    In-vitro demonstration of cell-mediated immunity to vaccinia virus in man

    Cell mediated immunity to vaccinia virus in man was studied by lymphocyte transformation. Vaccinia antigen, propagated on BHK-21 and Vero cells, could be used successfully for in-vitro testing after partial purification as well as crude infectious homogenates. Vaccinia antigen preparations were effective both in the infective and the inactivated state. Inactivation was usually accompanied with a certain loss of stimulating activity. Development of cell mediated immune response in-vitro after first vaccination was investigated in 17 adults. Vaccinia virus specific lymphocyte transformation was seen in the second week after vaccination in all cases. Following revaccination no increase of lymphocyte transformation ratio could be observed in 11 persons studied. At the same time the titers of humoral antibodies were elevated

    Cortical correlates of the basic and first harmonic frequency of Parkinsonian tremor

    Objective It has been hypothesized that the basic and first harmonic frequency of Parkinsonian tremor are somewhat independent oscillations the biological basis of which remains unclear. Methods We recorded 64-channel EEG in parallel with EMG of the forearm muscles most affected by rest tremor in 21 PD patients. EMG power spectrum, corticomuscular coherence spectra and EEG power spectra for each EEG electrode were calculated. The dynamics of the coherence and relative EMG and EEG power at the basic (tremor) frequency were calculated by a sliding, overlapping window analysis. Corticomuscular delays and direction of interaction were analysed by the maximizing coherence method for narrow band signals. Results The contralateral EEG electrodes with maximal coherence were different for the basic and first harmonic frequency. The dynamical coherence curves showed non-parallel time courses for the two frequencies. The mean EEG-EMG and EMG-EEG delays were all around 15–20 ms but significantly longer for the first harmonic than for the basic frequency. Conclusions Our data indicate different cortical representations and corticomuscular interaction of the basic and first harmonic frequencies of Parkinsonian tremor. Significance Separate central generators seem to contribute to the tremor via different pathways. Further studies on this complex tremor network are warranted

    Oxidative Stress and NO Signalling in the Root Apex as an Early Response to Changes in Gravity Conditions.

    Oxygen influx showed an asymmetry in the transition zone of the root apex when roots were placed horizontally on ground. The influx increased only in the upper side, while no changes were detected in the division and in the elongation zone. Nitric oxide (NO) was also monitored after gravistimulation, revealing a sudden burst only in the transition zone. In order to confirm these results in real microgravity conditions, experiments have been set up by using parabolic flights and drop tower. The production of reactive oxygen species (ROS) was also monitored. Oxygen, NO, and ROS were continuously monitored during normal and hyper- and microgravity conditions in roots of maize seedlings. A distinct signal in oxygen and NO fluxes was clearly detected only in the apex zone during microgravity, with no significant changes in normal and in hypergravity conditions. The same results were obtained by ROS measurement. The detrimental effect of D’orenone, disrupting the polarised auxin transport, on the onset of the oxygen peaks during the microgravity period was also evaluated. Results indicates an active role of NO and ROS as messengers during the gravitropic response, with probable implications in the auxin redistribution

    Cultivating compliance: governance of North Indian organic basmati smallholders in a global value chain

    Focusing on a global value chain (GVC) for organic basmati rice, we study how farmers’ practices are governed through product and process standards, organic certification protocols, and contracts with buyer firms. We analyze how farmers’ entry into the GVC reconfigures their agencements (defined as heterogeneous arrangements of human and nonhuman agencies which are associated with each other). These reconfigurations entail the severance of some associations among procedural and material elements of the agencements and the formation of new associations, in order to produce cultivation practices that are accurately described by the GVC’s standards and protocols. Based on ethnography of two farmers in Uttarakhand, North India, we find that the same standards were enacted differently on the two farmers’ fields, producing variable degrees of (selective) compliance with the ‘official’ GVC standards. We argue that the disjuncture between the ‘official’ scripts of the standards and actual cultivation practices must be nurtured to allow farmers’ agencements to align their practices with local sociotechnical relations and farm ecology. Furthermore, we find that compliance and disjuncture were facilitated by many practices and associations that were officially ungoverned by the GVC
