    Overall animal welfare assessment reviewed. Part 1: Is it possible?

    Several authors have concluded that scientists should not attempt to perform overall animal welfare assessment (OWA). They argue that scientists have continued to fail to make progress in this area and that value judgements are inherently involved in OWA for which science cannot provide answers. We take a more positive attitude toward OWA and argue that scientists should avoid creating a self-fulfilling prophecy. OWA is necessary for making actual moral and political decisions. Science has already accumulated much relevant information about welfare and this information should be applied in decision making. The task of OW A is to assess welfare based on knowledge of the biological needs of animals. Weighting of welfare relevant factors constitutes a problem. However, when scientists cannot provide empirical data to solve weighting issues, this does not mean that rational answers cannot be found, e.g. in the form of procedural rules. OWA is conceived as a problem of multi-criteria decision making with fuzzy information. It focuses on the descriptive aspect of welfare, i.e. on what the welfare status of the animals really is without taking an ethical stance. The welfare status of animals depends on their biology and on the way animals assess their own welfare. It does not depend on how it happens to be perceived by us. Even though OWA necessarily remains a human activity, it is not arbitrary, nor does it allow of multiple \u27correct\u27 answers.     In the scientific literature several attempts have been made to systematically assess the overall welfare-status of animals in relation to housing and management. This paper reviews assessment tables and schemes that have been constructed to this end. These tables and schemes have a tabular format that allows an assessment of housing systems using a list of welfare-relevant attributes (properties of the housing system). Rather than identifying deficits, the focus of this review is on finding positive recommendations for the purpose of developing a method for overall welfare assessment (OWA) on a scientific basis. The main recommendation is to use the tabular format as representation formalism for OWA. The concept of linked tables provides the key to performing OWA on a scientific basis in an explicit and systematic way

    Overall animal welfare reviewed. Part 3: Welfare assessment based on needs and supported by expert opinion

    Welfare concerns what matters to animals from their point of view. What matters to animals is their state of need. Satisfaction and frustration of needs are associated with emotional states, the subjective experience of which directly determines the welfare status of an animal. Because emotional states are difficult to assess, overall welfare assessment (OWA) is best approached as an assessment of needs. For actual OW A a list of needs must be formulated. Different authors have formulated different lists. From these lists a concept need-list was constructed. For validation the needs-based approach for OWA was discussed in interviews with experts (n = 21) in the field of ethology and other welfare related sciences. These experts generally used mental terminology to define welfare, but when asked to classify their definition of welfare, many preferred a definition in terms of measurable parameters or a combination of both mental terms (feelings) and measurables. Most experts believed that welfare can be assessed objectively and that the problem of OWA is indeed best approached through an assessment of needs. Experts differ as to the exact composition of the list of needs. A list of needs is formulated which we intend to use for OWA in the case of sows

    Linking Parenting and Social Competence in School-Aged Boys and Girls: Differential Socialization, Diathesis-Stress, or Differential Susceptibility?

    Girls generally demonstrate superior skill levels in social competence compared to boys. The exact relations of parenting with these gender differences are currently unclear. Gender differences may occur due to exposure to different parenting strategies (differential socialization model) or due to a different impact of similar parenting strategies for boys and girls (differential susceptibility and diathesis-stress model).Objective: In this study we assessed both hypotheses using a multi-method multi-informant approach. We investigated (1) to what extent different parenting strategies mediate the relation between gender and social competence and (2) whether gender and age moderate the relation between parenting strategies and social competence.Design: Parenting strategies were observed during home visits and social competence was assessed using parent and teacher questionnaires and performance-based neurocognitive tasks (N = 98, aged 4 to 8).Results: (1) Parenting strategies did not mediate the relation between gender and social competence. (2) Gender moderated the association between parental questioning style and children’s level of social competence: parents asking fewer questions was associated with poorer social cognitive skills in boys only. Parental supportive presence and intrusiveness were related to aspects of social competence irrespective of gender. Age moderated the relation between parenting and aspects of social competence, though in various (unexpected) directions.Conclusion: Our findings do not support the differential socialization hypothesis and provide partial evidence for a diathesis-stress model as an explanation for parental influence on gender differences in social competence

    During recent decades, bacteriophages have been at the cutting edge of new developments in molecular biology, biophysics, and, more recently, bionanotechnology. In particular filamentous viruses, for example bacteriophage M13, have a virion architecture that enables precision building of ordered and defect-free two and three-dimensional structures on a nanometre scale. This could not have been possible without detailed knowledge of coat protein structure and dynamics during the virus reproduction cycle. The results of the spectroscopic studies conducted in our group compellingly demonstrate a critical role of membrane embedment of the protein both during infectious entry of the virus into the host cell and during assembly of the new virion in the host membrane. The protein is effectively embedded in the membrane by a strong C-terminal interfacial anchor, which together with a simple tilt mechanism and a subtle structural adjustment of the extreme end of its N terminus provides favourable thermodynamical association of the protein in the lipid bilayer. This basic physicochemical rule cannot be violated and any new bionanotechnology that will emerge from bacteriophage M13 should take this into account

