    Theatre Is the People Who Make It. The Romanian Theatre-work Experience by an Italian Director

    The present paper presents the journey of an Italian director in Romanian theatre, from the first encounter with Romanian great performances abroad, in  Festivals, to the work on stage with Romanian actors, and for a Romanian audience, in Cluj-Napoca. The National Theatre in Cluj-Napoca offered the place of  this encounter of different energies, ways of living, making and approaching theatre, and the space for an artistic challenge: to work not only in the  experimental way the director was used to at the Laboratory in Pontedera, but to find the right path in the more strict system of a Repertory Theatre and for  a much larger audience in a shorter laps of time. This journey is not only about theatrical places but also and mainly about people met there, about  exchanges, work, challenges, wishes, artistic results in the past and the present and projections in the future

    An Automatic System for Modeling and Controlling Color Quality of Dyed Leathers in Tanneries

    Abstract This paper presents an automatic system for modeling and controlling color quality of dyed leathers of an Italian tannery. The proposed software is implemented within the IT company system, and is fully integrated with the machineries of the finishing line, that is, a spraying cabin with a robotic carousel, and an electronic tintometer system. Suitable experimental tests according to the Design of Experiments (DoE) are firstly defined, executed and analyzed for a series of color tones of interest. In order to derive and validate a set of colorimetric models able to evaluate and predict the color rendering of painted leathers, a set of recipes of basic dye pigments and data of light reflection measured by a multispectral camera are used. Principal Component Analysis is applied for dimensionality reduction, and linear least squares regression is employed to identify these data-driven models, which are then used for color control purpose. A color correction feedback strategy is indeed developed in order to converge towards the various target formulations. The control algorithm aims at reaching the multispectral reading values of the reference, that is, the first sample of unknown color recipe starting from the most similar archive base and appropriately updating the recipe of pigments, by using the measurement of leather samples prepared from time to time by the finishing line machineries. A set of company data are used to successfully validate the identified colorimetric models and the proposed color correction strategy

    A general method for the selection of high-level scFv and IgG antibody expression by stably transfected mammalian cells

    The isolation of mammalian cell lines capable of high-yield expression of recombinant antibodies is typically performed by screening multiple individual clones by limiting dilution techniques. A number of experimental strategies have recently been devised to identify high-expressing clones, but protocols are often difficult to implement, time consuming, costly and limited in terms of number of clones which can be screened. In this article, we describe new vectors for the expression of recombinant antibodies in IgG format and in other formats, based on the single-chain Fv module, as well as a high-throughput screening procedure, based on the direct staining of antibodies transiting the membrane of a stably transfected cell, followed by preparative sorting using a high-speed cell sorter. This procedure allows, in one step, to deposit single cells into individual wells of a 96-well microtiter plate (thus facilitating cloning) and to preferentially recover those rare cell populations which express dramatically higher levels of recombinant antibody. Using cell cultures followed by affinity purification techniques, we could confirm that the new vectors and the new screening procedure reliably yield high-expression clones and homogenous protein preparations.     Implementation of an Industry 4.0 system to optimally manage chemical plant operation

    The evolution of the process industry in the direction of automation and digitalization is nowadays a consolidated phenomenon. In this direction, Industry 4.0 paradigms are leading many industrial companies to significantly update their facilities. This paper presents a scheduling algorithm that takes the role of a real-time optimization (RTO) element in a larger project framework where the various network components are aimed to be all highly interconnected. The proposed methodology is applied to an Italian chemical industrial site, in order to best manage the production rates of the various products and the sales plan for the different clients. Numerous plants and processes are considered into the model: batch and continuous production lines, saleable and non-storable products. Concepts of linear optimization and batch operation scheduling are used in the algorithm construction. This whole structure lays the foundation for a full integration between different elements of the facility, that is, the control systems and the selling department

    Case report of an atypical early onset X-linked retinoschisis in monozygotic twins

