    The developmental stage of inactivation of rye origin rRNA genes in the embryo and endosperm of wheat x rye F1 hybrids

    To identify the developmental stage during which the preferential inactivation of rRNA genes from the rye parent occurs in wheat x rye hybrids, nucleolar activity was evaluated in the embryo and endosperm of developing seeds of the hybrids. The hybrids were obtained from crosses of euploid and aneuploid lines of hexaploid wheat, Triticum aestivum cv. Chinese Spring, with rye, Secale cereale cv. Centeio do Alto. The number of nucleolar organizing regions (NORs) and nucleoli present in the embryo and endosperm cells of wheat, and wheat × rye F1 hybrids, at different times after fertilization was scored by silver staining. The inactivation of rDNA of rye origin in F1 hybrids occurs simultaneously in the embryo and in the endosperm between 4 and 5 days after fertilization, when these have been through six and 10 cell cycles respectively. We conclude that the genomic interactions leading to the inactivation of the rye origin rDNA is a time-dependent process, related to the developmental stage and independent of the number of cell cycles (DNA replication rounds) they have been through

    Reprogramming of rye rDNA in triticale during microsporogenesis

    To test the hypothesis that interspecific genomic and chromosome interactions leading to nucleolar dominance could be reprogrammed in meiosis, we compared the expression of distinct nucleolar organizing region (NOR) loci in hexaploid triticale root tip meristematic cells, pollen mother cells and young pollen grains. Interphase and metaphase cells were silver stained to quantify nucleoli and active NOR loci respectively. A marked difference in the ribosomal RNA gene activity of each locus was observed when different types of cells were compared: in somatic and pollen mother cells, rRNA gene activity was mainly restricted to major wheat NORs (1B and 6B) with only a small contribution from rye NORs (1R). In contrast, in young pollen grains, all NORs present, including the 1R NORs, were consistently active. The expression of all NORs just after meiosis is considered to be a consequence of meiotic reprogramming of rye origin rDNA. Gene reprogramming mediated by the resetting of methylation patterns established early in embryogenesis is suggested to be responsible for the differential expression of the NORs of rye origin in distinct developmental stages of triticale

    Visual Assessment of the Impact of Agricultural Management Practices on Soil Quality

    The intensification of agricultural practices to increase food and feed outputs is a pressing challenge causing deterioration of soil quality and soil functions. Such a challenge demands provision of empirical evidence to provide context-sensitive guidance on agricultural management practices (AMPs) that may enhance soil quality. The objectives of this study are to identify the most promising AMPs (and their combinations) applied by farmers with the most positive effects on soil quality and to evaluate the sensitivity of the soil quality indicators to the applied AMPs. The effect of selected AMPs on soil quality was assessed using a visual soil assessment tool in a total of 138 pairs of plots spread across 14 study site areas in Europe and China covering representative pedo-climatic zones. The inventory and scoring of soil quality were conducted together with landowners. Results show that 104 pairs show a positive effect of AMPs on soil quality. Higher effects of the AMPs were observed in lower fertile soils (i.e., Podzols and Calcisols) as opposed to higher fertile soils (i.e., Luvisols and Fluvisols). For the single use applications, the AMPs with positive effects were crop rotation; manuring, composting, and no-tillage; followed by organic agriculture and residue maintenance. Cluster analysis showed that the most promising combinations of AMPs with the most positive effects on soil quality are composed of crop rotation, mulching, and min-till. The agreement between scientific skills and empirical knowledge in the field identified by the farmers confirm our findings and ensures their applicability

    Rye Bs disclose ancestral sequences in cereal genomes with a potential role in gametophyte chromatid segregation

