Semaphorin-Plexin Signaling Controls Mitotic Spindle Orientation during Epithelial Morphogenesis and Repair

© 2015 Elsevier Inc. Morphogenesis, homeostasis, and regeneration of epithelial tissues rely on the accurate orientation of cell divisions, which is specified by the mitotic spindle axis. To remain in the epithelial plane, symmetrically dividing epithelial cells align their mitotic spindle axis with the plane. Here, we show that this alignment depends on epithelial cell-cell communication via semaphorin-plexin signaling. During kidney morphogenesis and repair, renal tubular epithelial cells lacking the transmembrane receptor Plexin-B2 or its semaphorin ligands fail to correctly orient the mitotic spindle, leading to severe defects in epithelial architecture and function. Analyses of a series of transgenic and knockout mice indicate that Plexin-B2 controls the cell division axis by signaling through its GTPase-activating protein (GAP) domain and Cdc42. Our data uncover semaphorin-plexin signaling as a central regulatory mechanism of mitotic spindle orientation necessary for the alignment of epithelial cell divisions with the epithelial plane.University Medical Center Giessen and Marburg (UKGM) to T.W. WIP Regulates Persistence of Cell Migration and Ruffle Formation in Both Mesenchymal and Amoeboid Modes of Motility

    Get PDF
The spatial distribution of signals downstream from receptor tyrosine kinases (RTKs) or G-protein coupled receptors (GPCR) regulates fundamental cellular processes that control cell migration and growth. Both pathways rely significantly on actin cytoskeleton reorganization mediated by nucleation-promoting factors such as the WASP-(Wiskott-Aldrich Syndrome Protein) family. WIP (WASP Interacting Protein) is essential for the formation of a class of polarised actin microdomain, namely dorsal ruffles, downstream of the RTK for PDGF (platelet-derived growth factor) but the underlying mechanism is poorly understood. Using lentivirally-reconstituted WIP-deficient murine fibroblasts we define the requirement for WIP interaction with N-WASP (neural WASP) and Nck for efficient dorsal ruffle formation and of WIP-Nck binding for fibroblast chemotaxis towards PDGF-AA. The formation of both circular dorsal ruffles in PDGF-AA-stimulated primary fibroblasts and lamellipodia in CXCL13-treated B lymphocytes are also compromised by WIP-deficiency. We provide data to show that a WIP-Nck signalling complex interacts with RTK to promote polarised actin remodelling in fibroblasts and provide the first evidence for WIP involvement in the control of migratory persistence in both mesenchymal (fibroblast) and amoeboid (B lymphocytes) motility. © 2013 Banon-Rodriguez et al.Spanish Ministry of Science and Innovation (BFU2010-21374/BMC), (BFU2011-30097); CIBERNED (Instituto de Salud Carlos III); Comunidad Autónoma de Madrid; La Caixa; Consejo Superior de Investigaciones Científicas; Work; MRC (G1100041; co-applicant IMA); an EU FP7 award (237946, T3Net); EU (FP7-integrated project Masterswicht 223404 FP7).Peer Reviewe

    Get PDF
A commented checklist containing 139 species of marine fishes recorded at the Galician Bank seamount is presented. The list is based on nine prospecting and research surveys carried out from 1980 to 2011 with different fishing gears. The ichthyofauna list is diversified in 2 superclasses, 3 classes, 20 orders, 62 families and 113 genera. The largest family is Macrouridae, with 9 species, followed by Moridae, Stomiidae and Sternoptychidae with 7 species each. The trachichthyd Hoplostethus mediterraneus and the morid Lepidion lepidion were the most abundant species. Biogeographically, the Atlantic group, with 113 species (81.3%) is the best represented, followed by the Lusitanian one with 17 species (12.2%). Data on species abundance, as number of individuals caught, size and depth are reported. Habitat, distribution and vulnerability status are commented. Moreover, biometric data and meristic counts are also reported for several species. The results obtained showing a high fish biodiversity and a sensible number of threatened species, strongly support the future declaration of the Galicia Bank as a Marine Protected Area.En prensa0,974

First recorded case of leucism in the velvet belly lantern shark Etmopterus spinax (Squaliformes: Etmopteridae)

