    Building Authentic Biblical Community at the Pine Forge Seventh-day Adventist Church

    Problem A spirit of division at the Pine Forge Seventh-day Adventist Church is adversely affecting its ability to agree on mission and ministry focus. Method Over the course of 12 months, an investigation of the source of division was conducted through pastoral observation, surveys of the adult members, and a focus group of pastors of churches on boarding academy campuses. The data from the focus group and surveys was compared to that of the Pine Forge Seventh-day Adventist church. A Bible study series on building authentic biblical community was also conducted that taught biblical principles focusing on putting aside differences and dwelling together in spiritual unity. The success of the Bible study was measured by the positive changes in the attitudes of the members during discussions of ministry focus. Results It was discovered that the division over the ministry focus was promoted mostly by those who felt that they were unjustly terminated by the Academy. At one church, the surveys revealed that seven of 31 people who were asked if their departure from the institution was negative said, Yes. Of that group of seven, six indicated that they were against the church\u27s focusing its ministry primarily toward the academy and its students. The focus group also revealed a high level of apathy among those who had been terminated. Conclusion The study reveals that an examination of the history and demographics of the church assists the church in returning to its original student-focused ministry. There is also a need for a specific ministry for teaching biblical community principles as well as ministry care for those who suffered job loss in the community. Finally, churches that have polarizing missions need to find a ministry activity around which the members can build their ministry focus

    Comments on Black Holes in Matrix Theory

    The recent suggestion that the entropy of Schwarzschild black holes can be computed in matrix theory using near-extremal D-brane thermodynamics is examined. It is found that the regime in which this approach is valid actually describes black strings stretched across the longitudinal direction, near the transition where black strings become unstable to the formation of black holes. It is argued that the appropriate dynamics on the other (black hole) side of the transition is that of the zero modes of the corresponding super Yang-Mills theory. A suggestive mean field theory argument is given for the entropy of black holes in all dimensions.     Is string theory a theory of quantum gravity?

    Some problems in finding a complete quantum theory incorporating gravity are discussed. One is that of giving a consistent unitary description of high-energy scattering. Another is that of giving a consistent quantum description of cosmology, with appropriate observables. While string theory addresses some problems of quantum gravity, its ability to resolve these remains unclear.     Analyticity and the Holographic S-Matrix

    We derive a simple relation between the Mellin amplitude for AdS/CFT correlation functions and the bulk S-Matrix in the flat spacetime limit, proving a conjecture of Penedones. As a consequence of the Operator Product Expansion, the Mellin amplitude for any unitary CFT must be a meromorphic function with simple poles on the real axis. This provides a powerful and suggestive handle on the locality vis-a-vis analyticity properties of the S-Matrix. We begin to explore analyticity by showing how the familiar poles and branch cuts of scattering amplitudes arise from the holographic description. For this purpose we compute examples of Mellin amplitudes corresponding to 1-loop and 2-loop Witten diagrams in AdS. We also examine the flat spacetime limit of conformal blocks, implicitly relating the S-Matrix program to the Bootstrap program for CFTs. We use this connection to show how the existence of small black holes in AdS leads to a universal prediction for the conformal block decomposition of the dual CFT.Comment: 28+15 pages, 7 figures; v2: typos correcte

    We perform a general study of primordial scalar non-Gaussianities in single field inflationary models in Einstein gravity. We consider models where the inflaton Lagrangian is an arbitrary function of the scalar field and its first derivative, and the sound speed is arbitrary. We find that under reasonable assumptions, the non-Gaussianity is completely determined by 5 parameters. In special limits of the parameter space, one finds distinctive ``shapes'' of the non-Gaussianity. In models with a small sound speed, several of these shapes would become potentially observable in the near future.     PerR Confers Phagocytic Killing Resistance and Allows Pharyngeal Colonization by Group A Streptococcus

    The peroxide response transcriptional regulator, PerR, is thought to contribute to virulence of group A Streptococcus (GAS); however, the specific mechanism through which it enhances adaptation for survival in the human host remains unknown. Here, we identify a critical role of PerR-regulated gene expression in GAS phagocytosis resistance and in virulence during pharyngeal infection. Deletion of perR in M-type 3 strain 003Sm was associated with reduced resistance to phagocytic killing in human blood and by murine macrophages in vitro. The increased phagocytic killing of the perR mutant was abrogated in the presence of the general oxidative burst inhibitor diphenyleneiodonium chloride (DPI), a result that suggests PerR-dependent gene expression counteracts the phagocyte oxidative burst. Moreover, an isogenic perR mutant was severely attenuated in a baboon model of GAS pharyngitis. In competitive infection experiments, the perR mutant was cleared from two animals at 24 h and from four of five animals by day 14, in sharp contrast to wild-type bacteria that persisted in the same five animals for 28 to 42 d. GAS genomic microarrays were used to compare wild-type and perR mutant transcriptomes in order to characterize the PerR regulon of GAS. These studies identified 42 PerR-dependent loci, the majority of which had not been previously recognized. Surprisingly, a large proportion of these loci are involved in sugar utilization and transport, in addition to oxidative stress adaptive responses and virulence. This finding suggests a novel role for PerR in mediating sugar uptake and utilization that, together with phagocytic killing resistance, may contribute to GAS fitness in the infected host. We conclude that PerR controls expression of a diverse regulon that enhances GAS resistance to phagocytic killing and allows adaptation for survival in the pharynx