    Profiling of dynamics in protein–lipid–water systems: a time-resolved fluorescence study of a model membrane protein with the label BADAN at specific membrane depths

    Profiles of lipid-water bilayer dynamics were determined from picosecond time-resolved fluorescence spectra of membrane-embedded BADAN-labeled M13 coat protein. For this purpose, the protein was labeled at seven key positions. This places the label at well-defined locations from the water phase to the center of the hydrophobic acyl chain region of a phospholipid model membrane, providing us with a nanoscale ruler to map membranes. Analysis of the time-resolved fluorescence spectroscopic data provides the characteristic time constant for the twisting motion of the BADAN label, which is sensitive to the local flexibility of the protein–lipid environment. In addition, we obtain information about the mobility of water molecules at the membrane–water interface. The results provide an unprecedented nanoscale profiling of the dynamics and distribution of water in membrane systems. This information gives clear evidence that the actual barrier of membranes for ions and aqueous solvents is located at the region of carbonyl groups of the acyl chains

    Conformational studies of peptides representing a segment of TM7 from H+-VO-ATPase in SDS micelles

    The conformation of a transmembrane peptide, sMTM7, encompassing the cytoplasmic hemi-channel domain of the seventh transmembrane section of subunit a from V-ATPase from Saccharomyces cerevisiae solubilized in SDS solutions was studied by circular dichroism (CD) spectroscopy and fluorescence spectroscopy of the single tryptophan residue of this peptide. The results show that the peptide adopts an α-helical conformation or aggregated β-sheet depending on the peptide-to-SDS ratio used. The results are compared with published data about a longer version of the peptide (i.e., MTM7). It is concluded that the bulky, positively charged arginine residue located in the center of both peptides has a destabilizing effect on the helical conformation of the SDS-solubilized peptides, leading to β-sheet formation and subsequent aggregation

    SNP association study in PMS2-associated Lynch syndrome

    Lynch syndrome (LS) patients are at high risk of developing colorectal cancer (CRC). Phenotypic variability might in part be explained by common susceptibility loci identified in Genome Wide Association Studies (GWAS). Previous studies focused mostly on MLH1, MSH2 and MSH6 carriers, with conflicting results. We aimed to determine the role of GWAS SNPs in PMS2 mutation carriers. A cohort study was performed in 507 PMS2 carriers (124 CRC cases), genotyped for 24 GWAS SNPs, including SNPs at 11q23.1 and 8q23.3. Hazard ratios (HRs) were calculated using a weighted Cox regression analysis to correct for ascertainment bias. Discrimination was assessed with a concordance statistic in a bootstrap cross-validation procedure. Individual SNPs only had non-significant associations with CRC occurrence with HRs lower than 2, although male carriers of allele A at rs1321311 (6p21.31) may have increased risk of CRC (HR = 2.1, 95% CI 1.2–3.0). A polygenic risk score (PRS) based on 24 HRs had an HR of 2.6 (95% CI 1.5–4.6) for the highest compared to the lowest quartile, but had no discriminative ability (c statistic 0.52). Previously suggested SNPs do not modify CRC risk in PMS2 carriers. Future large studies are needed for improved risk stratification among Lynch syndrome patients

    The apparent genetic anticipation in PMS2-associated Lynch syndrome families is explained by birth cohort effect

    BACKGROUND: PMS2-associated Lynch syndrome is characterized by a relatively low colorectal cancer penetrance compared with other Lynch syndromes. However, age at colorectal cancer diagnosis varies widely, and a strong genetic anticipation effect has been suggested for PMS2 families. In this study, we examined proposed genetic anticipation in a sample of 152 European PMS2 families. METHODS: The 152 families (637 family members) that were eligible for analysis were mainly clinically ascertained via clinical genetics centers. We used weighted Cox-type random effects model, adjusted by birth cohort and sex, to estimate the generational effect on the age of onset of colorectal cancer. Probands and young birth cohorts were excluded from the analyses. Weights represented mutation probabilities based on kinship coefficients, thus avoiding testing bias. RESULTS: Family data across three generations, including 123 colorectal cancers, were analyzed. When compared with the first generation, the crude HR for anticipation was 2.242 [95% confidence interval (CI), 1.162-4.328] for the second generation and 2.644 (95% CI, 1.082-6.464) for the third generation. However, after correction for birth cohort and sex, the effect vanished [HR = 1.302 (95% CI, 0.648-2.619) and HR = 1.074 (95% CI, 0.406-2.842) for second and third generations, respectively]. CONCLUSIONS: Our study did not confirm previous reports of genetic anticipation in PMS2-associated Lynch syndrome. Birth-cohort effect seems the most likely explanation for observed younger colorectal cancer diagnosis in subsequent generations, particularly because there is currently no commonly accepted biological mechanism that could explain genetic anticipation in Lynch syndrome. IMPACT: This new model for studying genetic anticipation provides a standard for rigorous analysis of families with dominantly inherited cancer predisposition
