    Isolation of compounds from Cannabis sativa and their anti-inflammatory effect

    Predmetom záujmu tejto práce je tradičná a zároveň kontroverzná rastlina Cannabis sativa. V teoretickej časti sú popísané ubikvitárne polyfenolické látky – flavonoidy, taktiež prítomné v mnou študovanej rastline. Podrobnejšie sú popísané doposiaľ preskúmané a potvrdené rôzne účinky na ľudský organizmus a taktiež synergia s inými látkami. Experimentálna časť práce sa zaoberá izoláciou a identifikáciou látok chloroformového a hexánového podielu extraktu získaného z tejto rastliny. Pre izoláciu boli použité chromatografické metódy – tenskovrstvová a stĺpcová chromatografia a semipreparatívna HPLC. Získané látky boli identifikované pomocou UV, IR spektrofotometrie a NMR. Následne bola skúmaná antiflogistická aktivita získaných látok, ako aj celkového etanolického extraktu a jednotlivých podielov. Výsledkom experimentu bola izolácia jednej čistej látky, ktorá bola identifikovaná ako kyselina cannabidiolová. Experimentom na THP 1 bunkách bola demonštrovaná mierna antiflogistická aktivita nepolárnych frakcií a z nich izolovanej CBDA.The object of this thesis is a traditional and also controversial plant Cannabis sativa. The theoretical part is focused on ubiquitous polyphenolic compounds – flavonoids, also present in the studied plant. All previously researched and confirmed effects on human body are described in detail. The experimental part of this thesis describes the isolation and identification of substances of the chloroform and hexane yields of extract obtained from this plant. Chromatographic methods were used for isolation – thin layer and column chromatography and semipreparative HPLC. Obtained substances were characterized using UV, IR spectrophotometry and NMR. Then the anti-inflammatory activity of obtained substances and also of the ethanolic extract and its yields was observed. The result of the experimental part was the isolation of one pure substance which was identified as the cannabidiolic acid. Using the THP 1 assay, we demonstrated mild anti-inflammatory effect of the non-polar yields and isolated CBDA.

    Cílem diplomové práce bylo provést průzkum na vybraných úpravnách vody za účelem zjištění možných poruch, se kterými se mohou provozovatelé úpraven setkat. V rámci diplomové práce byly navštíveny tyto úpravny: ÚV Troubky, ÚV Hosov, ÚV Příkazy, ÚV Černovír. Dále bylo provedeno vyhodnocení zjištěných informací; výčet a četnost poruch vyskytující se na daných provozovnách.The aim of this thesis was to conduct a survey in selected water treatment plants to identify possible faults with which treatment providers may encounter. Within the thesis has been visited these treatments: ÚV Troubky, ÚV Hosov, ÚV Příkazy, ÚV Černovír. Further was performed evaluation of the collected information, specification and frequency disturbances occurring on those premises.

    Model Checking a C++ Software Framework, a Case Study

    This paper presents a case study on applying two model checkers, SPIN and DIVINE, to verify key properties of a C++ software framework, known as ADAPRO, originally developed at CERN. SPIN was used for verifying properties on the design level. DIVINE was used for verifying simple test applications that interacted with the implementation. Both model checkers were found to have their own respective sets of pros and cons, but the overall experience was positive. Because both model checkers were used in a complementary manner, they provided valuable new insights into the framework, which would arguably have been hard to gain by traditional testing and analysis tools only. Translating the C++ source code into the modeling language of the SPIN model checker helped to find flaws in the original design. With DIVINE, defects were found in parts of the code base that had already been subject to hundreds of hours of unit tests, integration tests, and acceptance tests. Most importantly, model checking was found to be easy to integrate into the workflow of the software project and bring added value, not only as verification, but also validation methodology.     Nákaza na finančních trzích v zemích s možností přistoupení do Evropské unie

    This thesis analyses financial contagion between a reference EU market - Germany and markets of five countries which are actively seeking to become a part of European Union - Montenegro, Serbia, Turkey, Bosnia and Macedonia in the period of March 2006 to March 2018. We apply quantile regression framework to analyse contagion which we base on the occurrence and degree of coexceedances between the reference and analysed market. The results indicate that contagion between stock markets exists, however in different degree for each of the analysed markets. In addition we apply the regression framework specifically for period of financial crisis of 2008 to demonstrate that contagion is stronger during turbulent market periods.     The object of this thesis is a traditional and also controversial plant Cannabis sativa. The theoretical part is focused on botanical side of this plant and its presently used medical forms. All previously isolated substances and their observed biological activity are described in detail. The experimental part of this thesis describes the isolation and identification of substances of the chloroform extract fraction obtained from this plant. Chromatographic methods were used for isolation – analytical and semipreparative HPLC. Obtained substances were identified using UV and IR spectrophotometry. Then the biological effect of obtained substances was observed on chosen microorganisms
