    In der vorliegenden Arbeit werden die Lobbyaktivitäten des Deutschen Bibliothekverbands e.V. hinsichtlich der Ausgestaltung von Urheberrechtsschranken zur öffentlichen Zugänglichmachung für Unterricht und Forschung (später geregelt in § 52a UrhG) sowie der Wiedergabe an elektronischen Leseplätzen (später geregelt in § 52b UrhG) untersucht. Schwerpunktmäßig wird aufgezeigt, welche Faktoren Einfluss auf die Veränderung von Positionen und Zielen genommen haben. Dabei spielt auch das problematische Verhältnis zum Börsenverein des Deutschen Buchhandels eine Rolle. Am Ende der Arbeit steht zudem eine Bewertung des Erfolgs der Lobbyarbeit

    Vps27 recruits ESCRT machinery to endosomes during MVB sorting

    Down-regulation (degradation) of cell surface proteins within the lysosomal lumen depends on the function of the multivesicular body (MVB) sorting pathway. The function of this pathway requires the class E vacuolar protein sorting (Vps) proteins. Of the class E Vps proteins, both the ESCRT-I complex (composed of the class E proteins Vps23, 28, and 37) and Vps27 (mammalian hepatocyte receptor tyrosine kinase substrate, Hrs) have been shown to interact with ubiquitin, a signal for entry into the MVB pathway. We demonstrate that activation of the MVB sorting reaction is dictated largely through interactions between Vps27 and the endosomally enriched lipid species phosphatidylinositol 3-phosphate via the FYVE domain (Fab1, YGL023, Vps27, and EEA1) of Vps27. ESCRT-I then physically binds to Vps27 on endosomal membranes via a domain within the COOH terminus of Vps27. A peptide sequence in this domain, PTVP, is involved in the function of Vps27 in the MVB pathway, the efficient endosomal recruitment of ESCRT-I, and is related to a motif in HIV-1 Gag protein that is capable of interacting with Tsg101, the mammalian homologue of Vps23. We propose that compartmental specificity for the MVB sorting reaction is the result of interactions of Vps27 with phosphatidylinositol 3-phosphate and ubiquitin. Vps27 subsequently recruits/activates ESCRT-I on endosomes, thereby facilitating sorting of ubiquitinated MVB cargoes

    Glycosylation-mediated phenylpropanoid partitioning in Populus tremuloides cell cultures

    <p>Abstract</p> <p>Background</p> <p>Phenylpropanoid-derived phenolic glycosides (PGs) and condensed tannins (CTs) comprise large, multi-purpose non-structural carbon sinks in <it>Populus</it>. A negative correlation between PG and CT concentrations has been observed in several studies. However, the molecular mechanism underlying the relationship is not known.</p> <p>Results</p> <p><it>Populus </it>cell cultures produce CTs but not PGs under normal conditions. Feeding salicyl alcohol resulted in accumulation of salicins, the simplest PG, in the cells, but not higher-order PGs. Salicin accrual reflected the stimulation of a glycosylation response which altered a number of metabolic activities. We utilized this suspension cell feeding system as a model for analyzing the possible role of glycosylation in regulating the metabolic competition between PG formation, CT synthesis and growth. Cells accumulated salicins in a dose-dependent manner following salicyl alcohol feeding. Higher feeding levels led to a decrease in cellular CT concentrations (at 5 or 10 mM), and a negative effect on cell growth (at 10 mM). The competition between salicin and CT formation was reciprocal, and depended on the metabolic status of the cells. We analyzed gene expression changes between controls and cells fed with 5 mM salicyl alcohol for 48 hr, a time point when salicin accumulation was near maximum and CT synthesis was reduced, with no effect on growth. Several stress-responsive genes were up-regulated, suggestive of a general stress response in the fed cells. Salicyl alcohol feeding also induced expression of genes associated with sucrose catabolism, glycolysis and the Krebs cycle. Transcript levels of phenylalanine ammonia lyase and most of the flavonoid pathway genes were reduced, consistent with down-regulated CT synthesis.</p> <p>Conclusions</p> <p>Exogenous salicyl alcohol was readily glycosylated in <it>Populus </it>cell cultures, a process that altered sugar utilization and phenolic partitioning in the cells. Using this system, we identified candidate genes for glycosyltransferases that may mediate the glycosylation, and for transporters that mediate the subcellular compartmentalization of sugars and phenolic glycosides. The suspension cells appear to represent a facile system for dissecting the regulation of phenolic carbon partitioning, and in turn, its effects on growth in <it>Populus</it>.</p

    Untangling methodological and scale considerations in growth and productivity trend estimates of Canada’s forests

    In view of the economic, social and ecological importance of Canada's forest ecosystems, there is a growing interest in studying the response of these ecosystems to climate change. Accurate knowledge regarding growth trajectories is needed for both policy makers and forest managers to ensure sustainability of the forest resource. However, results of previous analyses regarding the sign and magnitude of trends have often diverged. The main objective of this paper was to analyse the current state of scientific knowledge on growth and productivity trends in Canada's forests and provide some explanatory elements for contrasting observations. The three methods that are commonly used for assessments of tree growth and forest productivity (i.e. forest inventory data, tree-ring records, and satellite observations) have different underlying physiological assumptions and operate on different spatiotemporal scales, which complicates direct comparisons of trend values between studies. Within our systematic review of 44 peer-reviewed studies, half identified increasing trends for tree growth or forest productivity, while the other half showed negative trends. Biases and uncertainties associated with the three methods may explain some of the observed discrepancies. Given the complexity of interactions and feedbacks between ecosystem processes at different scales, researchers should consider the different approaches as complementary, rather than contradictory. Here, we propose the integration of these different approaches into a single framework that capitalizes on their respective advantages while limiting associated biases. Harmonization of sampling protocols and improvement of data processing and analyses would allow for more consistent trend estimations, thereby providing greater insight into climate-change related trends in forest growth and productivity. Similarly, a more open data-sharing culture should speed-up progress in this field of research
