Transepithelial Migration of Toxoplasma gondii Is Linked to Parasite Motility and Virulence

    Get PDF
After oral ingestion, Toxoplasma gondii crosses the intestinal epithelium, disseminates into the deep tissues, and traverses biological barriers such as the placenta and the blood-brain barrier to reach sites where it causes severe pathology. To examine the cellular basis of these processes, migration of T. gondii was studied in vitro using polarized host cell monolayers and extracellular matrix. Transmigration required active parasite motility and the highly virulent type I strains consistently exhibited a superior migratory capacity than the nonvirulent type II and type III strains. Type I strain parasites also demonstrated a greater capacity for transmigration across mouse intestine ex vivo, and directly penetrated into the lamina propria and vascular endothelium. A subpopulation of virulent type I parasites exhibited a long distance migration (LDM) phenotype in vitro, that was not expressed by nonvirulent type II and type III strains. Cloning of parasites expressing the LDM phenotype resulted in substantial increase of migratory capacity in vitro and in vivo. The potential to up-regulate migratory capacity in T. gondii likely plays an important role in establishing new infections and in dissemination upon reactivation of chronic infections

Comparing Methods of Detecting Mind Wandering While Driving

Driver distraction is a persistent threat to traffic safety. External distraction has been examined extensively, but few studies have focused on internal distraction such as mind wandering. Equivocal results from the few existing studies are likely due, at least in part, to different experimental methods. Mind wandering is commonly assessed using either a self-caught or probe-caught method. The current investigation sought to better understand the effects of mind wandering on driving performance using the self-caught method and the probecaught method. In the Self-Caught Experiment, lateral control measures such as, lateral position variability and steering reversal rate were greater when drivers reported on-task thoughts versus mind wandering. In the Probe-Caught Experiment, these results were not replicated using the traditional probe-caught analysis. Instead, when analyzing the results of the Probe-Caught Experiment in a similar manner as the Self-Caught Experiment, the results were replicated. These results highlight methodological concerns in detecting mind wandering while driving. Additional research is needed to determine which method should be employed in future studies

    Habitability: CAMELOT 4

    Get PDF
During 1988 to 1989 the NASA/USRA Advanced Design Program sponsored research and design efforts aimed at developing habitability criteria and at defining a habitability concept as a useful tool in understanding and evaluating dwellings for prolonged stays in extraterrestrial space. The Circulating Auto sufficient Mars-Earth Luxurious Orbital Transport (CAMELOT) was studied as a case in which the students would try to enhance the quality of life of the inhabitants by applying architectural design methodology. The study proposed 14 habitability criteria considered necessary to fulfill the defined habitability concept, which is that state of equilibrium that results from the interaction between components of the Individual Architecture Mission Complex, which allows a person to sustain physiological homeostatis, adequate performance, and acceptable social relationships. Architecture, design development, refinements and revisions to improve the quality of life, new insights on artificial gravity, form and constitution problems, and the final design concept are covered

    Non-singular Universes a la Palatini

    Get PDF
It has recently been shown that f(R) theories formulated in the Palatini variational formalism are able to avoid the big bang singularity yielding instead a bouncing solution. The mechanism responsible for this behavior is similar to that observed in the effective dynamics of loop quantum cosmology and an f(R) theory exactly reproducing that dynamics has been found. I will show here that considering more general actions, with quadratic contributions of the Ricci tensor, results in a much richer phenomenology that yields bouncing solutions even in anisotropic (Bianchi I) scenarios. Some implications of these results are discussed.Comment: 4 pages, no figures. Contribution to the Spanish Relativity Meeting (ERE2010), 6-10 Sept. Granada, Spai

Peculiar outburst of A 0535+26 observed with INTEGRAL, RXTE and Suzaku

A normal outburst of the Be/X-ray binary system A0535+26 has taken place in August 2009. It is the fourth in a series of normal outbursts that have occured around the periastron passage of the source, but is unusual by starting at an earlier orbital phase and by presenting a peculiar double-peaked light curve. A first "flare" (lasting about 9 days from MJD 55043 on) reached a flux of 440 mCrab. The flux then decreased to less than 220 mCrab, and increased again reaching 440 mCrab around the periastron at MJD 55057. Target of Opportunity observations have been performed with INTEGRAL, RXTE and Suzaku. First results of these observations are presented, with special emphasis on the cyclotron lines present in the X-ray spectrum of the source, as well as in the pulse period and energy dependent pulse profiles of the source.Comment: 6 pages, Accepted for publication on PoS, Proceedings of "The Extreme sky: Sampling the Universe above 10 keV", held in Otranto (Italy) in October 200

Obtencion de aleaciones al-mg de morfologia porosa

    Get PDF

Chondroinductive Hydrogel Pastes Composed of Naturally Derived Devitalized Cartilage

Hydrogel precursors are liquid solutions that are prone to leaking from the defect site once implanted in vivo. Therefore, the objective of the current study was to create a hydrogel precursor that exhibited a yield stress. Additionally, devitalized cartilage extracellular matrix (DVC) was mixed with DVC that had been solubilized and methacrylated (MeSDVC) to create hydrogels that were chondroinductive. Precursors composed of 10% MeSDVC or 10% MeSDVC with 10% DVC were first evaluated rheologically, where non-Newtonian behavior was observed in all hydrogel precursors. Rat bone marrow stem cells (rBMSCs) were mixed in the precursor solutions, and the solutions were then crosslinked and cultured in vitro for 6 weeks with and without exposure to human transforming growth factor β3 (TGF-β3). The compressive modulus, gene expression, biochemical content, swelling, and histology of the gels were analyzed. The DVC-containing gels consistently outperformed the MeSDVC-only group in chondrogenic gene expression, especially at 6 weeks, where the relative collagen II expression of the DVC-containing groups with and without TGF-β3 exposure was 40- and 78-fold higher, respectively, than that of MeSDVC alone. Future work will test for chondrogenesis in vivo and overall, these two cartilage-derived components are promising materials for cartilage tissue engineering applications
