    Differentiation of definitive endoderm from human induced pluripotent stem cells on hMSCs feeder in a defined medium

    Background: The Definitive Endoderm (DE) differentiation using the undefined media and non-human feeders can cause contaminations in the generated cells for therapeutic applications. Therefore, generating safer and more appropriate DE cells is needed. This study compared five different methods to establish an appropriate method for inducing an efficient DE differentiation from Human Induced Pluripotent Stem Cells (hiPSCs) on an appropriate feeder in a more defined medium. Methods: Human Induced Pluripotent Stem Cells (hiPSCs) were cultured on inactivated feeders. Passaged hiPSCs, without feeder, were incubated for three days with Activin-A and different endodermal differentiation media including 1-FBS, 2-B27, 3- ITS and albumin fraction-V, 4-B27 and ITS and 5-like the third medium. The feeder cells in the first four methods were Mouse Embryonic Fibroblasts (MEFs) and in the fifth method were human adult bone marrow Mesenchymal Stem Cells (hMSCs). DE markers FOXA2, SOX17 and CXCR4 and also pluripotency marker OCT4 were evaluated using qRT-PCR, as well as FOXA2 by the immunocytochemistry. Results: QRT-PCR analysis showed that after three days, the expression levels of DE and pluripotency markers in the differentiated hiPSCs among all five groups did not have any significant differences. Similarly, the immunocytochemistry analysis demonstrated that the differentiated hiPSCs expressed FOXA2, with no significant differences. Conclusion: Despite this similarity in the results, the third differentiation medium has more defined and cost effective components. Furthermore, hMSC, a human feeder, is safer than MEF. Therefore, the fifth method is preferable among other DE differentiation methods and can serve as a fundamental method helping the development of regenerative medicine. © 2016, Avicenna Journal of Medical Biotechnology.     N-acetylcysteine compared to metformin, improves the expression profile of growth differentiation factor-9 and receptor tyrosine kinase c-kit in the oocytes of patients with polycystic ovarian syndrome

    Background: Paracrine disruption of growth factors in women with polycystic ovarian syndrome (PCOS) results in production of low quality oocyte, especially following ovulation induction. The aim of this study was to investigate the effects of metformin (MET), N-acetylcysteine (NAC) and their combination on the hormonal levels and expression profile of GDF-9, BMP-15 and c-kit, as hallmarks of oocyte quality, in PCOS patients. Materials and Methods: This prospective randomized, double-blind, placebo controlled trial aims to study the effects of MET, NAC and their combination (MET+NAC) on expression of GDF-9, BMP-15 and c-kit mRNA in oocytes [10 at the germinal vesicle (GV) stage, 10 at the MI stage, and 10 at the MII stage from per group] derived following ovulation induction in PCOS. Treatment was carried out for six weeks, starting on the third day of previous cycle until oocyte aspiration. The expression of GDF9, BMP15 and c-kit were determined by quantitative real time polymerase chain reaction (RT-qPCR) and western blot analysis. Data were analyzed with one-way ANOVA. Results: The follicular fluid (FF) level of c-kit protein significantly decreased in the NAC group compared to the other groups. Significant correlations were observed between the FF soluble c-kit protein with FF volume, androstenedione and estradiol. The GDF-9 expression in unfertilized mature oocytes were significantly higher in the NAC group compared to the other groups (P<0.001). Similar difference was not observed between the MET, NAC+MET and control groups. The c-kit expression in unfertilized mature oocytes were significantly lower in the NAC group compared to the other groups (P<0.001). Similar difference was not observed between the MET, NAC+MET and control groups (Registration number: IRCT201204159476N1).     Efficacy of lactobacilli to normalize production of corticosterone induced by unpleasant handling of broilers