The capture of a female specimen of the velvet belly lantern shark Etmopterus spinax with abnormal skin coloration is reported. The specimen was captured off northern Spain (Bay of Biscay, North-eastern Atlantic) in 2021 by bottom trawling. The lack of dermal melanophores combined with a normal retinal pigmentation, lead us to considerate the specimen as leucistic. This is the first case of leucism in E. spinax and the second record of colour aberration in the family Etmopteridae. A comprehensive updated review of published literature on albinism and leucism in chondrichthyans is also provided.En prens

This paper reports a new population of Bulinus truncatus (Audouin, 1827) (Gastropoda, Planorbidae) found in the province of Almería (Southeast Spain). B. truncatus is an intermediate host of Schistosoma haematobium, the trematode which causes urinary schistosomiasis in humans. Individuals were identified to species level by double-nested PCR, resulting in 100% homology. This population is located under the driest climate conditions of the Iberian Peninsula. Data on the distribution of this species in the Iberian Peninsula was gathered and is provided in this paper. Improved knowledge of the distribution of Bulinus truncatus is key to assess the risk of new outbreaks of schistosomiasis in the Iberian Peninsula.

Polyphosphate degradation by Nudt3-Zn²⁺ mediates oxidative stress response

    Get PDF
Polyphosphate (polyP) is a polymer of hundreds of phosphate residues present in all organisms. In mammals, polyP is involved in crucial physiological processes, including coagulation, inflammation, and stress response. However, after decades of research, the metabolic enzymes are still unknown. Here, we purify and identify Nudt3, a NUDIX family member, as the enzyme responsible for polyP phosphatase activity in mammalian cells. We show that Nudt3 shifts its substrate specificity depending on the cation; specifically, Nudt3 is active on polyP when Zn2+ is present. Nudt3 has in vivo polyP phosphatase activity in human cells, and importantly, we show that cells with altered polyP levels by modifying Nudt3 protein amount present reduced viability upon oxidative stress and increased DNA damage, suggesting that polyP and Nudt3 play a role in oxidative stress protection. Finally, we show that Nudt3 is involved in the early stages of embryo development in zebrafish

Automatic, efficient and scalable provenance registration for FAIR HPC workflows

    Get PDF
Provenance registration is becoming more and more important, as we increase the size and number of experiments performed using computers. In particular, when provenance is recorded in HPC environments, it must be efficient and scalable. In this paper, we propose a provenance registration method for scientific workflows, efficient enough to run in supercomputers (thus, it could run in other environments with more relaxed restrictions, such as distributed ones). It also must be scalable in order to deal with large workflows, that are more typically used in HPC. We also target transparency for the user, shielding them from having to specify how provenance must be recorded. We implement our design using the COMPSs programming model as a Workflow Management System (WfMS) and use RO-Crate as a well-established specification to record and publish provenance. Experiments are provided, demonstrating the run time efficiency and scalability of our solution. This work has been supported by the Spanish Government (PID2019-107255GB-C21), by Generalitat de Catalunya (contract 2017-SGR-01414) and the EU’s Horizon research and innovation programme under Grant agreement No 101058129 (DT-GEO). Updating the national checklist of marine fishes in Spanish waters: An approach to priority hotspots and lessons for conservation

    Get PDF
In response to a request from the Spanish Ministry of Farming, Fishing, Food, and Environment (Spanish: Agricultura, Pesca, Alimentación y Medio Ambiente) in 2015, a fish expert group was formed to provide a reference list of marine fish species according to five regions (marine demarcations) established by Spanish Law 41/2010 on the protection of the marine environment. The objective of this article was to update and analyse the data compiled in the marine fish species checklist in order to: 1) provide a complete list of marine fish species in the Spanish Exclusive Economic Zone; 2) compare this checklist between bio-geographical areas; and 3) identify possible priority hotspots for their conservation. We applied several indices, such as the total number of species in each area, species richness, and the Biodiversity Conservation Concern index. We discuss gaps in knowledge and the lessons learned for conservation purposes. A total of 1075 marine fishes were reported in Spanish waters. Most of these fish were well determined, whereas a few were treated as uncertain. The marine demarcation with the most species is the Canary Islands with 795 species, followed by the Spanish north coast demarcation with 506 species. However, the marine demarcations with the most species per area are the Spanish coast of the Gulf of Cádiz and the Strait of Gibraltar-Alboran Sea