    Research ArticleTwo sequence families, E3900 and D1100, are amplified on the subtelomeric domain of the long arm of rye B chromosomes, the region that controls its drive mechanism. In this work, polymerase chain reaction (PCR) with a number of primers spanning E3900 shows that the organization and nucleotide sequence of E3900-related portions are present and highly conserved on rye A chromosomes as well as in other cereals. Quantitative Real-Time PCR estimates two E3900 sequences to be represented in 100–150 copies on Bs and at least as single copies on As. A novel E3900- related sequence, with a deletion that results in a frameshift and subsequently an open reading frame with putative DNA binding motifs, is identified. Expression analysis of E3900 indicates identical transcription levels in leaves from plants with and without Bs, showing that the expression of these sequences must be silenced on Bs and tightly regulated on As in leaves. In contrast, E3900 transcription is upregulated during meiosis exclusively in plants with Bs, maintaining a high level of transcription in the gametophyte. Interestingly, Bs not only influence their own chromatid segregation but also that of the regular chromosome complement of both rye and wheat. There is a drastic increase in frequency of disrupted metaphase and anaphase cells in the first mitosis of pollen grains carrying Bs, which appears to be due to anomalous adherences between sister chromatids. Taken together, this work provides insight into how E3900 sequences are potentially associated with important evolutionary mechanisms involved in basic cellular processes

    Faculty versus Student Repeatability on Evaluating Translucency of the Anterior Dentition

    The objective was to compare the repeatability between dental faculty, whose clinical practice was primarily restorative dentistry, and final year dental students in categorizing the inherent translucency of images selected at random using either a 3- or 7-point scale (translucent to opaque). Digital images of anterior dentition were randomly selected based on inherent translucency. Thirty images (five were repeated) were randomized and categorized by 20 dental students and 20 faculty on their inherent translucency. Statistical analysis was performed using an F test for analysis of variance at 95% confidence interval. A covariance parameter estimate (CPE) was accomplished to compare the inter-rater variability of the dental faculty and dental students. Statistically, more variability occurred between Slides (CPE of 0.185 (p = 0.001)) and between Subject and Slide (CPE of 0.122 (p = 0.0002)) than within subjects (CPE of 0.021 (p = 0.083)). Viewing repeat Slides, Students (CPE = 0.16) were more consistent (p \u3c 0.05) than faculty (CPE = 1.8) using the 3- point scale, while the CPE was the same (CPE = 0.669) using 7-point scale. Dental students and faculty were consistent using the 7-point scale to judge repeat slides, while dental students in this limited pilot study were more consistent when viewing a repeat slide using the 3-point scale

    As masterclasses internacionais em física de partículas em Portugal

    Todos os anos, desde 2005, institutos de investigação e universidades de todo o mundo convidam estudantes do ensino secundário a experimentar a linha da frente da investigação fundamental. São as Masterclasses Internacionais em Física de Partículas, um programa do International Particle Physics Outreach Group (IPPOG) que oferece aos estudantes a oportunidade de serem cientistas por um dia, analisando dados reais recolhidos no acelerador LHC (Large Hadron Collider) do CERN, o Centro Europeu de Física de Partículas. Em 2015, participaram nas Masterclasses estudantes de 42 países, recebidos em 210 universidades e laboratórios. A participação de países de todo o mundo reflete a internacionalização das colaborações em Física de Partículas. Os estudantes vivenciam este aspeto na videoconferência com que terminam o seu dia de pesquisa, apresentando os resultados aos participantes de outros países, com a moderação de cientistas no CERN. Portugal participa desde a primeira edição, com a colaboração do LIP - Laboratório de Instrumentação e Física Experimental de Partículas, promotor nacional da atividade, e o apoio da Agência Ciência Viva. Em 2014, cerca de dois mil estudantes e centena e meia de professores participaram nas Masterclasses organizadas em 13 instituições do ensino superior um pouco por todo o País: de Faro a Braga e de Bragança a Ponta Delgada, passando por Beja, Évora, Lisboa, Covilhã, Coimbra, Aveiro, Porto e Vila Real. Foi ainda prestado apoio à realização das Masterclasses na Universidade de São Tomé e Príncipe. A dimensão e o sucesso das Masterclasses em Portugal não são a regra na generalidade dos países europeus, tendo-se notado um crescimento acentuado desde o inicio da realização de Escola de Física no CERN para professores em língua portuguesa. Nos últimos anos, verifica-se que cerca de 20% dos participantes nas Masterclasses são portugueses. Além da aquisição de conhecimentos ou da perceçâo do fascínio da investigação fundamental, aspetos como a cooperação internacional na ciência, a comunicação entre cientistas de diferentes línguas e culturas, ou a estreita relação de interdependência entre ciência fundamental e tecnologia, que o CERN tão bem representa, são aprendizagens a valorizar. Neste contexto, importa hoje sobretudo refletir sobre as formas de maximizar o ganho para todos os participantes

