    Astrocytes regulate brain extracellular pH via a neuronal activity-dependent bicarbonate shuttle

    Brain cells continuously produce and release protons into the extracellular space, with the rate of acid production corresponding to the levels of neuronal activity and metabolism. Efficient buffering and removal of excess H+ is essential for brain function, not least because all the electrogenic and biochemical machinery of synaptic transmission is highly sensitive to changes in pH. Here, we describe an astroglial mechanism that contributes to the protection of the brain milieu from acidification. In vivo and in vitro experiments conducted in rodent models show that at least one third of all astrocytes release bicarbonate to buffer extracellular H+ loads associated with increases in neuronal activity. The underlying signalling mechanism involves activity-dependent release of ATP triggering bicarbonate secretion by astrocytes via activation of metabotropic P2Y1 receptors, recruitment of phospholipase C, release of Ca2+ from the internal stores, and facilitated outward HCO3− transport by the electrogenic sodium bicarbonate cotransporter 1, NBCe1. These results show that astrocytes maintain local brain extracellular pH homeostasis via a neuronal activity-dependent release of bicarbonate. The data provide evidence of another important metabolic housekeeping function of these glial cells

    Health-related quality of life of children born very preterm: a multinational European cohort study

    Purpose This study aims to (1) describe the health-related quality of life (HRQoL) outcomes experienced by children born very preterm (28–31 weeks’ gestation) and extremely preterm (< 28 weeks’ gestation) at five years of age and (2) explore the mediation effects of bronchopulmonary dysplasia (BPD) and severe non-respiratory neonatal morbidity on those outcomes. Methods This investigation was based on data for 3687 children born at < 32 weeks’ gestation that contributed to the EPICE and SHIPS studies conducted in 19 regions across 11 European countries. Descriptive statistics and multi-level ordinary linear squares (OLS) regression were used to explore the association between perinatal and sociodemographic characteristics and PedsQL™ GCS scores. A mediation analysis that applied generalised structural equation modelling explored the association between potential mediators and PedsQL™ GCS scores. Results The multi-level OLS regression (fully adjusted model) revealed that birth at < 26 weeks’ gestation, BPD status and experience of severe non-respiratory morbidity were associated with mean decrements in the total PedsQL™ GCS score of 0.35, 3.71 and 5.87, respectively. The mediation analysis revealed that the indirect effects of BPD and severe non-respiratory morbidity on the total PedsQL™ GCS score translated into decrements of 1.73 and 17.56, respectively, at < 26 weeks’ gestation; 0.99 and 10.95, respectively, at 26–27 weeks’ gestation; and 0.34 and 4.80, respectively, at 28–29 weeks’ gestation (referent: birth at 30–31 weeks’ gestation). Conclusion The findings suggest that HRQoL is particularly impaired by extremely preterm birth and the concomitant complications of preterm birth such as BPD and severe non-respiratory morbidity.This study was supported by a grant from the European Union’s Horizon 2020 Research and Innovation Programme under grant agreement No 633724.     CO2 signaling mediates neurovascular coupling in the cerebral cortex

    Neurovascular coupling is a fundamental brain mechanism that regulates local cerebral blood flow (CBF) in response to changes in neuronal activity. Functional imaging techniques are commonly used to record these changes in CBF as a proxy of neuronal activity to study the human brain. However, the mechanisms of neurovascular coupling remain incompletely understood. Here we show in experimental animal models (laboratory rats and mice) that the neuronal activity-dependent increases in local CBF in the somatosensory cortex are prevented by saturation of the CO2-sensitive vasodilatory brain mechanism with surplus of exogenous CO2 or disruption of brain CO2/HCO3− transport by genetic knockdown of electrogenic sodium-bicarbonate cotransporter 1 (NBCe1) expression in astrocytes. A systematic review of the literature data shows that CO2 and increased neuronal activity recruit the same vasodilatory signaling pathways. These results and analysis suggest that CO2 mediates signaling between neurons and the cerebral vasculature to regulate brain blood flow in accord with changes in the neuronal activity

    In Vitro Evaluation of the Antiviral Potential of Guettarda angelica Against Animal Herpesviruses

    Background: The number of antiviral studies using plant extracts has increased in the last decades, and the results have shown that plants are potential sources of compounds that are able to inhibit and/or decrease viral infections. The selection of these plants by ethnopharmacological criteria increases the probability of finding new substances with significant pharmacological and biological activities. Hence, Brazil has an advantage in this area due to its extensive biodiversity and ethnological diversity. Guettarda angelica is a plant from the Brazilian Caatinga region the roots of which are popularly used for various therapeutic purposes, including veterinary use. The aim of this work was to investigate the in vitro antiviral activity of extracts of plant parts from G. angelica against three animal herpesviruses: bovine (BoHV-1), suid (SuHV-1) and equine (EHV-1) herpesviruses type 1. Materials, Methods & Results: The extracts of roots, leaves and seeds of G. angelica were initially screened for in vitro antiviral activity against these herpesviruses using the virus yield reduction assay. The MDBK cells were used in assays with BoHV-1 and SuHV-1, and the Vero cells with EHV-1. For these assays, the cells previously treated with the extracts in non-cytotoxic concentrations were inoculated with logarithmic dilutions of each virus, The viral inhibitory activity of extracts was calculated by difference of virus titer between treated infected cells and non-treated infected cells. Only the aqueous extract from seeds (AEs) showed a significant antiviral activity (P < 0.01, ANOVA followed by Tukey test) against all herpesviruses leading continuous studies, Thus, the selectivity index (SI) of this extract was determined by MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] colorimetric assay by calculating the ratio CC(50)over IC50. The 50% cytotoxic concentration (CC50) was defined as the extract concentration that reduced the cell viability by 50% when compared to untreated controls; the 50% inhibitory concentration (IC50) was defined as the concentration of the extract that inhibited 50% of viral replication when compared to the virus control. The CC50 and IC50 obtained from nonlinear regression analysis of concentration-effect curves by the GraphPad Prism 5 Demo program and represented the means +/- standard deviation of three independent experiments. The CC5(0) for Vero cells was 400.60 +/- 0.20 mu g/mL, while the CC50 for MDBK cells was 920.50 +/- 0.19 mu g/mL. The IC50 values of the AEs on the BoHV-1, SuHV-1 and EHV-1 were 22.79 mu g/mL, 91.30 mu g/mL and 19.95 mu g/mL, respectively. The SI values of this extract for each virus obtained from these data were 40.39, 10.08 and 20.08 for BoHV-1, SuHV-1, and EHV-1, respectively. Discussion: To ensure the antiviral activity of a plant extract and consequently its future use as antiviral agent is crucial the obtainment of its selectivity index or safety index. It is guarantee of a true antiviral effect and not the result of cytotoxicity of the extract on cells, and that could be confused with an antiviral activity. Other important point are the extract IC50 values less than 100 mu g/mL. The results of the AEs of G. angelica are in accordance with these considerations indicating that the Go angelica seeds may be a potential source of antiviral compounds insurance and encouraging further investigation of them.40
