus labrax L. (Moronidae, Perciformes, Teleostei) is one of the most important fishes in European marine aquaculture; growing genomic resources put it on its way to serve as an economic model. Results End sequencing of a sea bass genomic BAC-library enabled the comparative mapping of the sea bass genome using the three-spined stickleback Gasterosteus aculeatus genome as a reference. BAC-end sequences (102,690) were aligned to the stickleback genome. The number of mappable BACs was improved using a two-fold coverage WGS dataset of sea bass resulting in a comparative BAC-map covering 87% of stickleback chromosomes with 588 BAC-contigs. The minimum size of 83 contigs covering 50% of the reference was 1.2 Mbp; the largest BAC-contig comprised 8.86 Mbp. More than 22,000 BAC-clones aligned with both ends to the reference genome. Intra-chromosomal rearrangements between sea bass and stickleback were identified. Size distributions of mapped BACs were used to calculate that the genome of sea bass may be only 1.3 fold larger than the 460 Mbp stickleback genome. Conclusions The BAC map is used for sequencing single BACs or BAC-pools covering defined genomic entities by second generation sequencing technologies. Together with the WGS dataset it initiates a sea bass genome sequencing project. This will allow the quantification of polymorphisms through resequencing, which is important for selecting highly performing domesticated fish.Peer Reviewe
