    Association patterns of volatile metabolites in urinary excretions among Type-2 Non-Insulin dependent diabetes patients

    Background: Patterns of volatile metabolites in urine are important to detect abnormalities associated with diabetes. Present study was conducted to find out the excretion patterns of endogenously produced alcohols in urine for type 2 (Non-Insulin Dependent) diabetes mellitus. A cross sectional analytical study was conducted with duration extended from Jan to Mar 2015.Methods: The current study included 40 patients with chronic type 2 diabetes mellitus. In total, 10 sex and age matched subjects with no history of any disease were considered as controls. Blood sugar was estimated by autoanalyzer using standard kit of Merck following manufacturer`s instructions. Urine sugar was quantitatively detected by biuret reagent using titration technique. Urinary alcohol was identified and estimated by gas chromatography.  Urinary ketone bodies were estimated by urinary strip.Results: It was observed that level of fasting blood sugar was significantly increased (P<0.001) in patients as compared to their controls. The blood sugar and urinary alcohol in patients were 3.0% and 6.0% respectively. Urinary ketone bodies were found to be 2+. On the other hand urine sugar, alcohol and ketone bodies were not detected in the negative control subjects.Conclusions: It is concluded that urinary alcohol is endogenously produced in patients with type 2 diabetes due to uncontrolled hyperglycemia. However further work is needed to find out the ratio of urinary and blood alcohol which may confirm the present findings

    Detection and comparison of light metals in hair among workers of different industries using Particle Induced X-Ray Emission (PIXE) for forensic casework

    Background: The study was planned to evaluate the deposition of some metals in the scalp hair of the people working in the particular environment. Scalp hair samples were collected from different industry workers including pharmaceutical, textile and paint industry and analyzed for the determination of aluminum (Al), calcium (Ca), potassium (K) and sulfur (S). Hair analysis provides a better assessment of light metals present in the surroundings, as well as monitoring of variation of metals from place to place.Methods: The hair samples were collected from 21-55 years old employees working in industry for more than four years. Workers of the age less than 20 years and less than 4 years working experience were not including in this study. Particle Induced X-ray Emission (PIXE) system was used to detect the light metals from hair and probable matches were searched through National Institute of Standards and Technology (NIST) library.Results: The metals Al, S, K and Ca concentration was found maximum in the pharmaceutical industry workers. i.e., 575.1 ”g/g, 190.7 ”g/g, 11.1 ”g/g and 9.1 ”g/g respectively. The minimum concentration so these metals was found in paint industry workers i.e. 103.7 ”g/g, 8.87 ”g/g, 2.3 ”g/g, and 1.7”g/g respectively.Conclusion: Light metal concentrations in hair samples showed a significant positive correlation. Our findings can play vital role for health departments and industrial environmental management system (EMS) authorities in policy making and implementation.     Human Cryptosporidiosis: An insight into Epidemiology, Modern Diagnostic Tools and Recent Drug Discoveries

    Cryptosporidiosis is an emerging food and water borne zoonotic disease, which is caused by genusCryptosporidium. The first Cryptosporidium spp. was isolated from mice in 1907 and gained importance when it was found in an HIV positive patient. It usually causes self-limiting diarrhea in young children and immunocompetent patients. However, it may lead to chronic diarrhea with life threatening condition in immunocompromised patients. Other complications related to this transmittable infection may include respiratory problems, skin rashes and headache. HIV/AIDS patients are highly susceptible host for this parasite. Cryptosporidium parvum and Cryptosporidium hominis are the known pathogenic species, prevalent among humans and they are being transmitted through contaminated food and water. Usually, the diagnosis of Cryptosporidium spp. is dependent on microscopic technique in many countries, which has a low sensitivity and specificity leading to false positive results. However, for a step forward to successful epidemiological studies, advanced techniques (Serological and DNA-based) provide us the better ways of diagnosis with more sensitivity and specificity. Furthermore, no antiparasitic drug has found to be effective againstCryptosporidium spp. except Nitazoxanide which is FDA-approved and effective only when administered along with antiretroviral therapy. In this regard, present review summarizes the various epidemiological studies conducted around the globe along with modern diagnostic tools and the suitable treatment available now a days. This systemized review will help the scientists to better understand all the aspects of cryptosporidiosis at one platform which may help in designing surveillance studies through selection of sensitive diagnostic techniques. The new drugs mentioned in this review may also help to better control this parasite in humans, especially immunocompromised individuals

    The development and validation of a scoring tool to predict the operative duration of elective laparoscopic cholecystectomy

