The Massive Progenitor of the Possible Type II-Linear Supernova 2009hd in Messier 66
We present observations of SN2009hd in the nearby galaxy M66. This SN is one
of the closest to us in recent years but heavily obscured by dust, rendering it
unusually faint in the optical, given its proximity. We find that the observed
properties of SN2009hd support its classification as a possible Type II-L SN, a
relatively rare subclass of CC-SNe. High-precision relative astrometry has been
employed to attempt to identify a SN progenitor candidate, based on a
pixel-by-pixel comparison between HST F555W and F814W images of the SN site
prior to explosion and at late times. A progenitor candidate is identified in
the F814W images only; this object is undetected in F555W. Significant
uncertainty exists in the astrometry, such that we cannot definitively identify
this object as the SN progenitor. Via insertion of artificial stars into the
pre-SN HST images, we are able to constrain the progenitor's properties to
those of a possible supergiant, with M(F555W)0>-7.6 mag and (V-I) 0>0.99 mag.
The magnitude and color limits are consistent with a luminous RSG; however,
they also allow for the possibility that the star could have been more yellow
than red. From a comparison with theoretical massive-star evolutionary tracks,
which include rotation and pulsationally enhanced mass loss, we can place a
conservative upper limit on the initial mass for the progenitor of <20 M_sun.
If the actual mass of the progenitor is near the upper range allowed by our
derived mass limit, then it would be consistent with that for the identified
progenitors of the SNII-L 2009kr and the high-luminosity SNII-P 2008cn. The
progenitors of these three SNe may possibly bridge the gap between lower-mass
RSG that explode as SNeII-P and LBV, or more extreme RSG, from which the more
exotic SNeII-n may arise. Very late-time imaging of the SN2009hd site may
provide us with more clues regarding the true nature of its progenitor.Comment: 19 pages, 9 figures, 3 tables, accepted for publication in Ap

Human activity in the last century has led to a substantial increase in nitrogen (N) emissions and deposition. This N deposition has reached a level that has caused or is likely to cause alterations to the structure and function of many ecosystems across the United States. One approach for quantifying the level of pollution that would be harmful to ecosystems is the critical loads approach. The critical load is dei ned as the level of a pollutant below which no detrimental ecological effect occurs over the long term according to present knowledge.
The objective of this project was to synthesize current research relating atmospheric N deposition to effects on terrestrial and aquatic ecosystems in the United States and to identify empirical critical loads for atmospheric N deposition. The receptors that we evaluated included freshwater diatoms, mycorrhizal fungi and other soil microbes, lichens, herbaceous plants, shrubs, and trees. The main responses reported fell into two categories: (1) biogeochemical, and (2) individual species, population, and community responses.

Angelman syndrome: advancing the research frontier of neurodevelopmental disorders
This report is a meeting summary of the 2010 Angelman Syndrome Foundation's scientific symposium on the neuroscience of UBE3A. Angelman syndrome is characterized by loss of speech, severe developmental delay, seizures, and ataxia. These core symptoms are caused by maternal allele disruptions of a single gene—UBE3A. UBE3A encodes an E3 ubiquitin ligase that targets certain proteins for proteasomal degradation. This biology has led to the expectation that the identification of Ube3a protein targets will lead to therapies for Angelman syndrome. The recent discovery of Ube3a substrates such as Arc (activity-regulated cytoskeletal protein) provides new insight into the mechanisms underlying the synaptic function and plasticity deficits caused by the loss of Ube3a. In addition to identifying Ube3a substrates, there have also been recent advances in understanding UBE3A's integrated role in the neuronal repertoire of genes and protein interactions. A developmental picture is now emerging whereby UBE3A gene dosage on chromosome 15 alters synaptic function, with deficiencies leading to Angelman syndrome and overexpression associated with classic autism symptomatology

