    Transcriptional modulation of mitochondria biogenesis pathway at and above critical speed in mice

    International audienceHigh- or moderate-intensity endurance training leads to mitochondrial biogenesis via the peroxisome proliferator-activated receptor γ co-activator 1α (PGC-1α)/mitochondrial transcription factor A (Tfam) signaling pathway. Although this pathway is stimulated during acute exercise, the relationship between its activity and the intensity of the exercise has not been characterized. In animal studies, individualized running speeds have not previously been assessed. Here, we sought to determine whether this pathway was modulated after a bout of exhaustive exercise at different relative intensities (at and over critical speed (CS)). Our starting hypotheses were that (i) exercise-induced overexpression of PGC-1α in skeletal muscle falls at intensities above CS, and (ii) transcriptional activity of the mitochondrial biogenesis signaling cascade is intensity-sensitive at and above CS. To test these hypothesis, male Friend Virus B-Type mice were divided into a control group and three exercise groups (exercising at CS, peak velocity (vPeak) and 150 % CS, respectively). mRNA expression levels for genes involved in mitochondrial biogenesis signaling were analyzed in the quadriceps muscle. PGC-1α was overexpressed at all exercise intensities. We also identified that, PGC-1α mRNA expression was negatively correlated with exercise intensity and blood lactate levels but not with maximal oxygen uptake, vPeak, or CS. Expression of the PGC-1α co-activator peroxisome proliferator-activated receptor β was negatively correlated with the exercise intensity. In contrast, expression levels of Tfam were dissociated from exercise intensity. Our data indicate that at the intensities used in endurance training, the expression of mitochondrial biogenesis genes is finely modulated by the relative intensity of exhaustive exercise. © 2015, Springer Science+Business Media New York

    Physical performance level in sarcomeric mitochondria creatine kinase knockout mouse model throughout ageing

    International audiencePurpose: The objective of the present study was to establish the role of sarcomeric mitochondrial creatine kinase (Mt-CK) in muscle energy output during exercise in a murine model of ageing (the Mt-CK knock-out mouse, Mt-CK-/-).Methods: Three age groups of Mt-CK-/- mice and control male mice (6, 9, and 18 months of age) underwent incremental treadmill running tests. The maximum speed (Vpeak) and maximal oxygen consumption (VO2 peak) values were recorded. Urine samples were analyzed using metabolomic techniques. The skeletal muscle (quadriceps) expression of proteins involved in mitochondria biogenesis, peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1 alpha) and dynamin-related GTPase mitofusin 2 (Mnf2) were quantified.Results: The VO(2)peak (normalized to heart weight: HW) of 18-month-old (mo) Mt-CK-/- mice was 27% (p < 0.001) lower than in 18-mo control mice. The VO(2)peak/HW ratio was 29% (p < 0.001) lower in 18-mo Mt-CK-/- mice than in 6-mo (p < 0.001) and 32% (p < 0.001) than 9-mo Mt-CK-/- mice. With a 0 degrees slope, Vpeak was 10% (p < 0.05) lower in 18-mo Mt-CK-/- mice than in 6-mo Mt-CK-/- mice but did not differ when comparing the 18-mo and 6-mo control groups. The skeletal muscles weight normalized on body weight in 6-mo Mt-CK-/- were 13 to 14% (p < 0.001, p < 0.05) lower versus the 6-mo control, in addition, the presence of branched-chain amino acids in the urine of 6-mo Mt-CK-/- mice suggests an imbalance in protein turnover (catabolism rather than anabolism) but we did not observe any age-related differences. The expression of PGC-1 alpha and Mnf2 proteins in the quadriceps showed that age-related effects were more prominent than genotype effects.Conclusion: The present study showed ageing is potentialized by Mt-CK deficiency with regard to VO(2)peak, Vpeak and mitochondrial protein expression. Our results support that Mt-CK-/- mice undergo physiological adaptations, enabling them to survive and to perform as well as wild-type mice. Furthermore, it is possible that these adaptations in Mt-CK-/- mice have a high energy cost and might trigger premature ageing

