    Inhomogeneous Gain Saturation in EDF: Experiment and Modeling

    Erbium-Doped Fiber Amplifiers can present holes in spectral gain in Wavelength Division Multiplexing operation. The origin of this inhomogeneous saturation behavior is still a subject of controversy. In this paper we present both an experimental methods and a gain's model. Our experimental method allow us to measure the first homogeneous linewidth of the 1.5 Ό\mum erbium emission with gain spectral hole burning consistently with the other measurement in the literature and the model explains the differences observed in literature between GSHB and other measurement methods

    Spectroscopic signature of phosphate crystallization in Erbium-doped optical fibre preforms

    In rare-earth-doped silica optical fibres, the homogeneous distribution of amplifying ions and part of their spectroscopic properties are usually improved by adding selected elements, such as phosphorus or aluminum, as structural modifier. In erbium ion (Er3+) doped fibres, phosphorus preferentially coordinates to Er3+ ions to form regular cages around it. However, the crystalline structures described in literature never gave particular spectroscopic signature. In this article, we report emission and excitation spectra of Er3+ in a transparent phosphorus-doped silica fibre preform. The observed line features observed at room and low temperature are attributed to ErPO4 crystallites

    Virus-like particle vaccines against BK and JC polyomaviruses

    Nearly all healthy adults are asymptomatically infected with human polyomaviruses. In immunosuppressed individuals, the infection can reactivate and cause disease. BK polyomavirus (BKV) frequently damages transplanted kidneys and causes severe bladder disease in bone marrow transplant patients. JC polyomavirus (JCV) causes a lethal brain disease, PML, in individuals on various immunosuppressive therapies. PML also affects immunodeficient individuals, including AIDS patients. The outer capsid proteins of polyomaviruses are structurally similar to the capsids of human papillomaviruses (HPVs). Building on the success of the NCI’s HPV virus-like particle (VLP) vaccine technologies, we have developed VLP vaccines targeting BKV and JCV. Preclinical testing in a monkey model indicates that the BKV and JCV VLP vaccines share the HPV vaccines’ exceptionally potent immunogenicity. Given our knowledge of the role that antibodies play in ameliorating polyomavirus pathologies, the new VLP vaccines are likely to protect at-risk patients against the development of BKV-induced urinary tract disease and JCV-induced brain disease. Each year, roughly 30,000 Americans join wait-lists for kidney transplantation. Additionally, roughly 300,000 Americans per year are diagnosed with diseases that might be treated with bone marrow transplantation. Emerging evidence indicates that antibody-producing plasma cells elicited by the BKV vaccine will persist after bone marrow transplantation and the vaccine should thus provide protection against post-transplant hemorrhagic cystitis. The highly effective multiple sclerosis therapy Tysabri (natalizumab) is associated with up to 2% risk of PML side effects. Rituxan (rituximab), which is used for treatment of rheumatoid arthritis and certain types of lymphoma, carries a black box warning for PML and a dozen additional immunosuppressive therapies are also known or suspected to have PML side effects. The JCV vaccine should be a useful preventive adjunct for these popular immunotherapies. Since there are currently no effective treatments for BKV or JCV diseases, the candidate vaccines seem likely to qualify for FDA’s Accelerated Approval Program.     Effect of Chemically Induced Hypoxia on Osteogenic and Angiogenic Differentiation of Bone Marrow Mesenchymal Stem Cells and Human Umbilical Vein Endothelial Cells in Direct Coculture

    Bone is an active tissue where bone mineralization and resorption occur simultaneously. In the case of fracture, there are numerous factors required to facilitate bone healing including precursor cells and blood vessels. To evaluate the interaction between bone marrow-derived mesenchymal stem cells (BMSC)-the precursor cells able to differentiate into bone-forming cells and human umbilical vein endothelial cells (HUVEC)-a cell source widely used for the study of blood vessels. We performed direct coculture of BMSC and HUVEC in normoxia and chemically induced hypoxia using Cobalt(II) chloride and Dimethyloxaloylglycine and in the condition where oxygen level was maintained at 1% as well. Cell proliferation was analyzed by crystal violet staining. Osteogenesis was examined by Alizarin Red and Collagen type I staining. Expression of angiogenic factor-vascular endothelial growth factor (VEGF) and endothelial marker-von Willebrand factor (VWF) were demonstrated by immunohistochemistry and enzyme-linked immunosorbent assay. The quantitative polymerase chain reaction was also used to evaluate gene expression. The results showed that coculture in normoxia could retain both osteogenic differentiation and endothelial markers while hypoxic condition limits cell proliferation and osteogenesis but favors the angiogenic function even after 1 of day treatment

    Osteoporosis and Bisphosphonate-Related Osteonecrosis of the Jaw Bone

    The aim of this longitudinal study is to present data from 76 female patients treated with bisphosphonates (BPs) for postmenopausal osteoporosis and referred to the Unit of Oral Diagnosis and Day Surgery of the University of Milano for diagnosis and treatment. All patients received a thorough oral examination. The diagnosis of osteonecrosis of the jaw bone (ONJ) was made from radiographic and clinical findings. 9% of individuals had BRONJ at first visit. Patients with dental or periodontal abscess were significantly more likely to develop BRONJ (OR: 2.9, 95% CI 0.5–15.9). Patients with osteoporosis receiving BPs may develop BRONJ, especially in the presence of an active infectious process in the mouth. Clinicians should carefully follow up on individuals receiving bisphosphonates therapy to avoid the occurrence of osteonecrotic lesions