    A study was conducted to investigate the effect of two Lactobacillus strains on hypothalamus pituitary adrenal (HPA) axis activity induced by supposedly unpleasant handling (UH) of broiler chickens. The three treatments were: (1) non-handled chicks fed basal diet (control); (2) unpleasantly handled-chicks fed basal diet (UH-BD) and (3) unpleasantly handled-chicks fed basal diet supplemented with the probiotic, lactobacilli (UH-BDL). Chicks were exposed to UH from days 1 to 21. Treatment UH-BDL received probiotics in their diet for the whole experimental period. Blood corticosterone (CS) concentrations were monitored at 14, 28, 35 and 42 days of age and selected caecal bacterial groups were enumerated in 14-day old birds. Unpleasant handling significantly increased blood CS concentrations at 14 and 28 days of age. Blood CS concentration decreased with age over the course of the experimental period. Lactobacillus supplementation did not reduce blood CS concentration in broilers in the UH-BDL treatment. Moreover, the UH treatment did not alter caecal bacterial numbers in the 14-day old broilers while the probiotic numerically increased total anaerobes and lactobacilli. Overall, it seems as if the efficacy of lactobacilli to modulate stress-related high HPA-axis activity depends on gastrointestinal tract microbial alteration. However, Lactobacillus salivarius subsp. salicinius JCM 1230 and L. agilis JCM 1048 were able to re-establish a proper microbial balance in the caecum of the chicken

    Evaluation and optimization of chitosan derivatives-based gene delivery system via kidney epithelial cells

    Purpose: Non-viral vectors have been widely proposed as safer alternatives to viral vectors, and cationic polymers have gained increasing attention because they can form self-assembly with DNA. Chitosan is also considered to be a good candidate for gene delivery systems, since it is already known as a biocompatible, biodegradable, and low toxic material with high cationic potential. However, low solubility and transfection efficiency need to be overcome prior to clinical trial. In this work, we focus on alkyl modified chitosan which might be useful in DNA condensing and efficient gene delivery. Methods: N, N- Diethyl N- Methyl (DEMC) and N- Triethyl Chitosan (TEC) were synthesized from chitosan polymer. In order to optimize the polymers for gene delivery, we used FITC-dextran (FD). Then the optimized polymer concentrations were used for gene delivery. Fluorescent microscope was used, in order to evaluate the polymers’ efficiency for gene delivery to human embryonic kidney epithelial cells (HEK 293T). Results: This modification increased chitosan’s positive charge, thus these chitosan derivatives spontaneously formed complexes with FD, green fluorescence protein plasmid DNA (pEGFP), red fluorescence protein plasmid DNA (pJred) and fluorescent labeled miRNA. Results gained from fluorescent microscope showed that TEC and DEMC were able to transfer FD, DNA and miRNA (micro RNA) to HEK cell line. Conclusion: We conclude that these chitosan derivatives present suitable characteristics to be used as non-viral gene delivery vectors to epithelial cells

    Selected microbial groups and short-chain fatty acids profile in a simulated chicken cecum supplemented with two strains of Lactobacillus.

    Among the bacterial fermentation end products in the chicken cecum, butyrate is of particular importance because of its nutritional properties for the epithelial cell and pathogen inhibitory effects in the gut. An in vitro experiment, operated with batch bioreactor, was conducted to quantify butyric-producing bacteria in a simulated broiler cecum supplemented with Lactobacillus salivarius ssp. salicinius JCM 1230 and Lactobacillus agilis JCM 1048 during 24 h of incubation. Selected bacterial species were determined by real-time PCR and short-chain fatty acids and lactate concentrations were monitored. The results showed that after 24 h of incubation, Lactobacillus supplementation significantly increased the number of lactobacilli, bifidobacteria and Faecalibacterium prausnitzii in medium containing cecal content and lactobacilli supple-mentation (Cc + L) compared with the control (Cc). Addition of lactobacilli did not alter Escherichia coli and Clostridium butyricum, whereas it significantly (P < 0.05) reduced Salmonella in treatment Cc + L compared with the Cc treatment. Propionate and butyrate formation were significantly (P < 0.05) increased in treatment Cc + L as compared with the Cc treatment. Lactate was only detected in treatment containing 2 Lactobacillus strains. After 24 h of incubation, acetate concentration significantly (P < 0.05) decreased in all treatments. It was suggested that lactate produced by Lactobacillus in the cecal content improved the growth of butyric producers such as F. prausnitzii, which significantly increased butyrate accumulation. Additionally, the results showed that butyrate and propionate inhibited Salmonella without influencing the E. coli profile

