    The Transcriptome of the Human Pathogen Trypanosoma brucei at Single-Nucleotide Resolution

    The genome of Trypanosoma brucei, the causative agent of African trypanosomiasis, was published five years ago, yet identification of all genes and their transcripts remains to be accomplished. Annotation is challenged by the organization of genes transcribed by RNA polymerase II (Pol II) into long unidirectional gene clusters with no knowledge of how transcription is initiated. Here we report a single-nucleotide resolution genomic map of the T. brucei transcriptome, adding 1,114 new transcripts, including 103 non-coding RNAs, confirming and correcting many of the annotated features and revealing an extensive heterogeneity of 5′ and 3′ ends. Some of the new transcripts encode polypeptides that are either conserved in T. cruzi and Leishmania major or were previously detected in mass spectrometry analyses. High-throughput RNA sequencing (RNA-Seq) was sensitive enough to detect transcripts at putative Pol II transcription initiation sites. Our results, as well as recent data from the literature, indicate that transcription initiation is not solely restricted to regions at the beginning of gene clusters, but may occur at internal sites. We also provide evidence that transcription at all putative initiation sites in T. brucei is bidirectional, a recently recognized fundamental property of eukaryotic promoters. Our results have implications for gene expression patterns in other important human pathogens with similar genome organization (Trypanosoma cruzi, Leishmania sp.) and revealed heterogeneity in pre-mRNA processing that could potentially contribute to the survival and success of the parasite population in the insect vector and the mammalian host

    Überinfusion von Verbrennungsopfern: häufig und schädlich

    Zusammenfassung: Hintergrund: Schwerbrandverletzte (mehr als 20% verbrannter Körperoberfläche bei Erwachsenen) weisen in der ersten Phase (8-48h) einen durch das massive Kapillarleck bedingten Verbrennungsschock auf, der einer Infusionstherapie bedarf, um die Hämodynamik wieder herzustellen. Bis in die 80erJahre stellte eine unzureichende Flüssigkeitstherapie (Unterinfusion) die Haupttodesursache von Verbrennungspatienten dar. Seither ist die übermäßige Flüssigkeitstherapie (Überinfusion) zu einer beachtenswerten Quelle von Komplikationen geworden: abdominales Kompartmentsyndrom, Entlastungsschnitte (Escharotomie), Verschlechterung des Gasaustauschs, Verlängerung der künstlichen Beatmung und des Spitalaufenthalts. Die Überinfusion hat Ende der 90erJahre begonnen, wo innerhalb der ersten 24h Flüssigkeitsmengen zugeführt wurden, die weit über den 4ml/kg/%BSA ("burn surface area") der Parkland-Formel lagen. Ziel: Dieser Beitrag analysiert die Faktoren, welche zu einer Überinfusion führen können und zeigt Möglichkeiten, dem durch eine strikte Kontrolle der präklinischen Infusionstherapie sowie durch eine permissive Hypovolämie vorzubeuge

    Quantum kinetic approach for studying thermal transport in the presence of electron-electron interactions and disorder

    A user friendly scheme based on the quantum kinetic equation is developed for studying thermal transport phenomena in the presence of interactions and disorder. We demonstrate that this scheme is suitable for both a systematic perturbative calculation as well as a general analysis. We believe that we present an adequate alternative to the Kubo formula, which for the thermal transport is rather cumbersome.Comment: 30 pages, 16 figure

    Осуществлен синтез бората алюминия на основе природных сырьевых материалов, изучено влияние добавки CaO на структуру и свойства материала. Было выявлено, что введение CaO в состав исходной сырьевой смеси способствует формированию игольчатых кристаллов и увеличению выхода фазы бората алюминия, а также снижению плотности материала. Показано, что двухстадийный обжиг позволяет повысить прочность образцов с добавкой CaO с сохранением низкой плотности

    Pilot Programs for Veterans Transition To Engineering Fields

    Veterans, through their active service, frequently receive training in highly skilled technical areas. However, they may lack a theoretical background in underlying engineering principles. They also need additional support with the transition from a highly structured military environment to an environment with more ambiguous time constraints and different sorts of responsibilities. Moreover they are facing challenges which are specific for their student population. Therefore, enabling multiple mechanisms which would support them and provide them necessary guidance are especially important at universities with large veteran populations such as at Old Dominion University in Norfolk, Virginia. Hence, there is a need for programs which build on the specialized training that veterans received and aid in their academic journey. This paper will introduce three pilot programs for advancing engineering education for military veterans focusing on forming a support base for veterans to assist them in overcoming traditional educational barriers

    Nonlinear Lattice Waves in Random Potentials

    Localization of waves by disorder is a fundamental physical problem encompassing a diverse spectrum of theoretical, experimental and numerical studies in the context of metal-insulator transition, quantum Hall effect, light propagation in photonic crystals, and dynamics of ultra-cold atoms in optical arrays. Large intensity light can induce nonlinear response, ultracold atomic gases can be tuned into an interacting regime, which leads again to nonlinear wave equations on a mean field level. The interplay between disorder and nonlinearity, their localizing and delocalizing effects is currently an intriguing and challenging issue in the field. We will discuss recent advances in the dynamics of nonlinear lattice waves in random potentials. In the absence of nonlinear terms in the wave equations, Anderson localization is leading to a halt of wave packet spreading. Nonlinearity couples localized eigenstates and, potentially, enables spreading and destruction of Anderson localization due to nonintegrability, chaos and decoherence. The spreading process is characterized by universal subdiffusive laws due to nonlinear diffusion. We review extensive computational studies for one- and two-dimensional systems with tunable nonlinearity power. We also briefly discuss extensions to other cases where the linear wave equation features localization: Aubry-Andre localization with quasiperiodic potentials, Wannier-Stark localization with dc fields, and dynamical localization in momentum space with kicked rotors.Comment: 45 pages, 19 figure

    The complex TIE between macrophages and angiogenesis

    Macrophages are primarily known as phagocytic immune cells, but they also play a role in diverse processes, such as morphogenesis, homeostasis and regeneration. In this review, we discuss the influence of macrophages on angiogenesis, the process of new blood vessel formation from the pre-existing vasculature. Macrophages play crucial roles at each step of the angiogenic cascade, starting from new blood vessel sprouting to the remodelling of the vascular plexus and vessel maturation. Macrophages form promising targets for both pro- and anti-angiogenic treatments. However, to target macrophages, we will first need to understand the mechanisms that control the functional plasticity of macrophages during each of the steps of the angiogenic cascade. Here, we review recent insights in this topic. Special attention will be given to the TIE2-expressing macrophage (TEM), which is a subtype of highly angiogenic macrophages that is able to influence angiogenesis via the angiopoietin-TIE pathway

    Micro-manufacturing : research, technology outcomes and development issues

    Besides continuing effort in developing MEMS-based manufacturing techniques, latest effort in Micro-manufacturing is also in Non-MEMS-based manufacturing. Research and technological development (RTD) in this field is encouraged by the increased demand on micro-components as well as promised development in the scaling down of the traditional macro-manufacturing processes for micro-length-scale manufacturing. This paper highlights some EU funded research activities in micro/nano-manufacturing, and gives examples of the latest development in micro-manufacturing methods/techniques, process chains, hybrid-processes, manufacturing equipment and supporting technologies/device, etc., which is followed by a summary of the achievements of the EU MASMICRO project. Finally, concluding remarks are given, which raise several issues concerning further development in micro-manufacturing
