    Artificial co-drivers as a universal enabling technology for future intelligent vehicles and transportation systems

    This position paper introduces the concept of artificial “co-drivers” as an enabling technology for future intelligent transportation systems. In Sections I and II, the design principles of co-drivers are introduced and framed within general human–robot interactions. Several contributing theories and technologies are reviewed, specifically those relating to relevant cognitive architectures, human-like sensory-motor strategies, and the emulation theory of cognition. In Sections III and IV, we present the co-driver developed for the EU project interactIVe as an example instantiation of this notion, demonstrating how it conforms to the given guidelines. We also present substantive experimental results and clarify the limitations and performance of the current implementation. In Sections IV and V, we analyze the impact of the co-driver technology. In particular, we identify a range of application fields, showing how it constitutes a universal enabling technology for both smart vehicles and cooperative systems, and naturally sets out a program for future research

Abstract To define the time course and potential effects of electrical stimulation on permanently denervated muscle, we evaluated excitation-contraction coupling (ECC) of rat leg muscles during progression to long-term denervation by ultrastructural analysis, specific binding to dihydropyridine receptors, ryanodine receptor 1 (RYR-1), Ca channels and extrusion Ca pumps, gene transcription and translation of Ca-handling proteins, and in vitro mechanical properties and electrophysiological analyses of sarcolemmal passive properties and L-type Ca current (ICa) parameters. We found that in response to long-term denervation: 1) isolated muscle that is unable to twitch in vitro by electrical stimulation has very small myofibers but may show a slow caffeine contracture; 2) only roughly half of the muscle fibers with "voltage-dependent Ca channel activity" are able to contract; 3) the ECC mechanisms are still present and, in part, functional; 4)ECC-related gene expression is upregulated; and 5) at any time point, there are muscle fibers that are more resistant than others to denervation atrophy and disorganization of the ECC apparatus. These results support the hypothesis that prolonged "resting" [Ca] may drive progression of muscle atrophy to degeneration and that electrical stimulation-induced [Ca] modulation may mimic the lost nerve influence, playing a key role in modifying the gene expression of denervated muscle. Hence, these data provide a potential molecular explanation for the muscle recovery that occurs in response to rehabilitation strategies developed based on empirical clinical observations.     Observation of the moon shadow using a new reconstruction technique in the CLUE experiment

    The CLUE experiment, located in La Palma island at 2200 m a.s.l., is an array of 3×3 telescope, detecting the UV (190–230 nm) ˇCerenkov light produced by atmospheric showers. Due to the higher atmospheric absorption in the UV range than in the visible one, CLUE cannot apply existing algorithms normally used in IACT experiments to determine primary cosmic ray direction. In this paper we present a new method developed by CLUE. The algorithm performances were evaluated using simulated showers. CLUE experiment collected data in the last two years pointing to AGN sources and to Moon. The preliminary results obtained using the new technique on Crab Nebula and on Markarian 421 were presented in a previous paper. Here, we present the preliminary observation of Moon Shadow employing the new method. As described in the paper, we expect in a near future improvements on AGN sources and on Moon Shadow measurement

    Correction of beta-thalassemia major by gene transfer in haematopoietic progenitors of pediatric patients

    Beta-thalassemia is a common monogenic disorder due to mutations in the beta-globin gene and gene therapy, based on autologous transplantation of genetically corrected haematopoietic stem cells (HSCs), holds the promise to treat patients lacking a compatible bone marrow (BM) donor. We recently showed correction of murine beta-thalassemia by gene transfer in HSCs with the GLOBE lentiviral vector (LV), expressing a transcriptionally regulated human beta-globin gene. Here, we report successful correction of thalassemia major in human cells, by studying a large cohort of pediatric patients of diverse ethnic origin, carriers of different mutations and all candidates to BM transplantation. Extensive characterization of BM-derived CD34(+) cells before and following gene transfer shows the achievement of high frequency of transduction, restoration of haemoglobin A synthesis, rescue from apoptosis and correction of ineffective erythropoiesis. The procedure does not significantly affect the differentiating potential and the relative proportion of haematopoietic progenitors. Analysis of vector integrations shows preferential targeting of transcriptionally active regions, without bias for cancer-related genes. Overall, these results provide a solid rationale for a future clinical translation

    The largest multicentre data collection on prepectoral breast reconstruction: The iBAG study

    Background and Objectives: In the last years, prepectoral breast reconstruction has increased its popularity, becoming a standard reconstructive technique by preserving pectoralis major anatomy and functionality. Nevertheless, the lack of solid and extensive data negatively impacts on surgeons\u2019 correct information about postoperative complication rates and proper patient selection. This study aims to collect the largest evidence on this procedure. Methods: A multicentre retrospective audit, promoted by the Barcelona Hospital, collected the experience of 30 centers on prepectoral breast reconstruction with Braxon ADM. The study had the scientific support of INPECS and IIB societies which provided the online database Clinapsis. Results: A total of 1450 procedures were retrospectively collected in a 6-year period. Mean age 52.4 years, BMI 23.9, follow-up 22.7 months. Reconstruction was carried out after a tumor in 77.1% of the cases, 20.1% had prophylactic surgery, 2.8% had revisions. Diabetes, smoke, and immunosuppression had an influence on complications occurrence, as well as implant weight. Capsular contracture was associated with postoperative radiotherapy, but the overall rate was low (2.1%). Complications led to implant loss in 6.5% of the cases. Conclusions: The international Braxon Audit Group multicentre data collection represents a milestone in the field of breast reconstruction, extensively improving the knowledge of this procedure
