    Cooperation and virulence in acute Pseudomonas aeruginosa infections

    BACKGROUND: Efficient host exploitation by parasites is frequently likely to depend on cooperative behaviour. Under these conditions, mixed-strain infections are predicted to show lower virulence (host mortality) than are single-clone infections, due to competition favouring non-contributing social 'cheats' whose presence will reduce within-host growth. We tested this hypothesis using the cooperative production of iron-scavenging siderophores by the pathogenic bacterium Pseudomonas aeruginosa in an insect host. RESULTS: We found that infection by siderophore-producing bacteria (cooperators) results in more rapid host death than does infection by non-producers (cheats), and that mixtures of both result in intermediate levels of virulence. Within-host bacterial growth rates exhibited the same pattern. Crucially, cheats were more successful in mixed infections compared with single-clone infections, while the opposite was true of cooperators. CONCLUSION: These data demonstrate that mixed clone infections can favour the evolution of social cheats, and thus decrease virulence when parasite growth is dependent on cooperative behaviours

    High-Level Magnetic Activity on a Low-Mass Close Binary: GSC 02038-0293

    Taking into account results obtained from light-curve analysis and out-of-eclipse analyses, we discuss the nature of GSC 02038-00293 and also its magnetic activity behaviour. We obtained light curves of the system during observing seasons 2007, 2008 and 2011. We obtained its secondary minimum clearly in I-band observations in 2008 for the first time. Analysing this light curve, we found the physical parameters of the components. The light-curve analysis indicates that the possible mass ratio of the system is 0.35. We obtained the remaining V-band light curves, extracting the eclipses. We modelled these remaining curves using the spotmodel program and found possible spot configurations of the magnetically active component for each observing season. The models demonstrated that there are two active longitudes for the active component. The models reveal that both active longitudes migrate in the direction of decreasing longitude. We also examined the light curves in out-of-eclipse phases with respect to minimum and maximum brightness, amplitude, etc. The amplitude of the curves during out-of-eclipse phases varies in a sinusoidal way with a period of ~8.9yr the mean brightness of the system is dramatically decreasing. The phases of the deeper minimum during out-of-eclipse periods exhibit a migration toward decreasing phase.Comment: 15 pages, 8 figures, 5 table

    Trace Metal Chemistry in the Water Column of the Angola Basin - A Contribution to the International GEOTRACES Program - Cruise No. M121, November 22, – December 27, 2015, Walvis Bay (Namibia) – Walvis Bay (Namibia)

    Meteor Cruise M121 was dedicated to the investigation of the distribution of dissolved and particulate trace metals and their isotopic compositions (TEIs) in the full water column of the Angola Basin and the northernmost Cape Basin. A key aim was to determine the driving factors for the observed distributions, which includes the main external inputs, as well as internal cycling and ocean circulation. The research program of the cruise is official part of the international GEOTRACES program (www.geotraces.org) and cruise M121 corresponds to GEOTRACES cruise GA11. Subject of the cruise was the trace metal clean and contamination-free sampling of waters and particulates for subsequent analyses of the TEIs in the home laboratories of the national and international participants. Besides a standard rosette for the less contaminant prone metals, trace metal clean sampling was realized by using for the first time a new dedicated, coated trace metal clean rosette equipped with Teflon-coated GO-FLO bottles operated via a plastic coated cable from a mobile winch of GEOMAR Kiel. The particulate samples were collected under trace metal clean conditions using established in-situ pump systems operated from Meteor’s Aramid line. The cruise track led from Walvis Bay northwards along the West African margin until 3°S, then turned west until the Zero Meridian, which was followed southwards until 30°S. Then the cruise track turned east again until the Namibian margin was reached and then completed the near shore track northwards until Walvis Bay. The track crossed areas of major external inputs including dust from the Namib Desert and exchange with the west African continental margin and with the oxygen depleted shelf sediments of the Benguela upwelling, as well as with the plume of the Congo outflow, that was followed from its mouth northwards. Our investigations of internal cycling included the extremely high productivity associated with the Benguela Upwelling and the elevated productivity of the Congo plume contrasting with the extremely oligotrophic waters of the southeastern Atlantic Gyre. The links between TEI biogeochemistry and the nitrogen cycle forms an important aspect of our study. The major water masses contributing the Atlantic Meridional Overturning Circulation were sampled in order to investigate if particular TEI signatures are suitable as water mass tracers, in particular near the ocean margin and in the restricted deep Angola Basin. A total of 51 full water column stations were sampled for the different dissolved TEIs, which were in most cases accompanied by sampling for particulates and radium isotopes using the in-situ pumps. In addition, surface waters were continuously sampled under trace metal clean conditions using a towed fish and aerosol and rain samples were continuously collected

    A Novel Tandem Mass Spectrometry Method for Rapid Confirmation of Medium- and Very Long-Chain acyl-CoA Dehydrogenase Deficiency in Newborns

    BACKGROUND:Newborn screening for medium- and very long-chain acyl-CoA dehydrogenase (MCAD and VLCAD, respectively) deficiency, using acylcarnitine profiling with tandem mass spectrometry, has increased the number of patients with fatty acid oxidation disorders due to the identification of additional milder, and so far silent, phenotypes. However, especially for VLCADD, the acylcarnitine profile can not constitute the sole parameter in order to reliably confirm disease. Therefore, we developed a new liquid chromatography tandem mass spectrometry (LC-MS/MS) method to rapidly determine both MCAD- and/or VLCAD-activity in human lymphocytes in order to confirm diagnosis. METHODOLOGY:LC-MS/MS was used to measure MCAD- or VLCAD-catalyzed production of enoyl-CoA and hydroxyacyl-CoA, in human lymphocytes. PRINCIPAL FINDINGS:VLCAD activity in controls was 6.95+/-0.42 mU/mg (range 1.95 to 11.91 mU/mg). Residual VLCAD activity of 4 patients with confirmed VLCAD-deficiency was between 0.3 and 1.1%. Heterozygous ACADVL mutation carriers showed residual VLCAD activities of 23.7 to 54.2%. MCAD activity in controls was 2.38+/-0.18 mU/mg. In total, 28 patients with suspected MCAD-deficiency were assayed. Nearly all patients with residual MCAD activities below 2.5% were homozygous 985A>G carriers. MCAD-deficient patients with one other than the 985A>G mutation had higher MCAD residual activities, ranging from 5.7 to 13.9%. All patients with the 199T>C mutation had residual activities above 10%. CONCLUSIONS:Our newly developed LC-MS/MS method is able to provide ample sensitivity to correctly and rapidly determine MCAD and VLCAD residual activity in human lymphocytes. Importantly, based on measured MCAD residual activities in correlation with genotype, new insights were obtained on the expected clinical phenotype