    BACKGROUND: X-linked Retinoschisis (XLRS) is one of the most common macular degenerations in young males, with a worldwide prevalence ranging from 1:5000 to 1:20000. Clinical diagnosis of XLRS can be challenging due to the highly variable phenotypic presentation and limited correlation has been identified between mutation type and disease severity or progression. CASE PRESENTATION: We report the atypical early onset of XLRS in 3-month-old monozygotic twins. Fundus examination was characterized by severe bullous retinal schisis with pre-retinal and intraretinal haemorrhages. Molecular genetic analysis of the RS1 was performed and the c.288G > A (p. Trp96Ter) mutation was detected in both patients. CONCLUSIONS: Early onset XLRS is associated with a more progressive form of the disease, characterized by large bullous peripheral schisis involving the posterior pole, vascular abnormalities and haemorrhages. The availability of specific technology permitted detailed imaging of the clinical picture of unusual cases of XLRS. The possible relevance of modifying genes should be taken into consideration for the future development of XLRS gene therapy

    Sclerosing Angiomatoid Nodular Transformation of the Adrenal Gland: A Case Report of a Novel Histopathological Entity

    The finding of an indeterminate adrenal mass at radiological investigations is a challenge for physicians. Complex diagnostic work-up, periodic follow-up, or surgical intervention are therefore needed to rule out malignant lesions. Tertiary care hospitals are provided with F-18-fludeoxyglucose (F-18-FDG) positron emission tomography (PET) and F-18-dihydroxyphenylalanine (F-18-DOPA) PET, which aid in the characterization of indeterminate adrenal masses. Nevertheless, the histopathological examination may be required to exclude malignancy or rare etiologies. A 54-year-old woman presented to our clinic 6 months after a cerebral hemorrhage. She was hypertensive and had recently discovered a left adrenal mass of 15 mm during an abdominal ultrasound. Contrast-enhanced CT, following adrenal protocol, revealed a 14-mm adrenal mass without characteristics suggestive of an adrenal adenoma. Tumor markers were negative. Functional tests excluded hormone hypersecretion. An F-18-DOPA PET was negative. An F-18-FDG PET showed mild uptake of both the adrenal glands, with a more circumscribed pattern in the left one (maximum standardized uptake value 5 4). As the clinical diagnosis was still indeterminate, we performed laparoscopic left adrenalectomy. The histopathological examination described a sclerosing angiomatoid nodular transformation (SANT) of the adrenal gland, a benign lesion already described as a rare occurrence only in the spleen. IgG4 levels were reduced. In conclusion, this is a report of a SANT of the adrenal gland, a novel entity that should be taken into consideration in the differential diagnosis of indeterminate adrenal masses at CT scan.     Deep learning for necrosis detection using canine perivascular wall tumour whole slide images

    Necrosis seen in histopathology Whole Slide Images is a major criterion that contributes towards scoring tumour grade which then determines treatment options. However conventional manual assessment suffers from inter-operator reproducibility impacting grading precision. To address this, automatic necrosis detection using AI may be used to assess necrosis for final scoring that contributes towards the final clinical grade. Using deep learning AI, we describe a novel approach for automating necrosis detection in Whole Slide Images, tested on a canine Soft Tissue Sarcoma (cSTS) data set consisting of canine Perivascular Wall Tumours (cPWTs). A patch-based deep learning approach was developed where different variations of training a DenseNet-161 Convolutional Neural Network architecture were investigated as well as a stacking ensemble. An optimised DenseNet-161 with post-processing produced a hold-out test F1-score of 0.708 demonstrating state-of-the-art performance. This represents a novel first-time automated necrosis detection method in the cSTS domain as well specifically in detecting necrosis in cPWTs demonstrating a significant step forward in reproducible and reliable necrosis assessment for improving the precision of tumour grading

    Pathobiology of Anaplastic Large Cell Lymphoma

    The authors revise the concept of anaplastic large cell lymphoma (ALCL) in the light of the recently updated WHO classification of Tumors of Hematopoietic and Lymphoid Tissues both on biological and clinical grounds. The main histological findings are illustrated with special reference to the cytological spectrum that is indeed characteristic of the tumor. The phenotype is reported in detail: the expression of the ALK protein as well as the chromosomal abnormalities is discussed with their potential pathogenetic implications. The clinical features of ALCL are presented by underlining the difference in terms of response to therapy and survival between the ALK-positive and ALK-negative forms. Finally, the biological rationale for potential innovative targeted therapies is presented