    SOI-based micro-mechanical terahertz detector operating at room-temperature

    We present a micro-mechanical terahertz (THz) detector fabricated on a silicon on insulator (SOI) substrate and operating at room-temperature. The device is based on a U-shaped cantilever of micrometric size, on top of which two aluminum half-wave dipole antennas are deposited. This produces an absorption extending over the ∌2−3.5\sim 2-3.5THz frequency range. Due to the different thermal expansion coefficients of silicon and aluminum, the absorbed radiation induces a deformation of the cantilever, which is read out optically using a 1.5ÎŒ1.5\mum laser diode. By illuminating the detector with an amplitude modulated, 2.5 THz quantum cascade laser, we obtain, at room-temperature and atmospheric pressure, a responsivity of ∌1.5×108\sim 1.5 \times 10^{8}pm/W for the fundamental mechanical bending mode of the cantilever. This yields an noise-equivalent-power of 20 nW/Hz1/2^{1/2} at 2.5THz. Finally, the low mechanical quality factor of the mode grants a broad frequency response of approximately 150kHz bandwidth, with a response time of ∌2.5ÎŒ\sim 2.5\mus

    ATN profile classification across two independent prospective cohorts

    BACKGROUND The ATN model represents a research framework used to describe in subjects the presence or absence of Alzheimer's disease (AD) pathology through biomarkers. The aim of this study was to describe the prevalence of different ATN profiles using quantitative imaging biomarkers in two independent cohorts, and to evaluate the pertinence of ATN biomarkers to identify comparable populations across independent cohorts. METHODS A total of 172 subjects from the Geneva Memory Clinic and 113 volunteers from a study on healthy aging at the University Hospital of Zurich underwent amyloid (A) and tau (T) PET, as well as T1-weigthed MRI scans using site-specific protocols. Subjects were classified by cognition (cognitively unimpaired, CU, or impaired, CI) based on clinical assessment by experts. Amyloid data converted into the standardized centiloid scale, tau PET data normalized to cerebellar uptake, and hippocampal volume expressed as a ratio over total intracranial volume ratio were considered as biomarkers for A, T, and neurodegeneration (N), respectively. Positivity for each biomarker was defined based on previously published thresholds. Subjects were then classified according to the ATN model. Differences among profiles were tested using Kruskal-Wallis ANOVA, and between cohorts using Wilcoxon tests. RESULTS Twenty-nine percent of subjects from the Geneva cohorts were classified with a normal (A-T-N-) profile, while the Zurich cohort included 64% of subjects in the same category. Meanwhile, 63% of the Geneva and 16% of the Zurich cohort were classified within the AD continuum (being A+ regardless of other biomarkers' statuses). Within cohorts, ATN profiles were significantly different for age and mini-mental state examination scores, but not for years of education. Age was not significantly different between cohorts. In general, imaging A and T biomarkers were significantly different between cohorts, but they were no longer significantly different when stratifying the cohorts by ATN profile. N was not significantly different between cohorts. CONCLUSION Stratifying subjects into ATN profiles provides comparable groups of subjects even when individual recruitment followed different criteria

    Conservation actions for restoring the coastal lagoon habitats: Strategy and multidisciplinary approach of LIFE Lagoon Refresh

    The Habitat Directive of European Union lists Costal Lagoons (habitat code 1150*) among priority habitats because they are in danger of disappearance. Natural ecosystems may recover from anthropogenic perturbations; however, the recovery can follow natural restoration or it can be redirected through ecological restoration by anthropogenic intervention. Accordingly, by collecting the available theoretical indications for restoration of estuarine and coastal areas, a methodological approach was detailed andit can be summarised into five issues: (i) Environmental context from which it began; (ii) Desired state to be achieved; (iii) Policies and socio-economic context; (iv) Typology of recovery and/or improvement of habitats and ecosystems; and (v) Methods for monitoring the impact of the project. The project strategy, management and measures of LIFE Lagoon Refresh were also presented and discussed, as a case study for the implementation of the multidisciplinary approach for restoration ecology in transitional waters. The project takes place in the northern Venice Lagoon (Italy), started in 2017 and it lasts 5 years. In the Venice Lagoon, since the 20th century, strong reductions of the typical salinity gradient of buffer areas between lagoon and mainland, and of reedbed extensions have occurred due to historic human interventions, with negative consequences on coastal lagoon habitats. To improve the conservation status of habitats and biodiversity of the area, the LIFE Lagoon Refresh project included several conservative actions, which are (i) the diversion of a freshwater flow from the Sile River into the lagoon; (ii) the restoration of intertidal morphology, through biodegradable structures; (iii) the reed and aquatic angiosperm transplantations with the involvement of local fishermen and hunters, and (iv) the reduction of hunting and fishing pressures in the intervention area. To achieve the restoration of the lagoon environment, the strategy of the project covered a combination of different aspects and tools, such as planning activities, through the involvement of local Institutions and communities; stakeholder's involvement to increase awareness of environment conservation and socioeconomic value improvement; an ecological engineering approach; numerical models as supporting tool for planning and managing of conservation actions; environmental monitoring performed before and after the conservation actions