    Os objetivos do estudo foram verificar a associação entre os métodos de quantificação da carga de treinamento, PSE da sessão e impulso de treinamento proposto por Lucía (LuciaTRIMP) e descrever a distribuição da intensidade de treinamento no futsal semiprofissional da categoria sub 18. Quinze jogadores (média e DP; idade: 17 ± 1 ano; massa corporal: 70,4 ± 6,5 kg; estatura: 175 ± 4 cm e IMC 23 ± 2 kg.m-2) foram submetidos a um teste de corrida incremental para determinar o limiar ventilatório (LVmax), o ponto de compensação respiratória (PCR) e o consumo máximo de oxigênio (VO2max). Apenas oito atletas foram monitorados por meio dos métodos PSE da sessão e LuciaTRIMP durante 78 sessões de treinamento antes da principal competição da temporada. Foram encontradas correlações individuais de magnitudes alta a quase perfeita (r = 0,64 - 0,91) entre os métodos PSE da sessão e LuciaTRIMP. O padrão de distribuição da intensidade de treinamento foi 76, 18 e 6% do tempo abaixo do LV (zona 1), entre LV e PCR (zona 2) e acima do PCR (zona 3), respectivamente. O método PSE da sessão se mostrou altamente relacionado com um método objetivo de quantificação da carga de treinamento. Além disso, as sessões de treinamento de uma equipe de futsal semiprofissional da categoria sub 18 apresentaram um padrão decrescente de distribuição das intensidades de treinamento.<br>The aims of this study were to verify the association between the methods of training load quantification session-RPE and training impulse -proposed by Lucia (LuciaTRIMP) -and to describe the training intensity distribution in under-18 semi-professional futsal. Fifteen male players (mean and SD; age: 17 ± 1 years; body mass: 70.4 ± 6.5 kg; height: 175 ± 4 cm and BMI 23 ± 2 kg.m²) performed an incremental treadmill test to determine the ventilatory threshold (VT), respiratory compensation point (RCP), and maximal oxygen consumption (VO2max). Only eight athletes had their training loads quantified by means of session-RPE and Lucia methods over a total of 78 training sessions before the main competition of the season. High to almost perfect correlations (r = 0.64 - 0.91) were found between session-RPE and LuciaTRIMP methods. The distribution of the training intensity was, respectively, 76, 18 e 6% of training duration below VT (zone 1), between VT and RCP (zone 2), and above RCP (zone 3). The session-RPE method was highly associated with an objective method of training load quantification. Furthermore, the training sessions of a semi-professional under-18 futsal team showed a decreasing training intensity distribution

    O treinamento físico é uma importante área de atuação profissional da Educação Física e do Esporte. Ela tem por objetivo precípuo, a melhoria do desempenho físico-esportivo através da aplicação de um processo organizado e sistemático composto por exercícios físicos. Nos últimos anos, os progressos tecnológicos e nos métodos de investigação científica nas diferentes subáreas relacionadas ao treinamento físico trouxeram um avanço significativo na obtenção deste objetivo. Neste artigo será discutido, do ponto de vista acadêmico-científico e também da prática profissional, o estado da arte do conhecimento associado à avaliação do treinamento, ao controle da carga de treinamento, aos modelos de organização da carga de treinamento e ao desenvolvimento das capacidades motoras. Esperamos que ao final, o leitor possa ter um bom entendimento destes diferentes componentes, como eles contribuem para a modificação do desempenho motor e como aplicá-los para a elaboração, implementação, avaliação e reformulação de programas de treinamento físico.<br>Physical training is an important field of work for the Physical Education and Sport professional. Its main goal is to develop the physical capacity and sports performance of an individual by means of an organized and systematic application of physical exercises. In the past years, the technological progress in the scientific investigation within the different disciplines related to physical training have contributed a great deal towards the accomplishment of such goal. In this manuscript we discuss, from both an academic-scientific and from a practical application point-of-view, the state of the art of the knowledge associated with the physical training evaluation, loading control, loading scheming, and the development of physical abilities. Our goal is to provide a comprehensive overview of the different components of the physical training and how they contribute to the development of the physical performance as well as how to apply these concepts in the elaboration, implementation, and evaluation of physical training programs