    Background: The ability to accurately predict operative duration has the potential to optimise theatre efficiency and utilisation, thus reducing costs and increasing staff and patient satisfaction. With laparoscopic cholecystectomy being one of the most commonly performed procedures worldwide, a tool to predict operative duration could be extremely beneficial to healthcare organisations. Methods: Data collected from the CholeS study on patients undergoing cholecystectomy in UK and Irish hospitals between 04/2014 and 05/2014 were used to study operative duration. A multivariable binary logistic regression model was produced in order to identify significant independent predictors of long (> 90 min) operations. The resulting model was converted to a risk score, which was subsequently validated on second cohort of patients using ROC curves. Results: After exclusions, data were available for 7227 patients in the derivation (CholeS) cohort. The median operative duration was 60 min (interquartile range 45–85), with 17.7% of operations lasting longer than 90 min. Ten factors were found to be significant independent predictors of operative durations > 90 min, including ASA, age, previous surgical admissions, BMI, gallbladder wall thickness and CBD diameter. A risk score was then produced from these factors, and applied to a cohort of 2405 patients from a tertiary centre for external validation. This returned an area under the ROC curve of 0.708 (SE = 0.013, p  90 min increasing more than eightfold from 5.1 to 41.8% in the extremes of the score. Conclusion: The scoring tool produced in this study was found to be significantly predictive of long operative durations on validation in an external cohort. As such, the tool may have the potential to enable organisations to better organise theatre lists and deliver greater efficiencies in care

    Biohydrogen is regarded as an attractive future clean energy carrier due to its high energy content and environmentally friendly conversion. Biohydrogen reactor is widely used in studies concerning the anaerobic co-digestion of food waste, sewage sludge, wastewater and other organic solids. Anaerobic digestion is a series of biological processes in which microorganisms break down biodegradable material (biomass or waste feedstock) in the absence of oxygen to produce biogas, which may generate electricity and heat, or can be processed into renewable natural gas and transportation fuels. This review article explains the scientific processes of anaerobic digestion process such as hydrolysis, acidogenesis, acetogenesis and hydrogenesis as well as methods to produce biohydrogen gas such as fermentation and biophotolysis for the waste management technology and sources of renewable energy and concludes with solutions that may allow anaerobic digestion to become more widely adopted throughout the developing countries to control the waste management system

    Genetic Characterization of Wakhi People from Hunza Valley of Pakistan by employing Mitochondrial DNA Control Region

    Background: Study of populations at genetic level marks high importance in terms of determination of population specific patterns. The study and analysis of population specific genetic patterns have wide ranging applications from medicine to forensic science. The study of mtDNA control regions gives the insight of maternal lineages of particular ethnic group and their evolutionary as well emigrational history. Methods: This research gives information of mitochondrial DNA haplotypes data of CR (control region) covering the area from 16024bp to 576bp of mitochondrial DNA of Wakhi population of Hunza valley from Pakistan. Samples of 40 unrelated Wakhi from upper Hunza were sequenced and their sequences encapsulating mtDNA control region was compared to rCRSs (revised Cambridge reference sequence) to see maternally inherited DNA variation at genetic level in this population.Keywords: Forensic Science; Control region; Haplogroup; Genetic diversity Keywords: Forensic Science; Control region; Haplogroup; Genetic diversity Results: The results showed that all forty (40) haplotypes are unique. The haplotypes corresponded to 67.9% West Eurasian haplogroups followed by the Middle East and variety of Asian haplogroups exhibiting admixed maternal genetics of this population. Wakhi population comes with high genetic diversity (0.998) in turn lowest random match probability (0.026) and high power of discrimination (0.974).Conclusion: This study gives interesting highlights important aspect of uni-parental genetics of Wakhi population and is also a contribution to mtDNA control region data of Pakistani populations for applications in criminal investigations.

    Prediction of Body Weight from Testicular and Morphological Characteristics in Indigenous Mengali Sheep of Pakistan using Factor Analysis Scores in Multiple Linear Regression Analysis

    The objective of the investigation was to predict body weight (BW) from testicular and morphological characteristics of the indigenous Mengali Sheep in Balochistan, Pakistan. The characteristics recorded were: body weight (BW), testicular length (TL), scrotal length (SL), scrotal circumference (SC), body length (BL), withers height (WH) and heart girth (HG) of 107 indigenous Mengali sheep in age range of 12 to 48 months. Only multiple linear regression analysis fitting to the available data showed tendency to multicollinearity problem with VIF >10 and didn’t present optimal solutions. With the purpose of removing the multicollinearity problem and especially making more correct interpretation biologically, the data were exposed to a combination of Factor and Multiple Linear Regression analyses. The appropriateness of factor analysis for the current data was determined by using criteria such as Kaiser-Meyer Olkin (KMO) measure of sampling adequacy and Bartlett’s test of spherity. These criteria showed that the data were appropriate for the factor analysis, one of the multivariate statistical analysis techniques. Thereafter, the factor analysis was applied to explanatory variables in order to derive new-uncorrelated latent explanatory variables from the variables. In the factor analysis, three new latent variables whose eigenvalues were greater than one were considered as explanatory variable for multiple linear regression analysis. Results obviously illustrated that 79.1% of variation in body weight was effectively explained by these new latent explanatory variables. Consequently, if confirmed at genetic level, the testicular and body traits might provide a significant contribution in the genetic improvement of the body weight of the indigenous Mengali for sheep breeding