Perioperative blood transfusion is associated with a gene transcription profile characteristic of immunosuppression: a prospective cohort study
INTRODUCTION
Blood transfusion in the perioperative period has frequently been associated with an excess of nosocomial infections. Whilst transfused whole blood induces specific host immune alteration that may predispose to nosocomial infections, the immunomodulating properties associated with leukodepleted blood remain incompletely understood. In this study, we explore the hypothesis that the transfusion of leukodepleted allogeneic blood during or following major gastrointestinal surgery is associated with an immunosuppressed phenotype, which may in turn predispose to postoperative infectious complications.
METHODS
Patients aged over 45 years undergoing scheduled inpatient major gastrointestinal surgery were recruited. Gene expression profiles of specific inflammatory genes were assayed from blood collected preoperatively, at 24 and at 48 hours after surgery. Genes were selected based on their ability to represent specific immune pathways. Gene expression was quantified using quantitative real-time polymerase chain reaction (qRT-PCR) to measure messenger RNA (mRNA) levels. Postoperative infections were documented using predefined criteria.
RESULTS
One hundred and nineteen patients were recruited. Fifteen (13%) patients required blood transfusion within 24 hours of surgery, 44 (37%) patients developed infections and 3 (2%) patients died prior to discharge. Patients receiving a blood transfusion were more likely to develop postoperative infections (P =0.02) and to have lower tumour necrosis factor alpha (TNFα), interleukin (IL)-12, IL-23 and RAR-related orphan receptor gamma T (RORγt) gene expression in the postoperative period (P <0.05). The TNFα/IL-10 mRNA ratio at 24 hours (P =0.0006) and at 48 hours (P =0.01) was lower in patients receiving a blood transfusion over this period. Multivariable analysis confirmed that these observations were independent of the severity of the surgical insult.
CONCLUSIONS
An association between an immunosuppressive pattern of gene expression and blood transfusion following major elective gastrointestinal surgery is described. This gene expression profile includes a reduction in the activity of innate immunity and T helper cell type 1 (Th1) and T helper cell type 17 (Th17) pathways in those patients receiving a blood transfusion. Blood transfusion was also associated with an excess of infectious complications in this cohort. A mechanistic link is suggested but not proven
Therapeutic Targeting of STAT3 (Signal Transducers and Activators of Transcription 3) Pathway Inhibits Experimental Autoimmune Uveitis
Mice with targeted deletion of STAT3 in CD4+ T-cells do not develop experimental autoimmune uveitis (EAU) or experimental autoimmune encephalomyelitis (EAE), in part, because they cannot generate pathogenic Th17 cells. In this study, we have used ORLL-NIH001, a small synthetic compound that inhibits transcriptional activity of STAT3, to ameliorate EAU, an animal model of human posterior uveitis. We show that by attenuating inflammatory properties of uveitogenic lymphocytes, ORLL-NIH001 inhibited the recruitment of inflammatory cells into the retina during EAU and prevented the massive destruction of the neuroretina caused by pro-inflammatory cytokines produced by the autoreactive lymphocytes. Decrease in disease severity observed in ORLL-NIH001-treated mice, correlated with the down-regulation of α4β1 and α4β7 integrin activation and marked reduction of CCR6 and CXCR3 expression, providing a mechanism by which ORLL-NIH001 mitigated EAU. Furthermore, we show that ORLL-NIH001 inhibited the expansion of human Th17 cells, underscoring its potential as a drug for the treatment of human uveitis. Two synthetic molecules that target the Th17 lineage transcription factors, RORγt and RORα, have recently been suggested as potential drugs for inhibiting Th17 development and treating CNS inflammatory diseases. However, inhibiting STAT3 pathways completely blocks Th17 development, as well as, prevents trafficking of inflammatory cells into CNS tissues, making STAT3 a more attractive therapeutic target. Thus, use of ORLL-NIH001 to target the STAT3 transcription factor, thereby antagonizing Th17 expansion and expression of proteins that mediate T cell chemotaxis, provides an attractive new therapeutic approach for treatment of posterior uveitis and other CNS autoimmune diseases mediated by Th17 cells

Synthesis and Assessment Product
The U.S. Government's Climate Change Science Program (CCSP) is responsible for providing the best science-based knowledge possible to inform management of the risks and opportunities associated with changes in the climate and related environmental systems. To support its mission, the CCSP has commissioned 21 "synthesis and assessment products" (SAPs) to advance decision making on climate change-related issues by providing current evaluations of climate change science and identifying priorities for research, observation, and decision support. This Report-SAP 4.4-focuses on federally managed lands and waters to provide a "Preliminary Review of Adaptation Options for Climate-Sensitive Ecosystems and Resources." It is one of seven reports that support Goal 4 of the CCSP Strategic Plan to understand the sensitivity and adaptability of different natural and managed ecosystems and human systems to climate and related global changes.
The purpose of SAP 4.4 is to provide useful information on the state of knowledge regarding adaptation options for key, representative ecosystems and resources that may be sensitive to climate variability and change. As its title suggests, this report is a preliminary review, defined as "the process of collecting and reviewing available information about known or potential adaptation options.
