    Tyrophagus putrescentiae predating adult insects of Aedes aegypti and Aedes albopictus in laboratory

    The present study aimed at identifying a mite infesting a colony of Aedes aegypti and Ae. albopictus as well as investigating the source of infestation. The mite species was identified after it was mounted on slides in Hoyer's medium and examined under an optical microscope. It showed to be Tyrophagus putrescentiae. Fish and rabbit food samples were kept in an environmental chamber at 27ºC and 90% humidity for 21 days and were weekly examined. The presence of T. putrescentiae was only observed in rabbit food. It is assumed that T. putrescentiae had been introduced into the colonies through phoresy among guinea pigs, which were kept at the time near the insectary under favorable conditions for their development (28.7ºC, 72% humidity, photoperiod of 14L:10D hours).O objetivo do estudo foi identificar ácaro infestando colônia de Aedes aegypti e Ae. albopictus, além da investigação de fonte de contaminação. A espécie de ácaro foi identificada pela montagem, em meio de Hoyer, entre lâmina e lamínula e observados em microscopia óptica, revelando tratar-se de Tyrophagus putrescentiae. Amostras de ração para peixe e de ração para coelho foram mantidas em estufa BOD a 27°C e 90% de umidade, por 21 dias, sendo examinadas semanalmente. A presença de T. putrescentiae foi somente observada em amostras de ração para coelho. Presume-se que T. putrescentiae tenha sido introduzido nas colônias pela forésia em cobaias guinea-pigs, encontrando em ambiente de insetário na ocasião do evento (28,7°C, 72% de umidade e fotoperíodo de 14L:10E horas) condições favoráveis para o seu desenvolvimento

    Demandas e perspectivas para a Acarologia no Brasil

    Scientific papers about research demands and perspectives on acarology in Brazil are scarce. This article aims to prospect and characterize former developed research, as well as identify actual and future demands on research in acarology in Brazil. To achieve these goals, a bibliographic review of papers published between 1996 and 2006 was done and the First Brazilian Symposium of Acarologia was organized. Studies about mites of medicalveterinarian and agricultural represent the majority of the reviewed papers, and the families Ixodidae, Tetranychidae and Phytoseiidae were the most studied. These studies were developed chiefly in the Brazilian Southeast region. Among the main research demands, especial attention was given to the training of taxonomists to identify new important agricultural and medical-veterinary species, the survey of new predatory mites species for biological control, the role of the mites in the nutrients cycling in ecosystems, and the mitigation of the negative impacts cause by synthetic acaricides. Nesse trabalho, objetivou-se prospectar e caracterizar os estudos em acarologia já realizados e identificar as principais demandas presentes e futuras em pesquisas em acarologia no Brasil. Esses estudos foram realizados, principalmente, na região Sudeste do Brasil. Dentre as principais demandas levantadas, destacaram-se o treinamento de taxonomistas para identificação de espécies de importância agrícola e médico-veterinária, o levantamentos de novas espécies de ácaros predadores para emprego no controle biológico aplicado, o estudo do papel dos ácaros na ciclagem de nutrientes em ecossistemas e a mitigação dos impactos negativos advindos do uso de acaricidas sintéticos.     Ornithodoros faccinii n. sp. (Acari: Ixodida: Argasidae) parasitizing the frog Thoropa miliaris (Amphibia: Anura: Cycloramphidae) in Brazil

    Abstract\ud \ud Background\ud Most argasid ticks from the Neotropical region are parasites of mammals and birds, with a few records from reptiles. Many species of the genus Ornithodoros are known only through larval descriptions, and their chaetotaxy and morphological characteristics have been used to separate the taxa. In the present study, we describe the larva and the nymph of first instar of a new species of the genus Ornithodoros that was collected from frogs of the species Thoropa miliaris.\ud \ud \ud Methods\ud Larvae of Ornithodoros were collected from frogs of the species T. miliaris at waterfalls in the state of Rio de Janeiro, southeastern Brazil. The larval and nymphal description was based on optical and scanning electron microscopy. Molecular analysis using the argasid 16S rRNA sequences available in GenBank was also conducted.\ud \ud \ud Results\ud \ud Ornithodoros faccinii sp. n. is closely related to Ornithodoros clarki Jones & Clifford, Ornithodoros marinkellei Kohls, Clifford & Jones, Ornithodoros capensis Neumann and Ornithodoros sawaii Kitaoka & Susuki. However, the larval morphology of the new species is unique. The mitochondrial 16S rDNA partial sequence of O. faccinii generated in the present study was deposited in GenBank under the number KP861242.\ud \ud \ud Conclusions\ud The larvae collected from Thoropa miliaris are a new species, Ornithodoros faccinii n. sp. This is the first report of argasid ticks on frogs in Brazil, the second on frogs and the third on Amphibia in the Neotropical region

    Análise do microbioma procarionte de fêmeas e cultura celular embrionária de Rhipicephalus sanguineus linhagens tropical e temperada de duas localidades no Brasil

    Two lineages of Rhipicephalus sanguineus are known in Brazil: the temperate or southern and the tropical or northern populations. The distribution patterns of both lineages of R. sanguineus have epidemiological implications that can affect vectorial competence concerning Ehrlichia canis, the agent of canine monocytic ehrlichiosis. Intending to identify the microbiomes of both lineages and compare microorganisms in R. sanguineus, we used the 16S rRNA (V4-V5 region) gene-based metataxonomic approach, through NGS sequencing on the MiSeq Illumina platform. We selected specimens of females from the environment and samples of primary embryonic cell cultures, from both lineages, and this was the first study to investigate the prokaryotic microbiome in tick cell cultures. The results showed that many bacterial taxa detected in the samples were typical members of the host environment. A significant diversity of microorganisms in R. sanguineus females and in embryonic cell cultures from both lineages was found, with emphasis on the presence of Coxiella in all samples, albeit in different proportions. The Coxiella species present in the two lineages of ticks may be different and may have co-evolved with them, thus driving different patterns of interactions between ticks and the pathogens that they can harbor or transmit to vertebrate hosts.Duas linhagens de Rhipicephalus sanguineus são conhecidas no Brasil: populações da linhagem temperada ou do sul, e tropical ou do norte. Os padrões de distribuição de ambas as linhagens de R. sanguineus têm implicações epidemiológicas, podendo afetar a competência vetorial de Ehrlichia canis, o agente etiológico da erliquiose monocítica canina. Com a intenção de identificar os microbiomas de ambas as linhagens e comparar microrganismos de R. sanguineus, foi utilizada a metataxonomia, baseada no gene 16S rRNA (região V4-V5), por meio do sequenciamento de nova geração na plataforma MiSeq Illumina. Foram selecionadas amostras de fêmeas do ambiente e cultivo primário de células embrionárias, considerando-se as duas linhagens conhecidas do Brasil. Este é o primeiro estudo que investiga o microbioma procariótico de células de cultura de carrapato. Os resultados mostram que muitos grupos de bactérias detectadas nas amostras são membros típicos do ambiente do hospedeiro. Uma diversidade significativa de microrganismos em fêmeas e cultura de células embrionárias nas duas linhagens de R. sanguineus foi encontrada, com ênfase na presença de Coxiella em todas as amostras, ainda que em diferentes proporções. Possivelmente, as espécies de Coxiella presentes nas duas linhagens de carrapatos são diferentes e co-evoluíram com essas linhagens, conduzindo a diferentes padrões de interação entre carrapatos e patógenos que podem abrigar ou transmitir aos hospedeiros vertebrados