    Biomimetic coatings enhance tribocorrosion behavior and cell responses of commercially pure titanium surfaces

Our aims were ( 1) to create bioactive Ti-coatings doped with Ca, P, Si, and Ag produced by microarc oxidation ( MAO) to improve the surface properties of biomedical implants, ( 2) to investigate the TiO2 layer stability under wear and corrosion, and ( 3) to evaluate human mesenchymal stem cells ( hMSCs) responses cultured on the modified surfaces. Tribocorrosion and cell experiments were performed following the MAO treatment. Samples were divided as a function of different Ca/P concentrations and treatment duration. Higher Ca concentration produced larger porous and harder coatings compared to the untreated group ( p<0.001), due to the presence of rutile structure. Free potentials experiments showed lower drops ( 0.6 V) and higher coating lifetime during sliding for higher Ca concentration, whereas lower concentrations presented similar drops ( 0.8 V) compared to an untreated group wherein the drop occurred immediately after the sliding started. MAO-treated surfaces improved the matrix formation and osteogenic gene expression levels of hMSCs. Higher Ca/P ratios and the addition of Ag nanoparticles into the oxide layer presented better surface properties, tribocorrosive behavior, and cell responses. MAO is a promising technique to enhance the biological, chemical, and mechanical properties of dental implant surfaces.     Polymerization cycles on hardness and surface gloss of denture bases

    The aim of this study was to evaluate different polymerization cycles on the hardness and surface gloss of acrylic resins for denture bases. Classico and Vip Cril Plus acrylic resins samples were made in dental stone molds. Powder-liquid ratio and resin manipulation were according to the manufacturers’ instructions, and the resulting mass pressed in metallic fl asks. The polymerization cycles were A - hot water bath at 74°C for 9 hours; B - hot water bath at 74°C for 8 hours + 100°C for 1 hour, and C - hot water bath at 74°C for 2 hours + 100°C for 1 hour. After polymerization, the samples were defl asked and submitted to finishing and polishing procedures, and stored in water at 37°C for 24 hours. A hardness indenter with load of 25 gf for 10 s evaluated the Knoop hardness values. A gloss meter evaluated the surface gloss using a light incidence of 60°. Data were submitted to ANOVA and Tukey’s test (α = 0.05). Hardness: There were signifi cant differences between resins regardless of polymerization cycles (Classico = 22.28 and Vip Cril Plus = 25.83). Significant differences occurred among polymerization cycles regardless of resins (A = 25.83, B = 24.64 and C = 21.73). There was similarity for the resin and cycle interaction (Classico: A = 24.51, B = 22.68 and C = 19.65; Vip Cril Plus: A = 27.15, B = 26.53 and C = 23.81). Surface gloss. Significant differences were shown between resins regardless of polymerization cycle (Classico = 57.26 and Vip Cril Plus = 49.38) and between polymerization cycles regardless of resin (A = 48.82, B = 53.46 and C = 57.68). Statistical diff erences were also found for the resin and cycle interaction (Classico: A = 52.32, B = 63.79 and C = 55.67; Vip Cril Plus: A = 45.32, B = 43.14 and C = 59.69). Diff erent polymerization cycles showed similar eff ects on hardness and different effects on the surface gloss of denture base acrylic resins