    Stress induced polarization of immune-neuroendocrine phenotypes in Gallus gallus

    Immune-neuroendocrine phenotypes (INPs) stand for population subgroups differing in immune-neuroendocrine interactions. While mammalian INPs have been characterized thoroughly in rats and humans, avian INPs were only recently described in Coturnix coturnix (quail). To assess the scope of this biological phenomenon, herein we characterized INPs in Gallus gallus (a domestic hen strain submitted to a very long history of strong selective breeding pressure) and evaluated whether a social chronic stress challenge modulates the individuals’ interplay affecting the INP subsets and distribution. Evaluating plasmatic basal corticosterone, interferon-γ and interleukin-4 concentrations, innate/acquired leukocyte ratio, PHA-P skin-swelling and induced antibody responses, two opposite INP profiles were found: LEWIS-like (15% of the population) and FISCHER-like (16%) hens. After chronic stress, an increment of about 12% in each polarized INP frequency was found at expenses of a reduction in the number of birds with intermediate responses. Results show that polarized INPs are also a phenomenon occurring in hens. The observed inter-individual variation suggest that, even after a considerable selection process, the population is still well prepared to deal with a variety of immune-neuroendocrine challenges. Stress promoted disruptive effects, leading to a more balanced INPs distribution, which represents a new substrate for challenging situations.Fil: Nazar, Franco Nicolas.     The purpose of this paper is to characterize the weak efficient solutions, the efficient solutions, and the isolated efficient solutions of a given vector optimization problem with finitely many convex objective functions and infinitely many convex constraints.     A multiphysics-based artificial neural networks model for atherosclerosis

    Atherosclerosis is a medical condition involving the hardening and/or thickening of arteries' walls. Mathematical multi-physics models have been developed to predict the development of atherosclerosis under different conditions. However, these models are typically computationally expensive. In this study, we used machine learning techniques, particularly artificial neural networks (ANN), to enhance the computational efficiency of these models. A database of multi-physics Finite Element Method (FEM) simulations was created and used for training and validating an ANN model. The model is capable of quick and accurate prediction of atherosclerosis development. A remarkable computational gain is obtained using the ANN model compared to the original FEM simulations

    On the Image Reconstruction of Capacitively Coupled Electrical Resistance Tomography (CCERT) with Entropy Priors

    Regularization with priors is an effective approach to solve the ill-posed inverse problem of electrical tomography. Entropy priors have been proven to be promising in radiation tomography but have received less attention in the literature of electrical tomography. This work aims to investigate the image reconstruction of capacitively coupled electrical resistance tomography (CCERT) with entropy priors. Four types of entropy priors are introduced, including the image entropy, the projection entropy, the image-projection joint entropy, and the cross-entropy between the measurement projection and the forward projection. Correspondingly, objective functions with the four entropy priors are developed, where the first three are implemented under the maximum entropy strategy and the last one is implemented under the minimum cross-entropy strategy. Linear back-projection is adopted to obtain the initial image. The steepest descent method is utilized to optimize the objective function and obtain the final image. Experimental results show that the four entropy priors are effective in regularization of the ill-posed inverse problem of CCERT to obtain a reasonable solution. Compared with the initial image obtained by linear back projection, all the four entropy priors make sense in improving the image quality. Results also indicate that cross-entropy has the best performance among the four entropy priors in the image reconstruction of CCERT
